Abstract: In a first aspect, the invention provides the composition of substrate for the artificial cultivation of Cordyceps mushrooms. It provides the method of producing the same in a shorter duration with enhanced medicinal properties and active ingredients. The composition of the substrate comprises of proportions of carbon, nitrogen & amino acids. In the present invention Bambusoideae seeds in combination with silk protein sericin forms the novel substrate combination. This amino acid composition of sericin being superior in terms of Aspartic acid which is crucial for mycelial growth can replace the otherwise expensive source-peptone. The present invention depicts the optimum combination of Bambusoideae seeds as the major carbohydrate source of 60% to support mycelial growth, Sericin a natural source of amino acids critical for mycelial growth & fructification which includes 16% Aspartic acid and 30% Serine. This combination depicts enhanced mycelial growth, enhances biomass production and productivity, improves extracellular Cordycepinc and other bioactive production and productivity, increases yield, enhances fructification, reduces duration of production to 30 days as opposed to 365 days in nature & enhances the active ingredient composition of Cordycepin, Cordycepic acid, polysaccharides etc.,
Claims:1. A Nutrient substrate composition comprising nutritionally enhanced combination of raw materials for artificial cultivation of medicinal mushrooms
2. The composition in claim 1 wherein, the composition constitutes 50 parts of Bambusoideae seeds
3. The composition in claim 1 wherein, the composition constitutes 1 part of Sericin protein which comprises amino acids such as Aspartic acid, Serine and glutamic acid among other 18 amino acids
4. The composition in claim 2 wherein, the Bambusoideae seeds constitute 60% carbohydrate, Vitamin B6, Calcium, Phosphorous, Potassium
5. A synergistic combination comprising optimum Carbon and Nitrogen source wherein, the medicinal mushrooms possess enahnced pharmacological properties, bioactive ingredients, increased biomass yield and faster rate of fructification
, Description:The shortcoming of the above prior art is overcome by the technical intervention in the present invention, by utilizing nutritionally enriched naturally available resources to promote quicker growth, increased yield and richer bioactive concentration. Making this a feasible, cost effective production technique. The culture method techniques for mother culture activation, nutrient media solution for culture growth and inoculation into novel substrate formulation for culture growth. The shortcoming in the carbon & nitrogen which are primary essential components for the culture growth is overcome through a novel combination of vegetarian substrate.
The conventional or traditional method of artificial cultivation of Cordyceps uses Rice as the major substrate. The present invention provides a kind of cordyceps plantation substrate preparation by the combination of agricultural byproducts that is simple, cost effective, increased rate of production that completely meets the growth period of Cordyceps with desired nutritional composition. The substrate of the present invention ensures rapid mycelial growth, high biological transformation ratio & quality product. Substrate of the present invention cultivates Cordyceps under artificial condition and provides the starting material of a large amount of high-quality Cordyceps.
In this present invention the Bambusoideae seeds is used as a major substrate. This rich carbohydrate source is complemented with Sericin liquid, a rich source of amino acids like aspartic acid, serine etc., This novel combination complements each other in increasing the rate and yield of production and further, does not require the supplementation of other vitamins and minerals, making it completely natural. Cordyceps cultivated through this technique has an edge when compared to the conventional combination in terms of duration of production, number of fruiting bodies produced, weight of the biomass produced and content of the bioactives produced.
Highest mycelial growth is observed in Cordyceps cultivated on substrates with specific Carbon sources and they may be selected from monosaccharides and disaccharides like D(−)-Fructose, D(+)-Galactose, D(+)-Glucose, D(+)-Cellobiose, Lactose, Maltose, Sucrose, polysaccharides like Dextrin, Malt extract, Pectin, Soluble Starch etc., and Alditols like D-Mannitol and D-Sorbitol. Additionally, amino acids play a crucial role in mycelial growth & fructification and these may be selected from the range of Amino Acids which contribute for the same like Aspartic Acid, Glutamine, Serine etc., The right combination of carbon, complex organic nitrogen, inorganic nitrogen, amino acids, vitamins and minerals play an important role in ensuring economical production of Cordyceps mushrooms with enhanced biomass and yield. The above given carbon, nitrogen etc., sources are only exemplary and further selective combination and sources may form part of the present invention.
The source of the Carbon, nitrogen and amino acid are optimized based on the requirement for optimal growth.
In one embodiment of the present invention the natural vegetarian substrate is Bambusoideae seeds. Bambusoideae seeds are grown out of dying bamboo shoots. Bambusoideae seeds is a rich source of various nutrients carbohydrates, proteins, amino acids, fibre, vitamins and minerals, has a low glycemic index, so it is a good option for those with diabetes, has low or no-fat, so a healthy substitute for overweight and obese people. It is rich in vitamin B complex which can be beneficial for pregnant women and loaded with minerals like calcium and phosphorous which can be a remedy for those suffering from joint pain. Bambusoideae seeds contains 218mg of phosphorous. Bambusoideae seeds is a rich source of Carbohydrates: 59%, and promotes faster and richer mycelial growth. It is also used to treat menstrual disorders, respiratory disease and infections. Hence, it is a factory of wellness and when used in combination with Cordyceps has miracle medicinal properties.
In one embodiment of the invention sericin, high molecular weight viscous protein obtained from the silk fibroin, composed of polypeptide with 18 kinds of amino acids. Majority of the amino acid is Serine, followed by aspartic acid and glutamic acid. The combination of these amino acids supports rapid mycelial growth.
The above mentioned complex is such that the medicinal properties of the Bambusoideae seeds fused together with that of Cordyceps results in enhanced pharmacological properties, rapid and dense mycelial growth. Additionally, the critical amino acids present in Sericin increases the biomass yield in 30 days as opposed to 365 days in nature and 60 days in traditional artificial cultivation. In the present invention the growth matrix or substrate constitutes Bambusoideae seeds comprising carbohydrates 60%, protein 7%, fibres 10%, Calcium, Phosphorus, & Vitamin B6 with Sericin comprising amino acids Serine 30% and Aspartic Acid 16%,. This combination provides the optimal combination of C/N ratio, essential minerals and vitamins which ensures higher yield, works as growth promoters for faster initiation of fructification. This reduces the need to supplement the combination with additional vitamins and minerals to promote growth, making it completely natural. It also ensures enhanced pharmacological properties on par with the naturally occurring cordyceps with enhanced properties and superior to the conventional techniques.
In preparation method preferably comprises by mass 50 part soaked & dried Bambusoideae seeds, 1 part sericin liquid replacing the conventional peptone– combination autoclaved and soaked in nutrient media, stirred evenly. Described Bambusoideae seeds is unhusked form of Bambusoideae seeds.
In accordance to embodiments of the present invention the combination of Bambusoideae seeds and Sericin liquid provide an optimum substrate for cultivation of Cordyceps mushrooms. The Cordyceps cultivation have rapid dense mycelial growth, uniformity in fructification, enhanced colour saturation, improved biological transformation ratio and consistent product.

In order to implement the present invention and for better understanding, particular embodiments is being explained below through non-limiting examples. It is also understood that the modification of the detail may be done without deviating from the scope of the invention.
Examples of composition of the substrate to produce desired mycelial yield & bioactive composition may be used:
Bambusoideae seeds comprising 60% of Carbohydrate, calcium, phosphorous, Potassium, Vitamin B6 and dietary fibres constituting 50 parts & 1 Part Sericin, subjected to steam at 121 ℃ for 15 min. Followed by culture inoculation with seed culture, the culture is subjected to cultivation in artificial conditions of alternate dark and light cycles emulating the natural conditions required for growth, resulting in biomass production, mycelial growth, fructification in 30 days.
