A Portable Clean Room For Separation And Manipulation Of Stem Cells For Clinical Application

Abstract: Central Chamber (A) enclosed in a thick plexiglass is flanked by two chambers, Al (right) and A2 (left). Chamber A is topped by a Hepa Filter of size 12" X 12" X 6" (IB) and Air Blower (2B) of capacity l/5th HP. Chambers Al and A2 are also topped by Filter-Blower Assembly. Chamber Al and A are connected by a sliding plexiglass door, similar is the link between Chambers A and A2. Manipulation is done in the Central Chamber A, through two Ports (PI and P2) guarded by gaskets, to which a pair of rubber latex gloves is fixed. Sliding plexiglass door separating the Chambers is either manual or motor driven. Pre-filter (6B) supplies dust free air into the clean room. Unit is mounted Stainless Steel Stand (9B). Four number castor wheels (10B), on which the SS Stand carrying the unit is supported and four number leveling jacks are provided.

This Invention, A PORTABLE CLEAN ROOM FOR SEPARATION AND MANIPULATION OF STEM CELLS FOR This Invention, A PORTABLE CLEAN ROOM FOR SEPARATION AND MANIPULATION OF STEM CELLS FOR CLINICAL APPLICATION is aimed at providing a portable HEPA filtered positive pressure environment, which provides for a sterile work place. Such a clean atmosphere is mandatory for the separation of stem cells from the fat tissue. The conventional methods consist of clean rooms, which are very expensive to construct and it is much more expensive.

In this Invention A PORTABLE CLEAN ROOM FOR SEPARATION AND MANIPULATION OF STEM CELLS FOR CLINICAL APPLICATION, a Clean room with specified HEPA filtered air under pressure, with air-locks with lower air-pressure of clean air for entry and exit of mass and material is needed for sterile processing of stem cells from cord blood/bone marrow or fat derived mesenchymal stem cells. The stem cells thus processed will be used for therapeutic purpose. The present Invention A PORTABLE CLEAN ROOM FOR SEPARATION AND MANIPULATION OF STEM CELLS FOR CLINICAL APPLICATION provides a satisfactory solution to all the above stated problems, by providing a bench top system, which could be handled with ease, as compared to the complicated and elaborate conventional system. Using this system, stem cell processing can be carried out be trained personnel at the same place, where bone marrow aspiration/Lipoaspirate or cord blood collection is done. Moreover, this A PORTABLE CLEAN ROOM FOR SEPARATION AND MANIPULATION OF STEM CELLS FOR CLINICAL APPLICATION is of light weight and requires minimal space. Further, as far as the cost of the equipment/system is concerned, it may come up to hardly 10% of the conventional method that is presently being used.

A PORTABLE CLEAN ROOM FOR SEPARATION AND MANIPULATION OF STEM CELLS FOR CLINICAL APPLICATION consists of a Central Chamber (A), which is enclosed in a thick plexiglass. The Central Chamber (A) is flanked by two chambers, Al to the right of the Central Chamber and A2, to its left. The top of the Chamber A accommodates a Hepa Filter of size 12" X 12" X 6" (1 of Figure B) and Air Blower (2 of Figure B) of capacity 1/5* HP. As in the case of A, chambers Al and A2 are also topped by Filter-Blower Assembly. Chamber Al is connected to the Chamber A by means of a sliding plexiglass door and similar is the link between Chamber A and Chamber A2. Manipulation is done in the Central Chamber A, through two Ports (P1 and P2), to which a pair of rubber latex gloves of suitable size is fixed. Moreover, the Ports P1 & P2 are guarded by suitable gaskets, to make it leak proof. The sliding plexiglass door separating the Chambers could be either manually operated or optionally could be motor driven. 6 of Figure (B) on Drawing Sheet No: 02 is a Pre-filter, which ensures supply of dust free air into the clean room. 9 of Figure (B) on Drawing Sheet No: 02 is a Stainless Steel Stand, on which the entire PORTABLE CLEAN ROOM FOR SEPARATION AND MANIPULATION OF STEM CELLS FOR CLINICAL APPLICATION is mounted. 10 of Figure (B) on Drawing Sheet No: 02 are four number castor wheels, on which the SS Stand carrying the entire unit is supported and four number leveling jacks have also been provided for the adjustment and proper balancing of the entire unit. 12 of Figure (B) on Drawing Sheet No: 02 represents the Exhaust duct.

In this newly invented A PORTABLE CLEAN ROOM FOR SEPARATION AND MANIPULATION OF STEM CELLS FOR CLINICAL APPLICATION, it is mandatory that the air pressure in the chamber A is always higher than the other two compartments, ie., Al & A2, the difference in the other two chambers being lower by 15 pascals. This is to make sure that as and when the prepared product is removed from the Chamber A, it does not compromise on the environment of A. Accordingly Chamber Al takes care of the environment in Chamber A, when the raw material to be processed is introduced into the Chamber A. Thus, chambers Al & A2 ensure that the required air pressure is maintained in the Chamber A, not only during the cleaning process, but also at the time of introducing the raw material into Chamber A for processing and also at the time of removal of the end product from the Chamber A. Moreover, the filter blower assembly fitted atop chamber A is capable of creating a class 100 atmosphere under pressure. The respective air pressures in the Chambers A, A1 and A2 is shown by the gauges fitted on the front face of the respective chambers and are shown as G, G1 and G2 in the Figure (A) of Drawing Sheet No: 01.

By making use of this A PORTABLE CLEAN ROOM FOR SEPARATION AND MANIPULATION OF STEM CELLS FOR CLINICAL APPLICATION, the sterile stem cell samples so obtained are free of toxicity and endotoxin. Moreover, this system drastically reduces the cost of preparing a stem cell sample, which can be used clinically. The biggest advantage of this A PORTABLE CLEAN ROOM FOR SEPARATION AND MANIPULATION OF STEM CELLS FOR CLINICAL APPLICATION is that it is possible to mover the sterile work station, to suit our convenience and requirement, thereby making it synonymous with the title, a portable clean room. Another added advantage of this system is that the time required for preparing stem cell sample is reduced drastically, as the procedure can now be done in the operation theatre of the relevant hospital itself. Further, such portable clean rooms can not only be used for tissue culture applications, but can be used for hard drive data recovery, as well.


We Claim,

1) A PORTABLE CLEAN ROOM FOR SEPARATION AND MANIPULATION OF STEM CELLS FOR CLINICAL APPLICATION consisting of a Central Chamber (A), which is enclosed in a thick plexiglass; the said Central Chamber (A) being flanked by two chambers, Al to the right of the Central Chamber and A2, to its left, wherein, the top of the Chamber A accommodates a Hepa Filter of size 12" X 12" X 6" (1B) and Air Blower (2B) of capacity 1/5th HP;

As in the case of A, Chambers Al and A2 are also topped by Filter-Blower Assembly and Chamber Al is connected to the Chamber A by means of a sliding plexiglass door and similar is the link between Chamber A and Chamber A2;

Manipulation is done in the Central Chamber A, through two Ports (P1and P2), to which a pair of rubber latex gloves of suitable size is fixed, wherein, the Ports P1 & P2 are guarded by suitable gaskets, to make it leak proof; A Pre-filter (6B) ensures supply of dust free air into the clean room and the entire PORTABLE CLEAN ROOM FOR SEPARATION AND MANIPULATION OF STEM CELLS FOR CLINICAL APPLICATION is mounted on a Stainless Steel Stand (9B) having four number castor wheels (10B), on which the SS Stand carrying the entire unit is supported and four number leveling jacks have also been provided for the adjustment and proper balancing of the entire unit with an Exhaust duct (12B) appropriately placed.

2) A PORTABLE CLEAN ROOM FOR SEPARATION AND MANIPULATION OF STEM CELLS FOR CLINICAL APPLICATION as claimed in Claim (1) herein above, wherein the Pressure in Chamber A is maintained at more than that in Chambers Al & A2, at least by 15 pascals

3) A PORTABLE CLEAN ROOM FOR SEPARATION AND MANIPULATION OF STEM CELLS FOR CLINICAL APPLICATION as claimed in Claims (1) & (2) herein above, wherein the sliding plexiglass door separating the Chambers could be either manually operated or optionally could be motor driven.
