A Wound Hydrating Composition For Topical Application And Method Of Preparation Thereof

Abstract: The present disclosure relates generally to wound healing compositions. More specifically, the disclosure provides a wound hydrating composition for topical application comprising amino acids obtained from egg yolk, bovine serum albumin, or their combination, Curcuma longa, glycerine, and 2-amino-2-methylpropanol. The composition supplements nutrients to the wounds and balances the amount of moisturization required for wound healing without drying or slowing the progress of healing. The disclosure also provides a method of preparing the composition.

Specification

The present disclosure relates generally to wound healing compositions. More specifically, the disclosure provides a wound hydrating composition for topical application comprising amino acids, Curcuma longa, and glycerine. The disclosure also provides a method of preparing the composition.
BACKGROUND OF THE INVENTION

It is not an admission that any of the information provided herein is prior art or relevant to the presently claimed invention, or that any publication specifically or implicitly referenced is prior art. [0003] Wound healing process involves expression of various proteins and amino acids along with anti-inflammatory molecules. However, the rigorous processes expressed during deep wound healing results into wound area dryness due to cell apoptosis. This results into slow healing and post healing marks. On the other hand, it is also not recommended to keep the wound moisturized as this also results in slow healing. Therefore, it becomes imperative to supply the wound area with sufficient moisture to fasten wound healing in a proper way by providing cell with supplements, by not letting the wound dry quickly as well as not keeping the wound moisturized for longer time.
[0004] The inventors of the present disclosure overcome such a need for a composition that can balance the moisturization on the wounds to promote healing.
OBJECTS OF THE INVENTION
[0005] An object of the present disclosure is to provide a wound hydrating composition for topical application that supplies nutrients to the wound, hydrates the wound and promotes wound healing.
[0006] An object of the present disclosure is to provide a wound hydrating composition that maintains the balance of not letting the wound dry up and not over hydrating both of which may slow healing.

[0007] Another object of the present disclosure is to provide a method of preparation of the wound hydrating composition.
SUMMARY OF THE INVENTION

simplified form that are further described below in Detailed Description section.
This summary is not intended to identify key features or essential features of the
claimed subject matter, nor is it intended to be used as an aid in determining the
scope of the claimed subject matter.
[0009] Aspects of the present disclosure provide a composition suitable for wound
hydration and healing.
[0010] In an aspect, the present disclosure provides a wound hydrating
composition for topical application comprising active components of amino acids
and Curcuma longa.
[0011] In an embodiment, the composition comprises amino acids obtained from
egg yolk, bovine serum albumin or combination thereof, extracts of Curcuma
longa, glycerine, and 2-amino-2-methylpropanol.
[0012] In an embodiment, the extract of Curcuma longa may be obtained from
rhizomes of Curcuma longa.
[0013] In an embodiment, the composition comprises amino acids obtained from
egg yolk, bovine serum albumin or combination thereof in a range of about
70%w/v to about 80%w/v of the composition, extracts of Curcuma longa in a
range of about 5%w/v to about 10%w/v of the composition, glycerine in a range
of about 7% v/v to about 10% v/v of the composition, and 2-amino-2-
methylpropanol in less than about 2%v/v of the composition.
[0014] In an embodiment, the composition comprises amino acids obtained from
egg yolk, bovine serum albumin or combination thereof in about 70%w/v of the
composition, extracts of Curcuma longa in about 5%w/v of the composition,
glycerine in about 7% v/v of the composition, and 2-amino-2-methylpropanol in
less than 2% v/v of the composition.

[0015] In an aspect, the present disclosure provides a method of preparation of a wound hydrating formulation for topical application, the method comprising the steps of: (a) mixing amino acids obtained from egg yolk, bovine serum albumin or combination thereof in a solvent and preparing an emulsion; (b) adding extracts of Curcuma longa to the emulsion; (c) mixing glycerine dropwise and homogenizing the emulsion; and (d) adding drops of 2-amino-2-methylpropanol and one or more additive(s) followed by lyophilizing to give the formulation. [0016] In another aspect, the present disclosure provides a method of treatment, prophylaxis or amelioration of wounds in a subject by administering an effective amount of the wound hydrating composition.

DETAILED DESCRIPTION OF THE INVENTION

composition for topical application comprising active components of amino acids
and Curcuma longa.
[0036] In an embodiment, the present disclosure provides a composition that
supplements essential nutrients to the wound. Primarily, the composition provides
amino acids that can rebuild the protein structures in the wounds.
[0037] Apart from being the active ingredient, Curcuma longa can also double up
as a preservative. In an embodiment, the composition may comprise whole, an
extract or a part of Curcuma longa.

[0038] In an embodiment, the extract or part may be obtained from the group
comprising of root, leaves, shoot, fruits, rhizome, seed, stem, barks, flower, sap,
bud or combinations thereof of Curcuma longa.
[0039] In an embodiment, the composition comprises amino acids obtained from
egg yolk, bovine serum albumin or combination thereof, extracts of Curcuma
longa, glycerine, and 2-amino-2-methylpropanol.
[0040] Glycerine works as a humectant, emollient or binder; while 2-amino-2-
methylpropanol functions as a buffering agent and surfactant.
[0041] In an embodiment, the composition comprises amino acids obtained from
egg yolk, bovine serum albumin or combination thereof in a range of about
70%w/v to about 80%w/v of the composition, extracts of Curcuma longa in a
range of about 5%w/v to about 10%w/v of the composition, glycerine in a range
of about 7% v/v to about 10% v/v of the composition, and 2-amino-2-
methylpropanol in an amount of less than 2% v/v of the composition.
[0042] In a preferred embodiment, the extract of Curcuma longa may be obtained
from the rhizomes of Curcuma longa. The extract may be obtained by any process
or technique well-known in the art. In an embodiment, the extract may be an
aqueous or alcoholic extract. In a preferred embodiment, the extract may be a dry
extract from rhizomes of Curcuma longa or turmeric powder.
[0043] In an embodiment, the composition comprises amino acids obtained from
egg yolk, bovine serum albumin or combination thereof in about 70%w/v of the
composition, extract of rhizome of Curcuma longa in about 5%w/v of the
composition, glycerine in about 7% v/v of the composition, and 2-amino-2-
methylpropanol in less than 2%v/v of the composition.
[0044] The composition of the present disclosure fastens the process of wound
healing by providing adequate moisture and hydration to the skin such that the
moisturization is balanced between being very high that can increase bacterial
growth and decrease wound healing, or being very low to cause dryness.
[0045] The composition is amino-acid-based such that the wound healing process
is fastened by providing cell with amino acid supplements obtained from natural
sources.

[0046] In an embodiment, the present disclosure provides a wound hydrating
formulation for topical application comprising amino acids obtained from egg
yolk, bovine serum albumin or combination thereof, extracts of Curcuma longa,
glycerine, 2-amino-2-methylpropanol, and one or more additive(s).
[0047] In an embodiment, the additive may be selected from a solvent,
humectants, fragrance, coloring agent, preservatives, antimicrobials, or
combinations thereof. However, a person of skill in the art would understand that
any other additive(s) may be employed without going beyond the spirit and scope
of the present disclosure.
[0048] In an embodiment, the additive may be present in a range of about 0.2%
w/v to about 0.5% w/v of the formulation.
[0049] In an embodiment, the solvent may be selected from water, oil, or
combinations thereof. However, a person of skill in the art would understand that
any other solvent(s) may be employed without going beyond the spirit and scope
of the present disclosure.
[0050] In an embodiment, the humectants may be selected from beeswax, shea
butter, cocoa butter, castor oil, or combinations thereof. However, a person of
skill in the art would understand that any other humectant(s) may be employed
without going beyond the spirit and scope of the present disclosure.
[0051] In an embodiment, the fragrance may be selected from rose, chamomile,
sandalwood, lavender, lemongrass, or combinations thereof. However, a person of
skill in the art would understand that any other fragrance(s) may be employed
without going beyond the spirit and scope of the present disclosure.

organic color including annatto, beta-carotene and turmeric extract or food grade

egg yolk, bovine serum albumin or combination thereof in about 70%w/v of the
formulation, extract of rhizome of Curcuma longa in about 5%w/v of the

methylpropanol in less than about 2%v/v of the formulation, and fragrance in
about 0.5% v/v of the formulation.
[0054] In an embodiment, the formulation is a solid, semi-solid, or liquid. In an
embodiment, the formulation may be a powder, capsule, paste, ointment, lotion,
emulsion, cream, microparticles, or nanoparticles. In a preferred embodiment, the
formulation is a dry powder.
[0055] In some embodiments, the formulation may be used by mixing the dry
powder with a solvent, preferably water and applying on the wound. The
formulation is topical so may be directly applied on the open wound or may be
applied via a cotton swab or clean cloth.
[0056] In another embodiment, the present disclosure provides a method of
preparation of a wound hydrating formulation for topical application, the method
comprising the steps of: (a) mixing amino acids obtained from egg yolk, bovine
serum albumin or combination thereof in a solvent and preparing an emulsion; (b)
adding extracts of Curcuma longa to the emulsion; (c) mixing glycerine dropwise
and homogenizing the emulsion; and (d) adding drops of 2-amino-2-
methylpropanol and one or more additive(s) followed by lyophilizing to give the
formulation.
[0057] In an embodiment, the emulsion may be prepared using a solvent such as
water, oil, or combinations thereof.
[0058] In another embodiment, the present disclosure provides a method of
treatment, prophylaxis or amelioration of wounds in a subject by applying an
effective amount of the wound hydrating composition or formulation on the
wound.
[0059] In an embodiment, the formulation or composition may be employed with
other anti-microbials or wound healing compositions.

Although methods and materials similar or equivalent to those described herein can be used in the practice of the disclosed methods and compositions, the exemplary methods, devices and materials are described herein. It is to be understood that this disclosure is not limited to particular methods, and experimental conditions described, as such methods and conditions may vary. [0062] MATERIALS: The chemicals were obtained from Sigma, Merck, HiMedia, or SRL chemicals. Turmeric extract was procured locally in the form of dry powder.
[0063] EXAMPLE 1: Preparation of wound hydrating formulation for topical application
[0064] A dry powder formulation was prepared as per the composition provided in Table 1. The amino acid was incorporated into an emulsion using water as a solvent. Added dry powder of turmeric extract into the emulsion. Dropwise mixed glycerine till homogenous solution was obtained. Added about 4 drops of 2-amino-2-methylpropanol (AMP) along with the rose fragrance, and lyophilized the paste by freezing for 10 minutes then primary drying, freezing again for 35 minutes then secondary drying for 51 minutes, followed by sealing and capping 1.3 minutes to give the formulation as dry powder, lg of the dry powder was mixed with 5 mL water and applied to the wounds.

[0065] From the foregoing, it will be appreciated that, although specific embodiments of the invention have been described herein merely for purposes of illustration, various modifications may be made without deviating from the spirit and scope of the invention and should not be construed so as to limit the scope of the invention or the appended claims in any way.

We Claim:

1. A wound hydrating composition for topical application comprising amino acids obtained from egg yolk, bovine serum albumin or combination thereof, Curcuma longa, glycerine, and 2-amino-2-methylpropanol.
2. The composition as claimed in claim 1, wherein the composition comprises whole, an extract or a part of Curcuma longa.
3. The composition as claimed in claim 2, wherein the extract or part is obtained from the group comprising of root, leaves, shoot, fruits, rhizome, seed, stem, barks, flower, sap, bud or combinations thereof of Curcuma longa.
4. The composition as claimed in claim 2, wherein the extract is a dry extract
from rhizomes of Curcuma longa or turmeric powder.
5. The composition as claimed in claims 1-4, wherein the composition comprises amino acids obtained from egg yolk, bovine serum albumin or combination thereof in a range of 70%w/v to 80%w/v of the composition, extracts oi Curcuma longa in a range of 5%w/v to 10%w/v of the composition, glycerine in a range of 7% v/v to 10% v/v of the composition, and 2-amino-2-methylpropanol in an amount of less than 2% v/v of the composition.

The composition as claimed in claim 5, wherein the composition comprises amino acids obtained from egg yolk, bovine serum albumin or combination thereof in 70%w/v of the composition, extract of rhizome of Curcuma longa in 5%w/v of the composition, glycerine in 7% v/v of the composition, and 2-amino-2-methylpropanol in less than 2%v/v of the composition.
7. A wound hydrating formulation for topical application comprising amino acids obtained from egg yolk, bovine serum albumin or combination thereof, extracts of

Curcuma longa, glycerine, 2-amino-2-methylpropanol, and one or more additive(s).
8. The formulation as claimed in claim 7, wherein the formulation is a powder, capsule, paste, ointment, lotion, emulsion, cream, microparticles, or nanoparticles.
9. The formulation as claimed in claim 7, wherein the formulation is a dry powder.
10. A method of preparation of a wound hydrating formulation for topical
application, the method comprising the steps of: (a) mixing amino acids obtained
from egg yolk, bovine serum albumin or combination thereof in a solvent and
preparing an emulsion; (b) adding extracts of Curcuma longa to the emulsion; (c)
mixing glycerine dropwise and homogenizing the emulsion; and (d) adding drops
of 2-amino-2-methylpropanol and one or more additive(s) followed by
lyophilizing to give the formulation.
