Abstract: DIETARY SUPPLEMENT COMPOSITION ABSTRACT The present invention is a dietary supplement which includes the composition for oral administration for patients with immune-system related issues including cancer to enhance the immune system. The composition includes Red Poha, Kheel (Puffed Rice), Palm Sugar, Inulin, Whey Protein, Ragi (Finger Millet), Samai (Little Millet) , Thinai (Foxtail Millet) , Kambu (Pearl Millet), Kollu ( Horse Gram), Jowar (Sorghum), Barley, Green Gram (Moong/Mung), Flak Seed, Soya bean Seed, Cinnamon, Fenugreek , Dry Ginger, Saffron, Ellachi (Cardamom), Walnut, Madhulai (Pomegranate), Turmeric, Vettiver (Camel Grass), Garlic, Karunjeeragam (Killi Black Cumin), Wheat Grass, Mavilanga Pattai, Thaneervitan (Neotea Asparagus Root Powder), Chitirai Moola Ver (White Leadwort Root), Vilvam (Indian Bael), Mullukatiri Ver, Sundakai ( Turkey Berry), Paal Muthukkan (Indian Kudzu Root), Pungampattai, Ashwagandha/Amukkara Kizhangu Powder, Aamanaku Veru (Castor Bean/Oil Plant), Mandhara Pattai ( Kachnar), Perumkurambai Ver, Mesha Shringi (Gymnema Sylvestre), Papaya, Custard Apple (Sugar Apple), Sweet Potato, Orilai Ver, Guggul and Neela Kurunji.
DESC:FIELD OF INVENTION
The present invention relates to the field of dietary supplement composition. More specifically it relates to the herbal composition to be used as a dietary supplement to enhance the immune system of cancer patients.
BACKGROUND OF THE INVENTION
Cancer is a group of diseases involving abnormal cell growth with the potential to invade or spread to other parts of the body. These contrast with benign tumors, which do not spread. Cancer begins in your cells, which are the building blocks of your body. Normally, your body forms new cells as you need them, replacing old cells that die. Sometimes this process goes wrong. New cells grow even when you don't need them, and old cells don't die when they should. These extra cells can form a mass called a tumor. Tumors can be benign or malignant. Benign tumors aren't cancer while malignant ones are. Cells from malignant tumors can invade nearby tissues. They can also break away and spread to other parts of the body.
Cancer is not just one disease but many diseases. There are more than 100 different types of cancer. Most cancers are named using their site of origin. For example, lung cancer starts in the lung, and breast cancer starts in the breast. The spread of cancer from one part of the body to another is called metastasis. Symptoms and treatment depend on the cancer type and how advanced it is. Most treatment plans may include surgery, radiation and/or chemotherapy. Some may involve hormone therapy, immunotherapy or other types of biologic therapy, or stem cell transplantation.
The most common types of cancer medications include alkylating agents, Nitrosoureas and antimetabolites. Some examples of antimetabolites include Capecitabine, Cladribine, Clofarabine, Floxuridine, Fludarabine, Gemcitabine, Hydroxyurea and Methotrexate etc. Cancer treatments and cancer can cause side effects. Side effects are problems that occur when treatment affects healthy tissues or organs. They are anemia, appetite loss, bleeding and bruising (Thrombocytopenia), constipation, delirium, diarrhea, edema (swelling), fatigue, fertility issues in boys and men, fertility issues in girls and women, flu-like symptoms, hair loss (Alopecia), infection and Neutropenia, Lymphedema, memory or concentration problems, mouth and throat problems, nausea and vomiting, nerve problems (Peripheral Neuropathy), immunotherapy and organ-related inflammation, pain, sexual health issues in men, sexual health issues in women, skin and nail changes, sleep problems, urinary and bladder problems. The side effects vary from person to person, even among people receiving the same type of cancer treatment.
Dietary modification is essential in accordance with the no-going treatment, several changes will be suggested. Several researches on the effects of following different types of diets, herbs, animal products/ algae during the pre-treatment/ treatment/ post-treatment stages are being studied. Adjuvants are substances which enhance the body's immune response to an antigen. They modify/enhance the activity of the principal medical treatment and can be used in addition to the principal treatment, especially in cancer treatment to avoid secondary growth.
There are several other patents that have developed a dietary supplement such as JP5118125B2 discuss about dietary supplements with high calorie dietary supplements that contain milk protein isolate as the primary or sole protein source. This comprises of a protein source, including milk protein isolate and / or canola plant protein, a fat source, and a carbohydrate source, the caloric content from about 2.25 to about 3 per milliliter. It is between 25 kilocalories, provides nutritional supplements viscosity of less than about 120 centipoise.
Another patent US6054128A relates to dietary supplements formulated to reduce risk or progression of cardiovascular disease by addressing the factors that influence the physiology and pathology of the heart, arteries and veins in a concurrent and synergistic manner. The supplement comprises of a combination of vitamin B6, vitamin B12 and folic acid.
The patent JP5094981B2 relates to a carbohydrate composition as a dietary supplement that is essential to produce a glycoprotein that is accurately configured and therefore functions properly. A dietary supplement composition comprising a nutritionally effective amount of aloe vera extract; and a nutritionally effective amount of gucci gum; glucosamine; arabinogalactan; tragacanth gum; and a glucose source.
To overcome the side-effect issues faced in the existing systems, the present invention formulates a dietary supplement without any side effects for the patients suffering from cancer and to generally improve the immune system which includes specific combinations of herbs and herbal extracts, particularly of oriental plants, and vitamins, minerals and nutrients.
Inulin is a type of prebiotic. It is not digested or absorbed in the stomach. It stays in the bowel and helps certain beneficial bacteria to grow. Finger millet, also known as ragi in India is an annual herbaceous plant widely grown as a cereal crop in the arid and semiarid areas in Africa and Asia.The Little Millet (samai) is believed to have originated in India. It is grown and used for food almost exclusively in our country. It belongs to the group of small millets (other members being Proso, Kodo, Barnyard and Finger millets), which are said to be nutritionally superior to rice and wheat. It is suitable for all ages of people. High phosphorus in Samai helps with fat metabolism, body tissue repair and creating energy, especially good for people who has low body mass. It is rich in phyto-chemicals and it has excellent antioxidant properties.
Foxtail millet, scientific name Setaria italica, is an annual grass grown for human food. Being highly nutritious for children and adults, it acts a great energy source. It is rich in fiber content and minerals like protein, iron, calcium, potassium, magnesium, zinc. Enriched with vitamin B 6, it helps in the regulation of the healthy nervous system.
The rich Iron content in Kambu aids in improving the hemoglobin level in the blood and prevents anemia. It is also rich in fiber, protein, minerals such as magnesium, zinc, manganese, folic acid, amino acids, lecithin, potassium, B complex vitamins, and calcium.
Kollu (Horse gram) is an excellent source of protein (17.9–25.3 %), carbohydrates (51.9–60.9 %), essential amino acids, energy, low content of lipid (0.58–2.06 %), iron molybdenum.
Jowar is globally known as the “new quinoa” for its gluten-free and whole-grain goodness. It goes by the name of Sorghum in English. In India, jowar is a staple food item. It is Antioxidant-Rich and good source of various phytochemicals like tannins, phenolic acids, anthocyanins, phytosterols and policosanols.
Moong/Mung or green gram is small, round, olive-green colored legumes with a sweet flavor. They are one of the healthiest sources of plant protein and are found in different forms like whole, split, de-husked (yellow) and ground. The plethora of nutrients present in green gram dal includes folate, thiamine, riboflavin, niacin, pyridoxine, zinc; magnesium and calcium help in lowering inflammation. While loaded with phytonutrients flavones it shows potent antioxidant properties that assist in triggering immunity.
Flaxseed is a plant-based food that provides healthful fat, antioxidants, and fiber. Flaxseed is a good source of many nutrients, including protein, fiber, omega-3 fatty acids, thiamine, and copper.
Soybean, (Glycine max), also called soja bean or soya bean, annual legume of the pea family (Fabaceae) and its edible seed. The soybean is high in fibre, high in protein, low in saturated fat, cholesterol free, lactose free, a good source of omega-3 fatty acids, a source of antioxidants, high in phytoestrogens.
One tablespoon (11.1 grams) of whole fenugreek seeds contains 35 calories and several nutrients namely Fiber: 3 grams, Protein: 3 grams, Carbs: 6 grams, Fat: 1 gram, Iron: 20% of the Daily Value (DV), Manganese: 7% of the DV and Magnesium: 5% of the DV. Anticancer potential of diosgenin was explored in in-vitro experiments. HT-29 human colon cancer cells were used and it was seen that diosgenin inhibited the proliferation of cells along with the induction of apoptosis.
Saffron is a spice derived from the flower of Crocus sativus, commonly known as the "saffron crocus". This spice is an excellent source of minerals like copper, potassium, calcium, manganese, iron, selenium, zinc and magnesium.
The nourishing components in elaichi (cardamom) make it a superior nutritional food. It is inherently rich in proteins and healthy fats, besides containing remarkable quantities of vitamin B6, vitamin B3, vitamin C, zinc, calcium, magnesium and potassium.
A walnut is the edible seed of a drupe. Walnuts are an excellent source of several vitamins and minerals, including Copper, Folic acid, Phosphorus, Vitamin B6, Manganese and Vitamin E. karunjeeragam contains protein: 8 of the 9 essential amino acids that are combined to make “whole” protein. Black Seed also has vitamin B1, vitamin B2, vitamin B3, folic acid, calcium, iron, copper, zinc and phosphorous.
Wheatgrass is the freshly sprouted first leaves of the common wheat plant, Nutrients in wheatgrass include: Vitamin A. Vitamin C. Vitamin E. A 1-ounce serving of wheatgrass juice contains Calories: 10, Protein: 2 grams, Fat: 0 grams, Carbohydrates: 2 grams, Fiber: 0 grams and Sugar: 2 grams.
A teaspoon of ground cinnamon weighing 2.6 g contains energy: 6.42 calories, carbohydrates: 2.1 g, calcium: 26.1 milligrams (mg), iron: 0.21 mg, magnesium: 1.56 mg, phosphorus: 1.66 mg, potassium: 11.2 mg and vitamin A: 0.39 micrograms. The aqueous extract and the fraction of cinnamon (procyanidins) from HPLC inhibit vascular endothelial growth factor subtype 2 (VEGFR2) kinase activity, thereby inhibiting the angiogenesis involved in cancer. Cinnamaldehydes have been synthesized and tested as inhibitors against angiogenesis CB403, a chemical that can be synthesized from 2'-hydroxycinnamaldehyde derived from cinnamaldehyde, can inhibit tumor growth. Cinnamic aldehyde inhibits the activity of NF-?B and the production of tumor necrosis factor alpha (TNFa-) induced interleukin-8 (IL-8) in A375 cells.
The anti-cancer properties of ginger are attributed to gingerol, which is found in large amounts in raw ginger. A form known as [6]-gingerol is viewed as especially powerful. Individuals at normal risk for colorectal cancer, 2 grams of ginger extract per day significantly reduced pro-inflammatory signaling molecules in the colon.
Organo-sulfur compounds found in garlic have been identified as effective in destroying the cells in glioblastomasTrusted Source, a type of deadly brain tumor. It is reported that three pure organo-sulfur compounds from garlic – DAS, DADS, and DATS – demonstrated efficacy in eradicating brain cancer cells, but DATS proved to be the most effective.
Giloy, a diterpenoid from Tinosporacordifolia has been reported for its chemopreventive potential in diethylnitrosamine (DEN) induced hepatocellular carcinoma (HCC) in rats by decreasing anti-oxidant activities via SOD, CAT and detoxification enzymes like GSH, GPx and subsequent increase in the activities of the hepatic markers ((Serum glutamic oxaloacetic transaminase)SGOT, (Serum Glutamic Pyruvate Transaminase) SGPT, LDH) and decreased serum transaminase level thus confirming its anti-tumor effects and promising application as a potent chemo preventive drug for HCC.
Curcumin from Haridra —improves heart health and prevent against Alzheimer’s and cancer. Curcumin may affect cancer growth and development.
The anticancer components in Ashwagandha i.e. withaferins-A (WA) and Withanolide, are responsible for apoptosis induction and exhibit anti-cancerous potential.
Thaneervittan Kilangu (Neotea Asparagus Root Powder) contains saponins, flavonoids, Terpenes and glycosides that possess anti-cancer activity. All these anti-cancer compounds are majorly present in Thaneervittan Kilangu roots.
White Leadwort root or Chitirai Moola Ver contains an acrid crystalline principle called ‘Plumbagin’ that is a powerful irritant and has well marked antiseptic properties. Plumbagin is one of the major bioactive widely investigated for anticancer and cytotoxic potential. plumbagin lacks toxicity on normal colon cells and showed its striking anti-survival effect on colon cancer cells. Plumbagin, which was previously reported as anticancer agent.
Vilvam ( Indian Bael) is loaded with a myriad of nutrients which include vitamins A, B1, B2, C and minerals calcium, potassium and iron. Aegle marmelos leaf extracts were effective in inhibiting the growth of leukemic K562, T-lymphoid Jurkat, B-lymphoid Raji, erythroleukemic HEL, melanoma Colo38, and breast cancer cell lines MCF7 and MDA-MB-231. Leaf extract is also shown to have antineoplastic effects on the Ehrlich ascites carcinoma in Swiss albino mice. 1-hydroxy-5,7-dimethoxy-2-naphthalene-carboxaldehyde (Marmelin) present in A. marmelos inhibiting the growth of epithelial cancer cells. Lupeol, another compound present in A. marmelos, possesses anti-neoplastic effects on various human neoplastic cell lines
Paal muthukkan or Kudzu root, also called Japanese arrowroot. In its raw form, kudzu root resembles other root tubers, such as potatoes or yams. It has tan skin, white flesh, and an oblong shape. Kudzu root contains more than 70 plant compounds, some of which may be responsible for the root’s potential health benefits. It is cytotoxic to various cancer cell lines like K562, MCF7, HEPG2, Jurkat cells, and A549 (Bulugonda et al., 2017). Furthermore, the anticancer and apoptotic potential of the hydroalcoholic tuber extract of P. tuberosa was investigated by cell viability assay. The extract showed a 50% inhibition of cell viability against human colon carcinoma (HT-29) cells at a concentration of 63.91 µg/ml.
Pungam pattai is a deciduous tree that grows to about 15-25 meters in height with a large canopy that spreads equally wide. It contains flavanoids, tocophorols, phenols and lauric acid. Ricinus communis, the castor bean or castor oil plant. It has shown activity against specific cancer cell lines such as melanoma, MCF7 (Breast cancer), HepG2 (Hepatic cancer), PC3 (Pancreatic cancer), and cervical cancer.
Cytotoxic effects of R. communis on various cell lines such as colon cancer, liver cancer, breast cancer, cervix cancer, ovarian cancer (OVCAR-5), skin melanoma (B16F10) and prostate cancer are studied. At lower concentrations of about 100 µg/mL, the R. communis extract was tested against these cancer cell lines and was shown to be effective.
Mantharai or Kachnar is rich in vital essential nutrients of vitamin C, B vitamins, key minerals of calcium, phosphorous, magnesium, iron, zinc, besides requisite macronutrients of reducing-sugar carbohydrates, dietary fibers, proteins and healthy unsaturated fats.
Meshashringi also known as Gymnema sylvestre is a shrub to promote metabolic wellness by maintaining blood sugar levels. The herb's bitter taste and astringent property.
Guggul comes from the resin of the Commiphora mukul, a small thorny tree that is known as the tree of myrrh. Guggul is the gum resin obtained from a variety of plants. It contains vitamin C, flavonoids, polyphenols, tannins and saponins, along with other potent plant compounds. These compounds help fight oxidative stress caused by free radicals, which are molecules that can damage cells and contribute to chronic disease. Guggul contains certain components that have anti-inflammatory and anti-arthritic properties. It suppresses the chemicals that lead to pain and inflammation in case of Rheumatoid arthritis
Strobilanthes kunthiana, known as Kurinji or Neelakurinji in Malayalam and Tamil, is a shrub.
MTT assay showed that the chloroform extract of callus (Perumkurambai Ver) has potent anticancer potential. The plant has a promising anticancer activity against human colon epithelium, lung carcinoma, and epidermoidal carcinoma cell lines. It was found to possess Topo as well as DNA polymerase inhibitory activity. C fragrans has revealed the presence of alkaloids, such as camptothecin (CPT), chonemorphine, and funtumafrine. CPT is a plant derived monoterpene indole alkaloid, currently is in clinical use for the treatment of various types of cancer.
The herbal ingredients described herein are commercially available in oriental natural food shops, natural health stores or pharmacies, or can be prepared as dry powders. All of the herbal components contained in this product are in the Generally Considered Safe (GCS) category. But the specific combination of requisite amounts of the ingredients for our dietary supplement formulation is not available or easily discernable by naturalists/ people skilled in the art.

The main objective of our present invention is to provide a dietary supplement for the cancer patients and to enhance the immune system. Further objective of the present invention is to provide a general herbal dietary supplement with immunomodulatory effect.
One aspect of the present invention is to provide an herbal dietary supplement without any side effects for cancer patients. Another aspect of the present invention is to provide a simple and low-cost herbal powder for cancer patients of all age groups.
Another aspect of the present invention to provide a dietary supplement which includes specific combinations of herbs and herbal extracts, particularly of oriental plants, and vitamins, minerals and nutrients.

The principles of operation, design configurations and evaluation values in these non-limiting examples can be varied and are merely cited to illustrate at least one embodiment of the invention, without limiting the scope thereof.

The current embodiment of the present invention which provides a dietary supplement for enhancing the immune system of cancer patients comprising of 13 herbs, grains, millets, fresh fruits, dry fruits, seeds and other ingredients (100) and its formulation is disclosed in Fig.1. All the ingredients are cleaned and dried under shadow to remove the moisture (101). It is processed in to a fine dry powder (103) and formulated as an oral preparation for the cancer patients (104).
The present invention provides a dietary supplement in unit dosage form adapted particularly for oral consumption by a subject in need of enhancement of the immune system. Nutritional supplementation with selected herbs and nutrients can improve certain aspects of the immune and blood system. The dietary supplement comprises an effective mixture of herbal extracts and powders, vitamins, minerals, amino acids, and various excipients. The dietary supplement described herein may be consumed as a tablet or capsule or may further be prepared as a composition for being consumed as a beverage wherein the supplement is prepared as an essentially dry mixture which is mixed with an ingestible liquid to form an aqueous beverage. Furthermore, the current embodiment of our invention has been formulated without the addition of any preservatives.
Each of these herbs or herbal extracts included in the current embodiment of our invention is known to have various effects beneficial to human health. However, it has been discovered that these herbs and herbal extracts, when consumed in the combinations described herein, have enhanced beneficial effects regarding immune enhancement. The present invention is a natural formulation which has further significant value in preventing undesirable side effects of radiation treatments and chemotherapy.

In addition to the active components described above, each composition may further include pharmaceutically acceptable medium. The dietary supplement compositions can be made into various forms suitable for oral administration, for example powder, granules, capsules, tablets, or liquid.
Studies have been conducted to determine the Anti-cancer adjuvant property of the dietary supplement by in vitro screening using MTT Assay, specifically one embodiment of our current invention labelled as Karrovita. The MTT Cell Proliferation Assay measures the cell proliferation rate and conversely, when metabolic events lead to apoptosis or necrosis, the reduction in cell viability. MTT Assay is a colorimetric assay that measures the reduction of yellow 3-(4,5-dimethythiazol- 2-yl)-2,5-diphenyl tetrazolium bromide (MTT) by mitochondrial succinate dehydrogenase. The assay depends both on the number of cells present and, on the assumption, that dead cells or their products do not reduce tetrazolium. The MTT enters the cells and passes into the mitochondria where it is reduced to an insoluble, dark purple-colored formazan crystals. The cells are then solubilized with a DMSO and the released, solubilized formazan reagent is measured spectrophotometrically at 570 nm. Cell viability was evaluated by the MTT Assay with three independent experiments with six concentrations of compounds in triplicates. Cells were trypsinized and preform the Thetryphan blue assay to know viable cells in cell suspension. Cells were counted by haemocytometer and seeded at density of 5.0 X 10 3 cells / well in 100 µl media in 96 well plate culture medium and incubated overnight at 37 0 C. After incubation, the old media was removed and fresh media 100 µl added with different concentrations of Karrovita with standard Cisplatin in represented wells in 96 plates. After 48 hours, the media solution is discarded and the fresh media with MTT solution (0.5 mg / mL-1) was added to each well and plates were incubated at 37 0 C for 3 hrs. At the end of incubation time, precipitates are formed as a result of the reduction of the MTT salt to chromophore formazan crystals by the cells with metabolically active mitochondria. The optical density of solubilized crystals in DMSO was measured at 570 nm on a microplate reader. The IC50 value was determined by using linear regression equation i.e. y = mx+c. Here, y = 50, m and c values were derived from the viability graph. The formulation treated with MCF 7cells showing the IC50 values is given under table 2. The dietary supplement (Karrovita) alone, and the dietary supplement formulation combination with standard drug Cisplatin shows the IC50 10.71µg/ml compared to cisplatin alone 12.53µM as shown in Table 2.

b. Cleaning, drying under shadow to remove the moisture content in all ingredients (101);
c. Processing of ingredients in to a fine dry powder (102) and
d. Oral supplement formulation for the cancer patients (103).
3. The dietary supplement composition as claimed in claim 1, wherein the supplement is further comprising of a pharmaceutically acceptable medium.
4. The dietary supplement composition as claimed in claim 1, wherein the supplement is formulated for oral administration.
5. The dietary supplement composition as claimed in claim 1, wherein the formulations for oral administration can be powders, granules, capsules, tablets or liquids.
