Abstract: The present disclosure discloses an anti-biofilm formulation comprising: (a) thymol; (b) citral; (c) at least one plasticizer; (d) at least one drug carrier; (e) at least one stabilizer; and (f) at least one solvent, wherein thymol has a weight percentage in a range of 0.0015-0.0045% with respect to the formulation, and citral has a weight percentage in a range of 0.0015-0.0045% with respect to the formulation. The present disclosure further discloses a process of obtaining the anti-biofilm formulation. Also disclosed is a process of coating the anti-biofilm formulation and its application as a coating material on biomedical devices.
I/We Claim:
1. An anti-biofilm formulation comprising:
a) thymol;
b) citral;
c) at least one plasticizer;
d) at least one drug carrier;
e) at least one stabilizer; and
f) at least one solvent,
wherein thymol has a weight percentage in a range of 0.0015-0.0045% with respect to the formulation, and citral has a weight percentage in a range of 0.0015-0.0045% with respect to the formulation.
2. The anti-biofilm formulation as claimed in claim 1, wherein thymol to citral has
a weight ratio in a range of 0.6:1 to 1.5:1.
3. The anti-biofilm formulation as claimed in claim 1, wherein the at least one
plasticizer is selected from the group consisting of poly (ethylene glycol) (PEG),
sorbitol, hydroxypropyl cellulose, epoxidized soybean oil (ESBO), epoxidized
linseed oil (ELO), vegetable oils, and combinations thereof, having a weight
percentage in a range of 5-10% with respect to the formulation.
4. The anti-biofilm formulation as claimed in claim 1, wherein the at least one drug carrier is selected from the group consisting of poly (vinyl alcohol) (PVC), poly (N-vinyl pyrrolidone), poly (ethylene-co-vinyl acetate), poly (vinyl acetate), poly (methyl methacrylate), polyurethanes, and combinations thereof, having a weight percentage in a range of 2-25% with respect to the formulation.
5. The anti-biofilm formulation as claimed in claim 1, wherein the at least one stabilizer is selected from the group consisting of chitosan, gelatin, starch, pectin, cellulose, and combinations thereof, having a weight percentage in a range of 1-4% with respect to the formulation.
6. The anti-biofilm formulation as claimed in claim 1, wherein the at least one solvent is selected from the group consisting of acetic acid, adipic acid, formic acid, lactic acid, malic acid, propionic acid, succinic acid, and combinations
thereof, having a weight percentage in a range of 1 to 2% with respect to the formulation.
7. The anti-biofilm formulation as claimed in claim 1, wherein the formulation
exhibits anti-biofilm activity against at least one microorganism selected from
the group consisting of Staphylococcus aureus, Pseudomonas aeruginosa,
Acinetobacter baumannii, and Klebsiella pneumonia.
8. A substrate coated with the anti-biofilm formulation as claimed in claim 1.
9. The substrate as claimed in claim 8, wherein the substrate is selected from the group consisting of titanium, stainless steel, glass, ceramic, polystyrene, and combinations thereof.
10. The substrate as claimed in claim 8 is an implantable device selected from the group consisting of baksi's prosthesis, charnley prosthesis, condylar blade plate, ender's nail, grosse-Kempf (GK) nail, kirschner wire, kuntscher nail, moore's pin rush nail, smith peterson (SP) nail, seidel nail, steinmann pin, and swanson prosthesis.
11. A process for obtaining the anti-biofilm formulation as claimed in claim 1, said process comprising:
a) heating at least one drug carrier in a vehicle at a temperature in a range of 70-95°C for a time period in a range of 30-120 minutes to obtain a first mixture;
b) contacting the first mixture with at least one plasticizer at a temperature in a range of 22-30°C for a time period in a range of 60-120 minutes to obtain a second mixture;
c) contacting at least one stabilizer with at least one solvent to obtain a third mixture;
d) contacting the third mixture with the second mixture at a temperature in a range of 22-30°C for a time period in a range of 20-60 minutes to obtain a fourth mixture;
e) contacting citral with thymol for a time period in a range of 60-120 minutes and at a temperature in a range of 22-3 0°C to obtain a fifth mixture; and
f) contacting the fourth mixture and the fifth mixture for a time period in a range of 20-100 minutes at a temperature in the range of 22-30°C for obtaining the anti-biofilm formulation.
12. A process for coating a substrate with an anti-biofilm formulation, said process
comprising:
a) obtaining an anti-biofilm formulation as claimed in claim 1;
b) coating the anti-biofilm formulation on a substrate by at least one coating method selected from the group consisting of spin coating, dip coating, layer-by-layer coating and spray coating to obtain a layer on the surface of the substrate; and
c) drying the substrate of step (b) at a temperature in a range of 30-60°C for
a time period in a range of 30-120 minutes to obtain a coated substrate.
13. The process as claimed in claim 11, wherein the vehicle is water having a weight
percentage in a range of 10 to 98% with respect to the formulation.
