Abstract: The present disclosure relates to a composition comprising pomegranate fruit extract and jamun seed extract which at specific ratios and concentrations can be used for the management of metabolic diseases. Also provided are methods of preparation and uses of the said composition.

FIELD OF INVENTION
[001] The present disclosure relates to the field of herbal compositions in general and compositions to treat metabolic diseases in particular. There is provided a composition comprising jamun seed extract and pomegranate fruit extract.
5 BACKGROUND OF INVENTION
[002] Diabetes and obesity belong to the large class of metabolic diseases, caused primarily by the increasingly sedentary lifestyles lead by people and the intake of excess calorific food. [003] Obesity is among the most common lifestyle diseases witnessed today, which
10 is marked by excess storage of fat in the body. Sedentary lifestyles, excessive food
intake in combination with the genetic disposition of an individual are the primary factors contributing to this disease. The development of obesity often leads to several other metabolic diseases including heart disease, type 2 diabetes, obstructive sleep apnea, certain types of cancer, and osteoarthritis [Haslam and James, "Obesity",2005
15 Lancet 366 (9492): 1197–209]. India ranks third in top ten countries with highest
number of obese people. [Marie et al., The Lancet, 2014, 6736(14)]. [004] Diabetes is very often an outcome of obesity-related ailments. It is predicted to be one of the most prevalent disease in Asia as an increasingly urbanized population feeds primarily on high-sugar and nutrient poor diets. India ranks second
20 for people with diabetes in top ten countries (IDF diabetes Atlas 5th edition, 2012
update). The disease is characterized by high blood glucose levels owing to reduced insulin levels.
[005] Metabolic diseases are multifactorial, with both intrinsic and extrinsic factors playing a major role in the development of these diseases. This makes it difficult to
25 find a panacea to encounter the varied symptoms of these diseases. An alternative is
to use a targeted approach. By regulating a few key metabolic players contributing to

the development of these diseases, both obesity and diabetes can be gradually controlled.
[006] Pancreatic alpha-amylases are the most important digestive enzymes in
humans and act as a catalysis in the reaction which involves the hydrolysis of the
5 alpha-1,4 glycosidic linkages of the starch. The inhibitors of the alpha-amylase may
prevent the digestion of carbohydrates and may be used in the treatment of type 2 diabetes and the like [Prashant and Ritika, RRJMHS, 2016: 5(4)]. [007] US3174901A relates to a method of treating diabetes by oral administration as well as to the pharmaceutical compositions useful therefor. The composition to be
10 administered orally contains an active ingredient- dimethyl biguanide.
[008] US6551627B1 is directed to a herbal medicinal composition for preventing or treating type II diabetes. The composition is comprised of extracts from Pterocarpus marsupium, Morus alba, Orthosiphon aristatus, Opiophogon japonicus, Rosa rugosa, Commelina communis, Trichosanthis kirilowii and Anemarrhena asphodeloides.
15 [009] US6559188B1 relates to a combination, such as a combined preparation or
pharmaceutical composition, respectively, which comprises nateglinide or repaglinide and at least one other antidiabetic compound, to delay of progression or treatment of diseases, especially metabolic disorders.
20 SUMMARY OF INVENTION
[0010] In an aspect of the present disclosure there is provided, a composition
comprising: (a) jamun seed extract; and (b) pomegranate fruit extract, wherein jamun
seed extract to pomegranate fruit extract w/w ratio in said composition is in the range
of 1:1- 1:2.
25 [0011] In an aspect of the present disclosure there is provided, a method of preparing
a composition comprising: (i) jamun seed extract; and (ii) pomegranate fruit extract,

said method comprising: (a) obtaining the jamun seed extract; (b) obtaining the
pomegranate fruit extract; and (c) contacting the jamun seed extract, and the
pomegranate fruit extract to obtain said composition, wherein the jamun seed extract
to the pomegranate fruit extract w/w ratio in said composition is in the range of 1:1-
5 1:2.

will be better understood with reference to the following description and appended

BRIEF DESCRIPTION OF ACCOMPANYING DRAWINGS

[0014] Figure 1 shows the effect of jamun seed extract and pomegranate fruit extract at different concentrations on the inhibition of alpha-amylase activity, in accordance with an embodiment of the present disclosure.

pomegranate fruit extract in different combination ratios, on the inhibition of alpha-amylase activity, in accordance with an embodiment of the present disclosure.

DETAILED DESCRIPTION OF THE INVENTION

understood that the present disclosure includes all such variations and modifications.

Definitions
[0017] For convenience, before further description of the present disclosure, certain
10 terms employed in the specification, and examples are delineated here. These
definitions should be read in the light of the remainder of the disclosure and understood as by a person of skill in the art. The terms used herein have the meanings recognized and known to those of skill in the art, however, for convenience and completeness, particular terms and their meanings are set forth below.

10 [0024] Metabolic disorders include diseases such as obesity and diabetes and are a
major problem arising out of the increasingly sedentary lifestyles and poor quality of food consumed by primarily urbanized populations. Several medicaments and compositions are available to manage these diseases, however, they still remain a major problem which needs to be addressed. Among the solutions available is to use
15 herbal compositions, which have limited side-effects, to treat these diseases in a
targeted manner. The enzyme alpha-amylase is one such target, that regulates the conversion of starch to sugars within the body, thus contributing to the increase of blood sugar levels. [0025] Natural organic compounds have been reported to inhibit the activity of
20 pancreatic alpha-amylase enzyme. Punica granatum is also known as pomegranate
and it belongs to the family Punicaceae. The fruit of the Punica granatum has extensively been used as a traditional remedy against acidosis, dysentery, microbial infections, diarrhea, helminth infection, hemorrhage and respiratory pathologies. Punica granatum is rich in polyphenols (Reza et al., Iran J Pharm Res. 2012 Spring;
25 11(2): 385–400).
[0026] Eugenia jambolana Lam. is also known as black plum or Jamun belongs to family Myrtaceae and native to India. Most of the plant parts such as fruit, pulp, seed, leaves and bark of Eugenia jambolana are used in traditional system of medicine. Literatures suggest that the various extracts of Eugenia jambolana Lam hold a range

of biological effect such as antibacterial, antifungal, antiviral, anti-genotoxic, anti-
inflammatory, anti-ulcerogenic, cardioprotective, anti-allergic, anticancer,
chemopreventive, radioprotective, free radical scavenging, antioxidant,
hepatoprotective, anti-diarrheal, hypoglycemic and antidiabetic effects (Manjeshwar
5 et al., Food Research International, 2011, 44(7): 1776–1789).
[0027] The present disclosure reports a surprising find, in that, the combination of jamun seed extract and pomegranate fruit extract at particular ratios inhibits alpha-amylase activity. Thus, the present disclosure provides a solution to the problem of metabolic diseases in the form of a composition which can manage these diseases
10 through the inhibition of alpha-amylase enzyme activity.
[0028] The present disclosure is not to be limited in scope by the specific embodiments described herein, which are intended for the purposes of exemplification only. Functionally-equivalent products, compositions, and methods are clearly within the scope of the disclosure, as described herein.
15 [0029] In an embodiment of the present disclosure there is provided, a composition
comprising: (a) jamun seed extract; and (b) pomegranate fruit extract, wherein jamun seed extract to pomegranate fruit extract w/w ratio in said composition is in the range of 1:1- 1:2. [0030] In an embodiment of the present disclosure there is provided, a composition

obtaining the pomegranate fruit extract; and (c) contacting the jamun seed extract, and pomegranate fruit extract to obtain said composition, wherein jamun seed extract to pomegranate fruit extract w/w ratio in said composition is in the range of 1:1- 1:2.

defined otherwise, all technical and scientific terms used herein have the same
meaning as commonly understood to one of ordinary skill in the art to which this
disclosure belongs. Although methods and materials similar or equivalent to those
15 described herein can be used in the practice of the disclosed methods and
compositions, the exemplary methods, devices and materials are described herein. It is to be understood that this disclosure is not limited to particular methods, and experimental conditions described, as such methods and conditions may vary.
Example 1
20 Material and Methods:
[00116] Preparation of pomegranate fruit extract: Stock solution of
pomegranate fruit extract was prepared using commercially available pomegranate fruit extract from Vidya Herbs, Bangalore. A stock solution pomegranate fruit extract powder was prepared by dissolving 100 mg/ml of solvent, preferably dimethyl
25 sulfoxide (DMSO - 100%). A 200 - 1000 μg/300 μL of pomegranate fruit extract was
prepared by dissolving 2.0 μL - 10 μL of stock solution in phosphate buffer.

Pomegranate fruit extract was standardized to 10% of polyphenols by TLC. The
polyphenols are present in the range of 10% to 80% in this extract.
[00117] Preparation of Eugenia jambolana Lam extracts: Stock solution of
Eugenia jambolana Lam extract was prepared by using commercially available
5 Eugenia jambolana Lam extract from Indena, Italia. A stock solution Eugenia
jambolana Lam extract powder was prepared by dissolving 100 mg/ml of solvent, preferably water: ethanol: dimethyl sulfoxide: (2:1:4 %). A 200 - 1000 μg/300 μL of Eugenia jambolana Lam extract was prepared by dissolving 2.0 μL - 10 μL of stock solution in phosphate buffer. Eugenia jambolana Lam extract was standardized to
10 1.5% of ellagic acid by HPLC. The Ellagic acids are present in the range of 0.5% to
2% in this extract.
[00118] Alpha amylase assay: Alpha-amylase activity can be measured in-vitro
by hydrolysis of starch in presence of alpha-amylase enzyme. This process was quantified by using iodine, which gives blue colour with starch. The reduced intensity
15 of blue colour indicates the enzyme-induced hydrolysis of starch into
monosaccharides. If the substance/extract possesses alpha-amylase inhibitory activity,
the intensity of blue colour will be more. In other words, the intensity of blue colour
in test sample is directly proportional to alpha- amylase inhibitory activity.
[00119] Alpha-amylase activity was carried out by starch-iodine method. 10
20 μL of pancreatic α- amylase solution was mixed with 250 μL of phosphate buffer
(0.02 M with 0.006 M NaCl, pH 7.0) containing 10 μl of different concentration of extracts. After incubation at 37 °C for 10 min, 20 μL of starch solution (1%) was added, and the mixture was re-incubated for 1 h at 37 °C. 10 μl of 1% iodine solution was added, and diluted with distilled water, the absorbance was taken at 565 nm.
25 Example 2
Inhibition of alpha-amylase activity by jamun seed extract and pomegranate fruit extract
[00120] In order to determine whether jamun seed extract and pomegranate
fruit extract, had any effect in inhibiting the activity of alpha-amylase, each of the

extracts was used using the in vitro assay as described above and tested for inhibitory
effect. Both the extracts were tested at 0.67 mg/mL, 1.67 mg/mL and 3.3 mg/mL.
[00121] As seen in Figure 1, the results indicate that both jamun seed extract
and pomegranate fruit extract possess the ability to inhibit the activity of the enzyme,
5 and the percent inhibition increases with increasing concentrations of the actives. In
the case of jamun extract, the maximum percent inhibition was observed at a
concentration at 3.3 mg/mL of approximately 65-70% inhibition, while at 1.67
mg/mL, the percent inhibition observed was at approximately 20-25%. In general,
pomegranate fruit extract was slightly less effective than jamun seed extract in its
10 inhibitory effect, the maximum percentage inhibition was observed at the
concentration of 3.3 mg/mL at 40-50% inhibition. While at concentrations lower than 3.3 mg/mL, pomegranate fruit extract did not show any inhibitory effect.
Example 3
15 Synergistic inhibition of alpha-amylase activity by the combination of jamun
seed extract and pomegranate fruit extract
[00122] As both the extracts under study demonstrated varying inhibitory
effects on the activity of the enzyme alpha-amylase, the combination of both jamun seed extract and pomegranate fruit extract was attempted. The concentrations of the
20 combinations tested are listed below in Table 1.

As seen from Figure 2, all combinations of the extracts could inhibit the activity of
alpha-amylase. However, among the combinations tested, a 1:1 and 1:1.54 ratios
showed a remarkably and highly significant (p= <0.05) percent inhibition of enzyme
activity.
5 [00123] At a 1:1 ratio of jamun seed extract to pomegranate fruit extract, the
percent inhibition observed was at 178.70%. This is over four to five times higher than the percent inhibition expected by merely mixing the two extracts as is extrapolated from Figure 1. In fact, the percent inhibition observed using a 1:1 ratio of the actives, is even two-fold higher from expected values if a combination of the
10 extracts at their individual maximum concentration was used.
[00124] Figure 2 further reveals that a 1:1.54 ratio of jamun seed extract and
pomegranate fruit extract also shows an enhanced effect in the inhibition of alpha-amylase activity. At 93.61% inhibition, the combination of the extracts at this ratio is at least 1.5 to two fold higher than the expected inhibition values if both the extracts
15 were mixed together as is evident form the concentration curve obtained from Figure
1.
Advantages of present disclosure:
Overall the present disclosure provides a composition comprising jamun seed extract and pomegranate fruit extract which shows significant synergism in inhibiting the

activity of the enzyme alpha-amylase, than would be expected by a mere admixture
of the two ingredients. This synergism is particularly evident at a 1:1 and a 1:1.54
ratio of jamun seed extract to pomegranate fruit extract, respectively. While the
1:1.54 ratio demonstrates 1.5 to two fold higher percentage inhibition than expected
5 values, a 1:1 ratio demonstrates four to five fold higher percentage inhibition than
expected values. However, from a range of combinations attempted, only these two
ratios demonstrate any synergistic effect. Thus, the composition of jamun seed extract
to pomegranate fruit extract cannot be obtained without undue experimentation and is
therefore not is not obvious to a person skilled in the art. This composition can thus
10 be utilised further to treat metabolic disorders including diabetes, obesity and the
complications thereof.
