Abstract: An organ-on-chip device (100) is disclosed. The device (100) includes a microfluidic platform (1) and a vascular channel (2) positioned on the microfluidic platform (1). One or more tissue chamber (6) is positioned on the microfluidic platform (1). The device (100) includes an interface region (9) connecting the tissue chamber (6) and the vascular channel (2). The interface region (9) includes a plurality of anchor points (10) configured to provide anchorage to cells of the tissue chamber and to stall flow of unpolymerized ECM in the interface region (9). The interface region (9) includes a plurality of connecting points (11) positioned adjacent to the plurality of anchor points (10). The plurality of connecting points (11) are configured to facilitate fluidic interaction between the tissue chamber (6) and the vascular channel (2), and to form a separation interface between the ECM’s of the tissue chamber (6) and the vascular channel (2). 1. An organ-on-chip device (100), comprising:
- a microfluidic platform (1) adapted for establishing a microphysiological system;
- a vascular channel (2) positioned on the microfluidic platform (1), the vascular channel
5 (2) comprising:
(a) at least one inlet (3) and at least one outlet (4); and
(b) one or more perfusion path (5) connecting the at least one inlet (3) and the at
least one outlet (4); and
- one or more tissue chamber (6) positioned on the microfluidic platform (1), said tissue
10 chamber (6) comprising:
a first inlet (7) configured to receive at least one of cells and extracellular matrix
(ECM), to be introduced into the tissue chamber (6);
a second inlet (8) separate from the first inlet (7), the second inlet (8) configured
to store and supply at least one of cell culture media and fluids for the tissue
15 chamber (6); and
- an interface region (9) connecting the tissue chamber (6) and the vascular channel (2),
the interface region (9) comprising:
a plurality of anchor points (10) configured to provide anchorage to cells of
the tissue chamber (6) and to stall flow of unpolymerized ECM; and
20 a plurality of connecting points (11) positioned adjacent to the plurality of
anchor points (10), said plurality of connecting points (11) configured to facilitate
fluidic interaction between the tissue chamber (6) and the vascular channel (2), and
said plurality of connecting points (11) configured as a separation interface between
the ECM of the tissue chamber (6) and an ECM of the vascular channel (2).

2. The organ-on-chip device (100) as claimed in claim 1, wherein at least a portion of the vascular
channel (2) comprises a basement membrane made of the extracellular matrix (ECM).
3. The organ-on-chip device (100) as claimed in claim 1, wherein the tissue chamber (6) is at
30 least one of a tissue microenvironment chamber, a tissue macroenvironment chamber and an
organ microenvironment chamber, and wherein the tissue chamber (6) is positioned on either

side of the vascular channel (2), and wherein each of the tissue chamber (6) being fluidically
connected to the vascular channel (2).
4. The organ-on-chip device (100) as claimed in claim 1, wherein the second inlet (8) isstructured
5 to have a dimensional configuration substantially larger than a dimensional configuration of
the first inlet (7), and wherein said dimensional configuration’s of the second inlet (8) and the
first inlet (7) is configured to be varied based on requirement.
5. The organ-on-chip device (100) as claimed in claim 1, wherein the plurality of connecting
10 points (11), connecting the tissue chamber (6) to the vascular channel (2), have a width of 20
µm and a depth of 40 µm, and wherein each of the connecting point (11) is separated from an
adjacent connecting point (11) by a distance of 210 µm.
6. The organ-on-chip device (100) as claimed in claim 1, wherein the vascular channel (2) is
15 configured to have a depth of 40 µm, width of 500 µm and a length of 11 mm.
7. The organ-on-chip device (100) as claimed in claim 1, wherein the plurality of anchor points
(10) are substantially circular shaped, and wherein the plurality of connecting points (11) are
substantially trapezoidal shaped, and wherein dimensional configuration’s of the plurality of
20 anchor points (10) and the plurality of connecting points (11) is configured to be varied based
on requirement.
8. The organ-on-chip device (100) as claimed in claim 7, wherein the plurality of anchor points
(10) have a diameter of 100 µm, wherein the plurality of anchor points (10) are separated by a
25 distance in range of 150 µm to 170 µm, wherein the plurality of anchor points (10) are arranged
in a zig-zag pattern, and wherein a diameter of the plurality of anchor points (10) are configured
to be varied based on requirement.
9. The organ-on-chip device (100) as claimed in claim 1, wherein the tissue chamber (6) has a
30 diameter of 12 mm, the first inlet (7) has a diameter of 7 mm, and the second inlet (8) has a
diameter of 1 mm.

10. The organ-on-chip device (100) as claimed in claim 1, wherein the first inlet (7) configured to
receive at least one of cells and the ECM, comprises receiving:
(a) organ specific cells embedded with 3D extracellular matrix (ECM), or
(b) ECM and organ specific cells embedded with 3D extracellular matrix (ECM), or
5 (c) ECM and organ specific cells.
