Novel Process To Produce Protein Concentrate From Horse Gram Seeds

Abstract: The present invention relates to the field of extraction of protein from horse gram seeds. In particular, it relates to the field of extraction of protein firstly by increasing the protein content by reducing the anti-nutritional components like phytates, tannins, and oxalic acid by germination of the horse gram seeds for 48 hours and then drying them at 50 deg C for 12 hours. This process increases the protein content from 17% to 42%. Protein from dried germinated horse gram seed powder is extracted by alkaline solubilization using NaOH and protein precipitation using glacial acetic acid. This process of extraction increases protein content to 70% and also increases lysine content by 60%.

Specification

DESC:The purpose of the present invention is to provide an improved process of protein concentrate from horse gram seeds. In one embodiment of the present invention, horse gram is germinated for 24, 48, and 72 hours. The germinated horse gram is dried at 50 deg C, 60 deg C, 70 deg C for 12 hours. The highest protein content is found to be at 48 hours and drying at 50 deg C gave the highest protein content increasing from 17% to 42%. The dried sprouted horse gram is powdered.
In another embodiment of the present invention horse gram powder is treated with 0.45% of NaOH Solution and kept at 30 deg C for 60 min. The initial pH is checked and is recorded as 11. The solution is filtered and the residue is kept separately. The filtered solution is centrifuged at 2000 RPM, the supernatant is collected separately and stored. The supernatant obtained is treated with glacial acetic acid and pH adjusted to 4. The solution is kept for 12 hours for protein to be precipitated. The precipitate is filtered and dried at 50 deg C for 10 hours until it is completely dried. The protein powdered obtained is tested for protein content and it is concentrated to 70%. The amino acid profiling is done and results indicated that horse gram protein concentrate has a better profile with increase in lysine content by 60% than the horse gram flour. EXAMPLES: In order to implement the present invention and for better understanding, particular embodiments is being explained below through non-limiting examples. It is also understood that the modification of the detail may be done without deviating from the scope of the invention. Example of germination time ranging from 24 hours to 72 hours to reduce the anti-nutritional factors and drying range be from 40?-60? decreases the moisture content, thus stopping the germination.
,CLAIMS:1. A process to increase protein content in horse gram seeds comprising steps of germination and extraction of protein for increased lysine content.
2. The process claimed in claim 1, wherein horse gram seeds are subjected to germination for 48 hours and drying at 50 deg C for 12 hours.
3. The process claimed in claim 1 where, alkali like NaOH, KOH, NH4OH is used for processing
4. The process claimed in claim 1, wherein the process of extraction is carried out at 30 deg C to 70 deg C for 30 min to 90 min
5. The process claimed in claim 1 wherein, formic acid or acetic acid or propionic acid can be used
6. Treatment with alkali as claimed in claim 3 wherein, the alkali concentration varies between 0.1% to 1.0% w/v
