Multiplexed Molecular Sensing By Hybridization Chain Reaction And Long Read Sequencing
Abstract: The present disclosure provides a hybridization chain reaction (HCR) kit and a method for detecting an analyte by employing the HCR kit of the disclosure followed by a long-read sequencing method. The HCR kit comprises an HCR initiator comprising a nucleic acid aptamer and a DNA barcode. The HCR kit also comprises a first hairpin probe and a second hairpin probe containing the same DNA barcode as the HCR initiator. The nucleic acid aptamer of the HCR initiator binds to an analyte in a sample thereby triggering the HCR resulting in an amplified analyte and the DNA barcode detects the presence or absence of the analyte in the sample upon a long-read sequencing.
