Abstract: The present invention relates to the method for in house production of a simple and cost effective 1 Kb molecular DNA ladders. Further, the present invention relates to the method of preparation of a recombinant plasmid construct for the production of 1Kb molecular DNA ladders. Furthermore, the present invention relates to the technology for preparing and producing cost effective 1Kb DNA ladders and their method of production thereof. The DNA ladder is also used to quantitatively analyse target DNA fragments. At present, there are two ways to prepare a DNA ladder: i) amplification by polymerase chain reaction (PCR); and ii) digestion of plasmid DNA by restriction endonucleases.
