DESC:FIELD OF THE INVENTION
[0001] The present disclosure pertains to a composition for syndet bar. Particularly the present disclosure relates to a composition comprising parabiotic for syndet based bar. In particular the present disclosure relates to a composition comprising parabiotic and syndet base for bathing bar, especially for sensitive skin, and humans having a compromised skin barrier.
BACKGROUND OF THE INVENTION
[0002] The background description includes information that may be useful in understanding the present invention. It is not an admission that any of the information provided herein is prior art or relevant to the presently claimed invention, or that any publication specifically or implicitly referenced is prior art.
[0003] The word syndet is derived from synthetic combined with detergent. Syndet bars comprise different detergents, also called surfactants or tension-active agents. Technically these detergents, having an affinity for oils, bind to them repelling water, surround dirt with small structures that are then removed by water.
[0004] Syndet bars referred as cleansing bars or dermatological bars occupy a vast majority of market as against the traditional vegetable oil soaps. Traditional vegetable oil based commercial soaps often contain free alkali which increases the pH and makes them harsh and drying to the skin. The long term use of traditional soaps is considered to causes irreversible damage to the acid mantle.
[0005] Syndet bars are generally recommended for cleansing sensitive skin and even baby skin, as they are far more gentle and skin-friendly than traditional soaps.
[0006] From a technical standpoint, syndet bars are considered to have advantages over traditional soap, that syndet allows including additives preferred by large population of consumers like colourants and fragrances that are not suitable to be included in traditional soaps due to the high pH environment.
[0007] However, conventional syndet soap compositions comprise sulphates, parabens, phthalates which are harmful for sensitive skin population with compromised skin barrier function. Often for this reason syndets are now not preferred for babies and those suffering from dermatological conditions.
[0008] Therefore, there is an unmet need in the art to provide a composition for syndet bar which is devoid of harmful constituents like sulphates, parabens, phthalates and still provide effective cleaning properties and without drying out the skin of those having sensitive skin.

Table 1:
Ingredient % Qty
Syndet base 98.80%
Fragrance 0.50%
Parabiotic 0.25%

Table 2:
1 2
Visit 1 36.8 41.3
Visit 2 26.8 25.8
SCORAD Index assess the extent and severity of Atopic Dermatitis (AD) and evaluates improvement in AD management post treatment. In addition to objective signs such as extension, severity of signs and symptoms like erythema, Oedema, lichenification and dryness, SCORAD Index also considers subjective signs like irritability and loss of sleep. After 30 days of skidel bar application, 32.6% reduction in SCORAD Index is observed indicating management of subjective signs and reduction in objective signs.
Example 3: Eczema Area and Severity Index (EASI): Severity Score
[00086] It is observed that 8.3 % reduction in atopic dermatitis severity observed post 30 days of daily skidel bar application as shown in Fig. 2 and below table 3:
Table 3:
1 2
Visit 1 3.0 4.2
Visit 2 3.0 3.6
EASI Score measures the severity of clinical signs in atopic dermatitis and evaluates improvement in AD management post treatment. It assesses four AD characteristics such as erythema, thickness, scratching and lichenification on a scale of 0 (absent) to 3 (severe) in four different part of the body, namely Head & Neck, Upper Limbs, trunk and lower limbs. After 30 days of skidel bar application, 8.3% reduction in EASI score is observed indicating reduction in severity of clinical signs of AD.
Example 4: Staphylococcus Aureus
[00087] It is observed that 25% showed no growth in staphylococcus aureus after 30 days of daily application using skidel bar application as shown in Fig. 3 and below table 4:
Table 4:

Visit 1 No growth Mild growth
Visit 2 No growth Scanty growth
(Staphylococcus aureus is associated with the severity, pathogenesis and exacerbation of AD. Increase in colonization triggers Th2 and IgE response which exacerbates inflammation and itching which further increases risk of various invasive infections. After 30 days of skidel bar application, 25% reduction in growth of Staphylococcus aureus is observed indicating reduction in severity of clinical signs of AD.
Example 5: Visual Analogue Scale (VAS)
[00088] There is 55.5% reduction in pruritus severity observed post 30 days of daily Skidel bar application as shown in Fig. 4 and below table 5:
Table 5:
1 2
Visit 1 4 5
Visit 2 2 2
Itching Score helps to assess severity of pruritis, a disease-defining symptom of atopic dermatitis. After 30 days of skidel bar application, 55.5% reduction in pruritis is observed indicating reduction in severity of clinical signs of AD.
Example 6: Moisture Content
[00089] For affected area, skin hydration at the epidermis and the upper dermis is increased by 24.2% after 30 days of daily skidel bar application as shown in Fig. 5 and below table 6:
Table 6:
1 2
Visit 1 15% 20%
Visit 2 24% 20%
Measuring moisture content is one of the important factors in AD as it helps to assess the skin’s integrity and the degree of skin hydration of the epidermis layer. After 30 days of Skidel bar application, 24.2% increase in skin hydration at the epidermis and the upper dermis is observed which helps in management of clinical signs of AD.
Example 7: Transdermal water loss
[00090] For affected area, transepidermal water loss (TEWL) is decreased by 34% after 30 days of daily skidel foam and Tep AD with parabiotic application as shown in Fig. 6 and below table 7:
Table 7:
1 2
Visit 1 12.8 17.8
Visit 2 9.6 10.6
TEWL helps in assessing the barrier function of skin in atopic dermatitis. It assesses the quantity of condensed water that diffuses across a fixed area of stratum corneum to the skin surface per unit time. Due to dysfunction of skin barrier, patients with AD display higher TEWL. After 30 days of skidel bar application, 34% decrease in TEWL is observed which indicates lesser water loss and better management of clinical signs of AD.
