Antibacterial Quinolones

Abstract: A novel methodology of reacting halogenated quinolines with piperazine derivatives using a chemical system comprising of alkali salts of aromatic acids with a ternary solvent mixture having pyridine or an alkylpyridine mixed with a polar solvent represented by hydrophobic alkanols or dimethyl sulfoxide and selecting N, N-dimethylglyoxime or biacetyl as the third member of this innovative chemical system. When reacted this way even equimolar amines, for example, piperazines gave good yields of the N-alkylated quinolones. Another innovation is achieved by the use of moist piperazines, including recycling of the unreacted piperazine solution in pyridines having substantial presence of water by utilizing surprisingly benefic effect of adding a dialkylsulphosuccinate to afford commercially important quinoline antibacterials.
