Abstract: The present disclosure provides a high-throughput method of hydroxyapatite stain quantification that is scalable, economical, quick, and more accurate than conventional methods of hydroxyapatite staining methods, including photometric methods.
CLIAMS:1. A high-throughput method of quantification of hydroxyapatite staining, said method comprising of:
a. drop casting hydroxyapatite film in a multi-well plate;
b. adding and incubating a pre-determined amount of at least one stain to said drop casted hydroxyapatite film in a multi-well plate;
c. removing excess amount of said stain from drop casted hydroxyapatite film; and
d. using a device to measure the coloration of said hydroxyapatite film.
2. A high-throughput method of identification of an ingredient or combinations thereof that inhibit teeth staining, said method comprising of:
a. drop casting hydroxyapatite film in a multi-well plate;
b. adding and incubating a pre-determined amount of at least one stain to said drop casted hydroxyapatite film in a multi-well plate;
c. removing excess amount of said stain from drop casted hydroxyapatite film;
d. adding and incubating a pre-determined amount of an ingredient or combinations thereof that can remove or inhibit teeth stain caused by the said stain;
e. removing said ingredient from drop casted hydroxyapatite film; and
f. using a device to measure the coloration of said hydroxyapatite film.
3. The high-throughput method as claimed in any of the claims 1-2, wherein coloration measurement is expressed in CIE L*a*b* (CIELAB) color space.
4. The high-throughput method as claimed in claim 3, wherein a combination of L*, a*, and b* values are used to express coloration measurement.
5. The high-throughput method as claimed in any of the claims 3-4, coloration of hydroxyapatite film is calculated as total color difference, ?E*, wherein ?E* = [(?L*)2+(?a*)2+(?b*)2 ]1/2.
6. The high-throughput method as claimed in any of the claims 1-2, wherein said at least one stain comprises of at least one chromophore.
7. The high-throughput method as claimed in any of the claims 1-2, wherein said device is a chromameter.
8. The high-throughput method as claimed in any of the claims 1-2, wherein said at least one statin comprises of at least one chromophore
