Genomic Signature And Pcr Primers For Specific Detection Of Streptomyces Species

Abstract: Bacteria of the genus Streptomyces are renowned for their prolific ability to synthesize diverse bioactive therapeutic compounds, having a significant bearing on the availability of drugs and therapeutics for the prevention and cure of human diseases. In this study, we undertook to , elucidate the conserved genomic regions with the aim of identifying signature nucleotide elements within the genomes of 18 distinct Streptomyces species. The genomic and pangenomic characteristics of the genus Streptomyces have enabled a deeper understanding of their diversity. The conserved regions within the genomes were identified through multiple genome alignments by employing the MAUVE tool, revealing conserved blocks across different species. Analysis of the uniqueness of sequences of the identified conserved regions was conducted by using BLASTn. Remarkably, among the total conserved blocks, only one region demonstrated uniqueness specific to thy genus Streptomyces. In-silica and in-vitro validation of the signature sequence within the Streptomyces genus was carried out by applying computational methodologies and by specific amplification of the target sequence using specifically designed primer pairs. The method involved identifying a highly conserved region within the target sequence to ensme specificity across different Streptomyces species. The designed primers exhibited perfect matches with the target region, thereby minimizing the likelihood of mismatches or gaps during amplification. This approach enhanced the accuracy and efficiency of PCR-based detection and analysis of the genus Streptomyces. The invention also relates to diagnostic kits for the detection of Streptomyces spp. in samples. The process and the primers were validated by agarose gel electrophoresis and DNA sequencing.
