An Antigen Suspension And Diagnostic Kit For Diagnosis Tuberculosis

Abstract: An antigen suspension comprising an antigen obtained by the steps of incubating a portion of M. tuberculosis wild strain growth in Sauton’s medium at 32 to 40ºC preferably 37ºC for 30 to 45 day till late log phase; collecting and dissolving the pellets there from in a solvent; inactivating the bacteria at 75 Cº to 85º C preferably 80º C for a period of 30 to 60 minutes followed by sonication for the period of 1 to 2 hrs; lyophilising the sonicated extract to free it of moisture; extracting the sonicated material with an extracting solvent and filtering followed by deprotinisation of the filtrate; separating, washing, filtering from the suspension a lower organic phase and removing the solvent from filtrate to yield dry lipid extract; removing the neutral lipids from said dried mixture followed by filtration until the lipids become whitish or colourless followed by filtration, purifying the lipids thus obtained to provide the antigen; phosphatidylcholine solution prepared as per conventional procedure and cholesterol and dye dissolved in alcohol for use in the manufacture of the test reagent for tuberculosis diagnosis. A kit for diagnosis of tuberculosis using the test reagent formed from the antigen suspension.
