Abstract: A formulation of turmeric complex with full spectrum turmeric and the process for preparation thereof [0030] The present invention relates to a formulation of turmeric (Curcuma longa) complex with full spectrum of turmeric. The formulation is obtained by the process of solvent extraction of ground rhizomes and is found to have high levels of purity. The formulation curcuminoids at a concentration of 90% to 95%, turmeric oil at a concentration of 5% to 10%, bisacurone at a concentration of 1% to 3%, and turmeric protein at a concentration of 1% to 3%. The obtained formulation is completely natural and yields a full spectrum of turmeric extract product which is useful in various other formulations. The full spectrum turmeric extract could be used in therapeutic applications. (Figure 1)

DESCRIPTION OF THE INVENTION

Technical Field of the Invention

[0003] The present invention discloses a formulation of a turmeric complex with full spectrum of turmeric. More specifically, the present invention discloses a formulation consisting of curcuminoids and bisacurone from turmeric oil and turmeric fiber or protein to obtain full spectrum of turmeric extract product. The present invention also discloses a process for preparation of the formulation by using solvent extraction.
Background of the Invention

[0004] Curcuma longa, commonly known as turmeric is an age-old herbaceous plant mainly used as spice, colorant and as a preservative. Turmeric belongs to Zingibereceae family. Rhizome (root) of turmeric is most often cultivated. Turmeric rhizome has a fragrant aroma and bitter, peppery taste. For thousands of years turmeric has been used in traditional herbal medicine both in fresh and dried form. Turmeric oleoresin is obtained when turmeric is subjected to solvent extraction. Turmeric oleoresins are orangish brown viscous fluids that contain non- volatile curcuminoids and volatile oil. The bioactive components of curcuminoids are curcumin, desmethoxycurcumin, bismethoxycurcumin and cyclic curcumin and that of volatile oil include sesquiterpenoids and monoterpenoids.
[0005] Curcuminoids are natural polyphenols that represent the major coloring pigment of turmeric with various therapeutic properties. Curcumin ((1E,6E)-1,7-bis (4-hydroxy- 3-methoxyphenyl) -1,6- heptadiene-3,5-dione) is the major component of curcuminoids. The volatile oil accounts for the aroma of turmeric rhizome. Sesquiterpenoids found in volatile oil exhibits better absorption than curcuminoids and have certain pharmacological properties.
[0006] Numerous biological activities of curcumin have been reported. Curcumin is known to have antioxidant, antimicrobial, anti- inflammatory, neuroprotective, anticancer, hepatoprotective and cardioprotective effects. Studies show volatile oil has therapeutic effects on skin, respiratory tract, and neurological pathways. The sesquiterpenoids of volatile oil have antioxidant, antiviral, anti-inflammatory and anticancer properties.
[0007] Traditionally turmeric is used as an aid for inflammation of various organs, liver and digestive tract problems, various respiratory conditions, and wound healing. Turmeric was also prescribed for pain and swelling. Turmeric is often incorporated with food in Eastern countries which helps in digestion, reduces gas and bloating. Turmeric milk helps to relieve cold and sore throat.
[0008] The patent application No. US9499529B2 entitled “Therapeutic uses of curcumin analogs for treatment of prostate cancer” relates to treating hormone refractory metastatic prostate cancer by curcumin analogs. The cancer cells are treated with various analogs of curcumin to find a suitable analog which has higher solubility, bioavailability and in vivo metabolic stability. The anticancer effect of curcumin influences number of growth factors within cells.
[0009] The patent application No. US7371766B2 entitled “Curcumin analogs with anti-tumor and anti-angiogenic properties” relates to a group of curcumin analogs which inhibit TF expression and VEGF expression which in turn blocks tumor angiogenesis and growth without exhibiting higher toxic levels. The formulation could also be used as a pharmaceutical accepted carrier.
[0010] The patent application No. EP2729138B1 entitled “Anticonvulsant activity of turmeric oil and bisabolene sesquiterpenoids” relates to the effect of sesquiterpenoids present in turmeric such as turmerone and altantone on the central nervous system. Bisabolene, a sesquiterpenoid found in essential oil extracted from rhizomes of turmeric is found to be an anticonvulsant. Sesquiterpenoids act as a therapeutic agent for epilepsy, tremors, pain, mood disorders etc.
[0011] The patent application No. CN101069735A entitled “Turmeric volatile oil extraction and curcumin chemical components purification method” relates to method of extraction and purification of curcumin from turmeric. The process offers a purification process which yields better volatile oil and curcumin content. The process involves use of polar solvent to obtain turmeric extract from solvent extraction. A solid absorber like silica gel is used to adsorb turmeric extract. A non- polar solvent is used to remove volatile oil from curcumin extract. Another polar solvent is used to remove curcumin compound and the solvents from previous steps are recovered. Volatile oil and curcumin are obtained with purity up to 95%.
[0012] The patent application No. CN102617316B entitled “Method of extracting curcumin from turmeric” relates to method incorporated in extraction of curcumin from turmeric. The dried turmeric rhizomes are powdered, mixed with solvents like methanol, ethanol and acetone then subjected to backflow extraction. The heavy metals present in the extract are separated. The precipitates are subjected to fractional distillation and curcumin is obtained.
[0013] Turmeric has its economical importance due to its medicinal and culinary uses. In most of the existing compositions curcumin, a curcuminoid is given utmost importance. The components present in volatile oil such as bisacurone a sesquiterpenoid have shown certain therapeutic effects. It is suitable to formulate a composition with high purity comprising of all the components of turmeric. Hence, such a formulation of turmeric with full spectrum turmeric extract product is of need.
Summary of the Invention
[0014] In order to overcome the drawbacks of the state of art, the present invention discloses a formulation consisting of curcuminoids and bisacurone from turmeric oil and turmeric fiber or protein to obtain full spectrum of turmeric extract product. In addition, the present invention also discloses a process for preparation of the formulation by using solvent extraction.
[0015] The formulation of the present invention is a full spectrum turmeric complex comprising curcuminoids at a concentration of 90% to 95%, turmeric oil at a concentration of 5% to 10%, bisacurone at a concentration of 1% to 3%, and turmeric protein at a concentration of 1% to 3%. The formulation is prepared in a powdered form.
[0016] The invention also discloses a process for preparation of formulation of the turmeric complex with full spectrum of turmeric using solvent extraction method.
[0017] The process of preparation starts with grinding the rhizomes of Curcuma longa and the ground rhizomes are extracted using a solvent namely ethyl acetate or ethanol or isopropyl alcohol to obtain turmeric oleoresin. The obtained oleoresin is subjected to purification, which yields curcuminoids up to 95% of purity. The obtained turmeric oil is said to have bisacurone, which is further purified to obtain enriched fraction. After obtaining turmeric oil and oleoresin, the spent residue is subjected to aqueous extraction. The extraction product of spent residue yields turmeric fiber or protein. The bioactive ingredients obtained from the process include curcuminoids from oleoresin, volatile oil, bisacurone and turmeric fiber or protein. Finally, the bioactive ingredients are powdered and uniformly mixed to get full spectrum of turmeric extract.
[0018] The turmeric formulation disclosed in the present invention comprises full spectrum turmeric product. The full spectrum extract maintains the full profile of the turmeric rhizome. The formulation of full spectrum of the present invention comprises bioactive components with high purity and useful for various therapeutic compositions of turmeric.

Detailed description of the invention

[0021] In order to more clearly and concisely describe and point out the subject matter of the claimed invention, the following definitions are provided for specific terms, which are used in the following written description.
[0022] The term “Curcuminoid” refers to a natural polyphenol with curcumin as a major component responsible for the bright yellow color of turmeric.
[0023] The term “Bisacurone” refers to one of sesquiterpenes which are responsible for the aroma of turmeric.
[0024] Turmeric (Curcuma longa) is a medicinal plant, which plays a pivotal role in managing various ailments like gastrointestinal disorders, inflammation, cancer etc. Turmeric root consists of various significant active components such as volatile oils, curcuminoids, sesquiterpenes etc. Composition of active components varies with species. In addition to curcuminoids, certain terpenoids also contribute to pharmacological effects of turmeric.
[0025] The present invention relates to a unique formulation consisting of curcuminoids, bisacurone from turmeric oil and turmeric fiber or protein to obtain a full spectrum turmeric extract product.
[0026] FIG 1 illustrates the composition of the formulation of the turmeric complex according to an embodiment of the invention. The formulation of the present invention comprises curcuminoids at a concentration of 90% to 95% , turmeric oil at a concentration of 5% to 10%, bisacurone at a concentration of 1% to 3%, and turmeric protein at a concentration of 1% to 3%.
[0027] The present invention also discloses a process for preparation of formulation of the turmeric complex with full spectrum of turmeric. The formulation is prepared by solvent extraction method.
[0028] FIG 2 illustrates the process for preparation of the turmeric complex according to an embodiment of the invention. The process (200) starts with a step (201) of grinding the rhizomes of Curcuma longa. At step (202), the ground rhizomes are extracted using a suitable solvent to obtain turmeric oleoresin. The solvents used in the present invention are selected from ethyl acetate or ethanol or isopropyl alcohol. At step (203), the obtained oleoresin is subjected to purification. The purification yields curcuminoids up to 95% of purity. At step (204), the obtained turmeric oil is said to have bisacurone. Bisacurone is further purified to obtain enriched fraction. At step (205), after obtaining turmeric oil and oleoresin, the obtained spent residue is subjected to aqueous extraction. The extraction product of spent residue yields turmeric fiber or protein. The bioactive ingredients obtained from the process include curcuminoids from oleoresin, volatile oil, bisacurone and turmeric fiber or protein. At step (206), the obtained bioactive ingredients are powdered and uniformly mixed to get full spectrum of turmeric extract.
[0029] The turmeric formulation disclosed in the present invention comprises full spectrum turmeric product. The full spectrum extract maintains the full profile of the turmeric rhizome. The bioactive components extracted have a high level of purity and have desirable composition. The full spectrum turmeric extract could be further used in various formulations. The formulation of full spectrum of the present invention comprises bioactive components with high purity and useful for various therapeutic compositions of turmeric.
,CLAIMS:
We Claim,
1. A formulation of turmeric complex for enhanced purity, the formulation comprises:
a. curcuminoids at a concentration in a range between 90%-95%;
b. turmeric oil at a concentration in a range between 5%-10%;
c. bisacurone at a concentration in the range of 1%-3%; and
d. turmeric protein at a concentration in the range of 1%-3%.
wherein the ingredients are uniformly mixed to form a full spectrum complex .
2. The formulation of liposphere as claimed in claim 1, wherein the ingredients are isolated from rhizomes of Curcuma longa.

3. The formulation of liposphere as claimed in claim 1, wherein the turmeric complex exhibits enhanced purity of one or more ingredients.

4. The formulation of liposphere as claimed in claim 1, wherein the formulation of turmeric complex is prepared in the powdered form.

5. A process for preparation of the formulation, the method (200) comprising the steps of:
a. grinding the rhizome of Curcuma longa (201);
b. extracting the ground rhizomes using a solvent to obtain turmeric oleoresin (202);
c. subjecting the obtained oleoresin to purification (203);
d. subjecting the turmeric oil of Step b containing bisacurone to obtain enriched fraction (204);
e. subjecting the spent residue after obtaining turmeric oil and oleoresin to aqueous extraction to yield turmeric fiber or protein (205); and
f. uniformly mixing the obtained one or more bioactive compounds (206)

wherein the process results in the formulation in the powdered form.
6. The method as claimed in Claim 5, the solvent used in extraction are selected from ethyl acetate or ethanol or isopropyl alcohol.

7. The method as claimed in Claim 5, the purification of oleoresin yields curcuminoids up to 95% of purity.

8. The method as claimed in Claim 5, the bioactive compounds include curcuminoids from oleoresin, volatile oil, bisacurone and turmeric fiber or protein.

9. The method as claimed in Claim 5, the process results in the formation of full spectrum turmeric complex to maintain complete profile of the turmeric rhizome.

10. The method as claimed in Claim 5, the formulation of said process is prepared in the powder form.
