Device For Extraction, Separation And Detection Of Biomolecules From A Bio Sample

Abstract: The subject matter herein describes a device (100) for extraction, separation, and detection of biomolecules from a bio-sample. The device (100) includes a base platform (102), a cartridge (104) mounted on the base platform for downstream flow based extraction, separation, and detection of the biomolecules from the bio-sample, and a pump (106) and a plurality of valves (108-114) mounted on the base platform and coupled to the cartridge for flowing the bio-sample, a diluting agent, an elution buffer and a wash buffer through the cartridge. The cartridge includes a lysis zone having a lysis channel couplable to electrodes for electrical lysis of the biomolecules flowing through the cartridge, a denaturation zone for denaturation of the biomolecules after the electrical lysis, and a binding and detection zone having a base chamber for preparing a functionalized nanostructure bed with a photoreactive moiety for binding and detection of the biomolecules from the bio-sample.

DESC:DEVICE AND METHOD FOR RAPID EXTRACTION, SCREENING AND DETECTION OF BIOMOLECULES ,CLAIMS:1. A device (100) for extraction, separation, and detection of biomolecules from a bio-sample, the device (100) comprising:
a base platform (102);
a cartridge (104) mounted on the base platform (102) for downstream flow based extraction, separation, and detection of the biomolecules from the bio-sample; and
a pump (106) and a plurality of valves (108, 110, 112, 114) mounted on the base platform (102) and coupled to the cartridge (104) for flowing the bio-sample, a diluting agent, an elution buffer, and a wash buffer through the cartridge (104),
wherein the cartridge (104) comprises:
a lysis zone (P2-P3) having a lysis channel couplable to electrodes for electrical lysis of the biomolecules flowing through the lysis channel;
a denaturation zone (P4-P5) for denaturation of the biomolecules after the electrical lysis; and
a binding and detection zone (P5-P6) having a base chamber for preparing a functionalized nanostructure bed with a photoreactive moiety for binding and detection of the biomolecules from the bio-sample.
2. The device (100) as claimed in claim 1, wherein the cartridge (104) comprises a magnetic separation zone (P3-P4) for magnetic separation of the biomolecules from the bio-sample after the electrical lysis.
3. The device (100) as claimed in claim 1, wherein the cartridge (104) comprises a magnetic separation zone for magnetic separation of the biomolecules from the bio-sample before the electrical lysis.
4. The device (100) as claimed in claim 2 or 3, wherein the device (100) comprises a magnetic block mounted on the base platform (102), wherein the magnetic block is positioned under the magnetic separation zone.
5. The device as claimed in claim 1, wherein the device (100) comprises a first electrical heating unit mounted on the base platform (102), wherein the first electrical heating unit is positioned under the lysis channel of the lysis zone (P2-P3).
6. The device (100) as claimed in claim 1, wherein the device (100) comprises a second electrical heating unit mounted on the base platform (102), wherein the second electrical heating unit is positioned under the denaturation zone (P4-P5).
7. The device (100) as claimed in claim 1, wherein the cartridge (104) comprises:
a sample chamber for holding the bio-sample;
a diluting agent chamber for holding the diluting agent;
a sample mixing zone (P1-P2) having a sample mixing fluid channel coupled to the sample chamber and the diluting agent chamber for mixing of the bio-sample with the diluting agent; and
a binding and functionalization reagent chamber for holding binding and functionalization reagents, wherein the binding and functionalization chamber is coupled to the binding and detection zone (P5-P6).
8. The device (100) as claimed in claim 7, wherein the cartridge (104) comprises:
a first orifice to top-load the bio-sample into the sample chamber; and
a second orifice to top-load the binding and functionalization reagent into the binding and fucntionalization chamber.
9. The device (100) as claimed in claim 7, wherein the device (100) comprises a diluting agent tank (502) coupled to the pump (106), a valve, from the plurality of valves (108, 110, 112, 114), and the cartridge (104) to supply the diluting agent to the diluting agent chamber.

The device (100) as claimed in claim 1, wherein the cartridge (104) comprises an elution buffer chamber, coupled at an end of the lysis zone (P2-P3), for holding the elution buffer, and wherein the device (100) comprises an elution buffer tank (504) coupled to the pump (106), a valve, from the plurality of valves (108, 110, 112, 114), and the cartridge (104) to supply the elution buffer to the elution buffer chamber.

The device (100) as claimed in claim 1, wherein the cartridge (104) comprises a wash buffer chamber, coupled at an end of the lysis zone (P2-P3), for holding the wash buffer, and wherein the device (100) comprises a wash buffer tank (506) coupled to the pump (106), a valve, from the plurality of valves (108, 110, 112, 114), and the cartridge (104) to supply the wash buffer to the wash buffer chamber.
12. The device (100) as claimed in claim 1, wherein the cartridge (104) comprises a biomolecule elution chamber, coupled at an end of the binding and detection zone (P5-P6), for collecting the biomolecules eluted based on detection in the binding and detection zone (P5-P6).
13. The device (100) as claimed in claim 1, wherein the cartridge (104) comprises a debris chamber, coupled at an end of the binding and detection zone (P5-P6), for collecting debris generated by the binding and detection zone (P5-P6).
14. The device (100) as claimed in claim 1, wherein the device (100) comprises a light source for photo-cleavage the photoreactive moiety bound to the biomolecules.
15. The device (100) as claimed in claim 1, wherein the device (100) comprises a power supply mounted on the base platform (102) and coupled to the electrodes for the electrical lysis.
16. The device (100) as claimed in claim 1, wherein the electrodes are made of one of aluminum and gold.
