Abstract: A process for preparing pheniramine maleate, comprising addition of a predetermined amount of an isopropyl alcohol and a maleic acid to obtain a mixture, heating the isopropyl alcohol and the maleic acid mixture at a predetermined temperature, adding the isopropyl alcohol and the maleic acid mixture to a N, N-dimethyl-3-phenyl-3-(pyridin-2-yl) propan-1-amine prepared by a predetermined process to obtain a reaction mass, heating the reaction mass at a temperature in the range of 60oC to 65oC for a predetermined period of time to isolate the product, cooling the isolated product at a temperature in the range of 10oC to 15oC and filtering and drying the isolated product to obtain a pure pheniramine maleate. Claims:1. A method for preparation of pheniramine maleate, the method comprising:
addition of a predetermined amount of an isopropyl alcohol and a maleic acid to obtain a mixture;
heating the isopropyl alcohol and the maleic acid mixture at a predetermined temperature;
adding the isopropyl alcohol and the maleic acid mixture to a N, N-dimethyl-3-phenyl-3-(pyridin-2-yl) propan-1-amine prepared by a predetermined process to obtain a reaction mass;
heating the reaction mass at a temperature in the range of 60oC to 65oC for a predetermined period of time;
cooling the content at a temperature in the range of 10oC to 15oC to isolate the product; and
filtering and drying the isolated product to obtain a pure pheniramine maleate.
2. The process as claimed in claim 1, wherein predetermined process of preparing the N, N-dimethyl-3-phenyl-3-(pyridin-2-yl) propan-1-amine comprises of:
condensing a predetermined amount of benzyl cyanide and a 2-Chloro pyridine using a condensing agent to form a phenyl(pyridin-2-yl)acetonitrile;
obtained phenyl(pyridin-2-yl)acetonitrile is condensed insitu with a dimethyl aminoethyl chloride, wherein the reaction mixture is maintained at a same temperature for 2 hours, followed by maintaining the reaction mixture at 65oC to 70oC for 2 hours and at 110oC to 115oC for a time period of 2hours;
cooling the reaction mixture to room temperature and addition of predetermined amount of methanol and distilled water to extract a 2-(phenyl)-4-(dimethylamino)-2-(pyridin-2-yl) butanenitrile in O-xylene by a phase separation technique;
decyanation of the 2-(phenyl)-4-(dimethylamino)-2-(pyridin-2-yl) butanenitrile is carried out with a potassium hydroxide in O-xylene at a high temperature,
addition of distilled water to extract the decyanated product into an organic layer by the phase separation technique;
concentrating the O-Xylene layer to obtain brown coloured crude oil of N, N-dimethyl-3-phenyl-3-(pyridin-2-yl) propan-1-amine; and
collecting N, N-dimethyl-3-phenyl-3-(pyridin-2-yl) propan-1-amine by a high vacuum fractional distillation.
3. The process as claimed in claim 2, wherein the condensing agent is sodium amide.
4. The process as claimed in claim 1, wherein the pH of the reaction mass is in the range of 5.0 to 6.0.
5. The process as claimed in claim 1, wherein the predetermined time comprises of 60 minutes.
6. The process as claimed in claim 2, wherein the high vacuum fractional distillation is carried out at NLT 650 mmHg at a temperature in the range of 155oC to 195oC.
7. The process as claimed in claim 2, wherein the reaction is monitored by a gas chromatography.
8. The process as claimed in claim 2, wherein the high temperature is in the range of 140oC to 155oC at the time of decyanation.

, Description:FIELD OF INVENTION
[0001] Embodiment of the present disclosure relates to a method for preparation of Pheniramine maleate and more particularly to a novel less hazardous method of preparing the same.
BACKGROUND OF THE INVENTION
[0002] Histamine is an amine that is produced as part of a local immune response to cause inflammation. More particularly, as part of the immune response to foreign pathogens, histamine is produced by basophils and by mast cells. It is also known that it is histamine which is responsible for causing watery eyes, runny or blocked nose, sneezing, skin rashes, itching, etc during an allergic reaction.
[0003] Pheniramine maleate is used as an antiallergic medication, when a body is exposed to any allergen such as pollen, animal dander, house dust, etc. The mechanism of action of pheniramine maleate involves blocking the action of histamine to relieve these symptoms.
[0004] US2567245 discloses the hydrolysis & decarboxylation of 2-(phenyl)-4-(dimethylamino)-2-(pyridin-2-yl) butanenitrile was performed by cooking with Sulfuric acid at 140oC to 150oC for 24 hours to obtain N, N-dimethyl-3-phenyl-3-pyridin-2-yl propan-1-amine.
[0005] The presently available processes are not safe and time consuming. Therefore, there is a need for an alternative safe, short duration and less hazardous process for obtaining pheniramine maleate.
SUMMARY OF THE INVENTION
[0006] In accordance with an embodiment of the present invention, a method for preparation of pheniramine maleate is disclosed, wherein the method comprises of addition of a predetermined amount of an isopropyl alcohol and a maleic acid to obtain a mixture, heating the isopropyl alcohol and the maleic acid mixture at a predetermined temperature, adding the isopropyl alcohol and the maleic acid mixture to a N, N-dimethyl-3-phenyl-3-(pyridin-2-yl) propan-1-amine prepared by a predetermined process to obtain a reaction mass, heating the reaction mass at a temperature in the range of 60oC to 65oC for a predetermined period of time, cooling the content reaction at a temperature in the range of 10oC to 15oC to isolate the product and filtering followed by drying the isolated product to obtain a pure pheniramine maleate.
[0007] In accordance with an embodiment of the present invention, wherein the predetermined process of preparing the N, N-dimethyl-3-phenyl-3-(pyridin-2-yl) propan-1-amine comprises of condensing a predetermined amount of benzyl cyanide and a 2-Chloro pyridine using a condensing agent to form a phenyl(pyridin-2-yl)acetonitrile, obtained phenyl(pyridin-2-yl)acetonitrile is condensed insitu with a dimethyl aminoethyl chloride, wherein the reaction mixture is maintained at a same temperature for 2 hours, followed by maintaining the reaction mixture at 65oC to 70oC for 2 hours and at 110oC to 115oC for a time period of 2hours, cooling the reaction mixture to room temperature and addition of predetermined amount of methanol and distilled water to extract a 2-(phenyl)-4-(dimethylamino)-2-(pyridin-2-yl) butanenitrile in O-xylene by a phase separation technique, decyanation of the 2-(phenyl)-4-(dimethylamino)-2-(pyridin-2-yl) butanenitrile is carried out with a potassium hydroxide in O-xylene at a high temperature, addition of distilled water to extract the decyanated product into an organic layer by the phase separation technique, concentrating the O-Xylene layer to obtain brown coloured crude oil of N, N-dimethyl-3-phenyl-3-(pyridin-2-yl) propan-1-amine and collecting N, N-dimethyl-3-phenyl-3-(pyridin-2-yl) propan-1-amine by a high vacuum fractional distillation.

Pheniramine maleate binds to histamine H1 receptors, thereby inhibiting phospholipase A2 and production of endothelium-derived relaxing factor, nitric oxide. Subsequent lack of activation of guanylyl cyclase through nitric oxide results in decreased cyclic GMP (cGMP) levels, thereby inhibiting constriction of smooth muscle tissue, and decreased capillary permeability and histamine-activated allergic reactions. It is chemically designated as N, N-dimethyl-3-phenyl-3-pyridin-2-ylpropan-1-amine.

EXAMPLE 1
Preparation of N, N-dimethyl-3-phenyl-3-(pyridin-2-yl) propan-1-amine-
[0046] In a Reactor with an Inert atmosphere, 3650 Liters of O-Xylene, 600 kg of Benzyl Cyanide and 570 kg of 2-Chloro Pyridine was added sequentially. To this mixture, a slurry mixture of 400 kg Sodium Amide in 1000 liter of O-Xylene is carefully added up to 55oC to 60oC. After maintaining the reaction for an hour, the solution of Dimethyl amino ethyl chloride in O-Xylene, which can be prepared by chlorinating 2-(Dimethyl amino) ethanol using Thionyl chloride, as its Hydrochloride salt and basifying the salt by the common technique followed by extracting in O-Xylene, is carefully added. The reaction is maintained at same temperature for 2 hours, then at 65oC to 70oC for 2 hours and further maintained for 2 hours at 110oC to 115oC. The reaction is monitored by gas chromatography during the maintaining period at 110oC to 115oC. The reaction mixture was cooled to room temperature and methanol 50 liter was added followed by 2000 liter water addition. The 2-(phenyl)-4-(dimethylamino)-2-(pyridin-2-yl) butanenitrile is extracted in O-Xylene by phase separation technique. Further reaction is heated to 140oC to 155oC with caustic Potash. The reaction conversion is monitored by gas chromatography. The reaction is brought to 70oC temperature and 2000 liter distilled water was added followed by phase separation to extract the decyanated product into an organic layer. The brown colored crude oil of N, N-dimethyl-3-phenyl-3-(pyridin-2-yl) propan-1-amine is obtained by concentrating the O-Xylene layer. The pure fraction was collected by high vacuum fractional distillation at the vacuum NLT (not less than) 650 mmHg at 155oC to 195oC.
EXAMPLE 2
Preparation of Pheniramine Maleate
[0047] In a reactor, sequentially added 600 liter of Isopropyl alcohol & 159 kg of Maleic Acid, on dry basis. This is heated to 50oC to 55oC to dissolve. In another reactor sequentially added 200 Liter Isopropyl alcohol & 350 kg N, N-dimethyl-3-phenyl-3-(pyridin-2-yl) propan-1-amine, prepared by the method as described in Example-1. The maleic acid solution is transferred in to above solution to adjust pH to 5.0 to 6.0. The reaction was heated to 60oC to 65oC for 60 minutes. The maleate salt formed is isolated by chilling to 10oC to 15oC, followed by filtration & drying. The Highly pure Pheniramine Maleate is obtained.
[0048] The pheniramine maleate obtained using the process of present invention is cheaper, efficient and furthermore industrially safer to produce.
[0049] More specifically, in the present disclosure, the decyanation of 2-(phenyl)-4-(dimethylamino)-2-(pyridin-2-yl) butanenitrile is performed by using Potassium hydroxide which is quite safe to handle rather than Sulfuric acid, considering an industrial safety. An additional advantage is that the reaction can be terminate for further work up in 10 to 12 hours of maintaining, which reduces the time and utility consumption as well.
[0050] Overall, the present disclosure discloses a one pot synthesis for decyanation of 2-(phenyl)-4-(dimethylamino)-2-phenyl-2-(pyridin-2-yl) butanenitrile, which can be synthesized by condensing Benzyl cyanide with 2-Chloro pyridine by using sodium amide as a condensing agent in O-Xylene to form phenyl(pyridin-2-yl)acetonitrile which further condensed insitu with dimethyl aminoethyl chloride. In the present disclosure, the cyanide group is removed by treating the Nitrile with Potassium Hydroxide in Xylene at high temperature.
