Abstract: Hydrogen Bond Surrogate (HBS)-Helicomimics for Triplet Code Selective Binding to dsDNA The present disclosure provides synthetic peptides that contain about 6-9 amino acids and a hydrogen bond surrogate between two amino acids which facilitates a stable alpha-helical configuration of the peptide. The peptides of the present disclosure bind a DNA triplet code comprising at least two thymine bases with nanomolar affinities. The present disclosure also provides a conjugate comprising the peptide and a moiety, wherein the moiety is linked at the N or C terminus of the peptide. The present disclosure further provides pharmaceutical compositions comprising the peptides or the conjugates described herein.
