Abstract: The present invention provides a nutritional drink composition for boosting immune system providing rich anti-oxidants, comprising of : a predefined quantity of fruit juice; a predefined quantity of extract from the herb Centella asiatica; and a predefined quantity fortifying agents. The ready to consume nutritional drink composition of the present invention is tasty and palatable and ensure patient compliance so as to provide necessary actives to the body. The nutritional drink composition also provides a high Oxygen Radical Absorbance Capacity (ORAC Value of 4620 µmol TE/200ml) for good health. In an embodiment of the present invention, provides a method of preparation of beverage composition rich in anti-oxidants and boosting immune system. DESC:Field of Invention
The present invention relates to the field of beverage composition and formulation, in particular relates to an anti-oxidant & immunity booster nutritional drink for general health and well-being formulated with an herb and natural fruit juice acting synergistically to provide a high anti-oxidant & immunity boosting benefit without any attenuating side-effects. The present invention also provides a proprietary process for making the said nutritional drink with immunity boosting properties to overcome fatigue due to oxidative stress and enabling faster recovery from infections.
Background of the Invention
In today’s world, majority of the population irrespective of age are suffering from various health issues such as low immunity, antibiotic resistance, nutritional deficiencies, increase stress level etc. Every day humans are subjected to exposure to environmental hazards like UV radiation, polluted air, water and exposure to a variety of toxins, stress, herbicides, pesticides, preservatives or additives in processed foods, unbalanced diets, obesity, which contribute to free radical accumulation and negatively affect our bodies with exacerbating signs of premature aging.
Free radicals are unstable molecules which take or scavenge electrons from other molecules. Free radicals are highly reactive chemicals that attack molecules by capturing the electrons and thus modifying chemical structures resulting in reducing immunity. When it takes an electron from a molecule, this second molecule becomes a free radical in return. This triggers a chain reaction where free radicals steal an electron from the nearest molecule, in turn creating a subsequent free radical. Ultimately, this cycle of free radical formation damages a living cell and results in many of the changes seen in the body. Exposure to free radicals is unavoidable in today’s fast paced world.
While the current social, environmental and industrial milieu does trigger free radical accumulation, there is much evidence to show that every system in the body can and do produce free radicals in the natural process of human physiology. For instance, Haemo metabolism has a chemical transitional component from ferric to ferrous and this produces OH ions one of the most dangerous molecules that could produce oxidative stress. During cell damage or break down of food, free radicals are generated for any morbidity.
Antioxidants are substances that reduce oxidative damage by neutralizing free radicals there by reducing the stress factor. Natural antioxidants may be defined as molecules that prevent cell damage against free radicals and are critical for maintaining optimum health. Every individual requires adequate levels of antioxidant defenses in order to avoid the harmful effect of an excessive production of reactive oxygen species (ROS) and to prevent damage to the immune cells. Since the Body will not have enough antioxidants to counter free radicals, they have to be supplemented to fight the menace of free radicals to stay fit and healthy for both sick and healthy person.
High antioxidant levels and low oxidative stress are associated with good health. A healthy and balanced diet loaded with strong antioxidants is the most effective way to deal. Plant-based foods are the best sources of antioxidants, which include fruits, vegetables, whole grains, nuts, seeds, herbs and spices are all useful sources of antioxidants.
Several researches have worked on the area of using herbs /food/drink/beverages and so on to counter oxidative stress.
US patent application 20190247325 provides Methods and compositions for reducing oxidative stress comprising administering an amount of a composition, said composition comprising an encapsulated cannabinoid compound and a terpene, thereby reducing oxidative stress in said subject, wherein said composition comprises an antioxidant.
European Patent 1740196 discloses Compositions and method for alleviating inflammation and oxidative stress in a mammal, An antioxidant-promoting composition comprising active ingredients comprising Bacopa monniera extract; Silybum marianum (milk thistle) extract, Withania somnifera (ashwagandha) powder, Camellia sinensis (green tea) extract, and Curcuma longa (turmeric) extract.
US patent US9889171B2 provides a composition for the topical application on a mammalian skin comprises one or more Brassica plant extracts selected from the group consisting of: Brassica juncea extract, Brassica oleracea italica extract, Brassica oleracea capitata extract, Brassica oleracea botrytis extract, and Brassica oleracea acephala extract. The composition further comprises one or more selected from the group consisting of: Curcuma longa extract, curcuminoids, tetrahydrocurcuminoids, metabolites of curcuminoids or tetrahydrocurcuminoids, and derivatives of curcuminoids or tetrahydrocurcuminoids. The composition further comprises one or more selected from the group consisting of: Camellia oleifera extract, Camellia sinensis extract, green tea extract and white tea extract; Wasabia japonica extract; Bacopa monnieri extract; Silybum marianum extract; and one or both of Piper nigrum extract or tetrahydropiperine.

All of these are believed to be significant contributors to the preventive effects of fruits through their antioxidant & immune boosting effect.
In recent times there has been a resurgence of interest in herbs rich in Meso-nutrients that have been safely used in herbal brews and beverages.
It is clear that there is a definite need for system that provides a daily dose of meso-nutrients with anti-oxidants that is readily consumed by people for relieving oxidative stress. While traditional knowledge does provide options for anti-oxidant & immunity boosting formulations with meso-nutrients, it is well-known that they are very unpopular due to their unpalatability. Still another object of the present invention is to provide a ready to consume nutritional drink composition that has zero free radicals and ensures a property of scavenging free radicals.
It is another object of the present invention is to provide a ready to consume nutritional drink composition that provides a daily allowance of essential minerals and vitamins, anti-oxidants in precise amounts, and a very high level of satiety.

The present invention provides a nutritional drink composition for boosting immune system and method of preparation thereof. The nutritional drink composition for boosting immune system and providing rich anti-oxidants according to an embodiment of the present invention, comprises of : a predefined quantity of fruit juice; a predefined quantity of extract from the quarantined herb Centella asiatica; and a predefined quantity fortifying agents. Wherein the said fruit juice, Centella asiatica extract and fortifying agents act synergistically to provide a rich meso-nutrients contributing Antioxidant and Immune Boosting Property. A Well balanced composition of natural ingredients which provides rich Anti-oxidant and immune boosting properties to the body to overcome oxidative stress caused by environmental hazard, unhealthy fast food, polluted air etc.
The present invention provides all three categories of nutrients namely Macro-nutrients (Carbohydrates, Protein, Dietary Fibres etc), Micro Nutrients (Vitamins & Minerals), meso-nutrients (Triterpenoids, Asiaticoside, Quercetin, Catechin & epicatechin, Anthocyanin, Phloretin, Beta-carotene, Polyphenols), which are required for the healthy body.
In an embodiment of the present invention, provides a method of preparation of nutritional drink composition rich in anti-oxidants and boosting immune system, comprises steps of : dry blending of fortifying agents; Preparing Sugar Syrup with Malic acid, Ascorbic acid, Sodium Citrate, Mineral, fruit Aroma, and Additive Mixture in a Sugar Syrup Tank, with feed hopper; Solution obtained is recirculated through High shear mixer for 15 minutes; Addition of Centella asiatica extract slowly in slurry form and further recirculated through High shear mixer for 15 minutes ; filtering all the solution through kitten filters(20-200 micron) to blending tank; & Addition of Vitamin Mixture slowly and mixed using an agitator; Addition of Flavour Mixture slowly and mixed using an agitator in blending tank; Performing batch standardization using treated water; Filtration and transferring the product to Ready Beverage Tank; Sterilization of the bulk solution stored in the Ready Beverage Tank; and Packing of the sterilized product packed into Form, Fill Seal in Tetra pack Laminate.
In the preferred embodiment of the present invention, Wherein said nutritional drink composition is packed in sterile condition in Tetrapak. The said package protects the nutritive properties of the ingredients maintaining high palatability and extended shelf life free from chemical preservatives.

Centella asiatica is proven to possess rich medicinal properties like antioxidant, memory and immunity booster, but unfortunately due to their acrid / pungent / bitter taste cannot be taken directly.

• dry blending of fortifying agents;
• Preparing Sugar Syrup with Malic acid, Ascorbic acid, Sodium Citrate, Fruit juices, Mineral, fruit Aroma, and Additive Mixture in a Sugar Syrup Tank, with feed hopper;
• All above ingredients are recirculated in high shear mixer for 15 minutes
• Wet blending by further adding Centella asiatica extract in slurry form;
• Centella asiatica slurry is recirculated in High shear mixer for 15 minutes
• filtering the solution through kitten filters (20-200 micron);
• blending of all the ingredients to obtain an uniform solution;
• Addition of Vitamin Mixture to the blending tank slowly and mixed using an agitator;
• Addition of Flavour Mixture slowly to the blending tank and mixed using an agitator;
• Volume makeup is done to the batch size accordingly, with the addition of High Purity water;
• Performing batch standardization using high purified water;
• Filtration and transferring the product to Ready Beverage Tank (RBT);
• Sterilization of the bulk solution stored in the Ready Beverage Tank;
• Packing of the sterilized product packed into Form using Aseptic packing technology; and
• QC release.
In the preferred embodiment of the present invention, Wherein dry blending of Vitamin mixture includes Thiamine Hydrochloride, Pyridoxine Hydrochloride, Calcium Pantothenate and Nicotinamide at 50% – 60% Relative Humidity (RH), maintaining a temperature of 30--32°Cin normal light radiation.
In the preferred embodiment of the present invention, Wherein dry blending of Vitamin mixture includes Natural Beta Carotene (10 % Dispersible), Riboflavin 5 Phosphate Sodium and Folic Acid in 50 – 60 % RH maintaining a temperature of 30 to 32° C without visible radiation conditions.
In the preferred embodiment of the present invention, Wherein dry blending of minerals mixture includes Zinc Sulphate, and Sodium Selenate are dry blended in an RH of 50 – 60 % maintaining a temperature of 30-to-32°Cin normal light conditions.
In the preferred embodiment of the present invention, Wherein dry blending of Additives includes Caramel colour are dry blended in an RH of 40%-60% maintaining a temperature of 30° C to 32°C in normal light conditions.
In the preferred embodiment of the present invention, Wherein wet blending of Flavours, Tocopherol Acetate and other are done in an RH of 40%-60% maintaining a temperature of 30° C to 32°C in normal light conditions.
In the preferred embodiment of the present invention, wherein said batch standardisation is done by checking the Bulk Solution for Sensory test like, Colour, Appearance & taste and Physical properties like Total Soluble Solids (TSS), Acidity and pH.
In the preferred embodiment of the present invention, wherein said sterilization performed is High Temperature Short Time ( HTST) sterilization, followed by rapid cooling which protects the nutritional drink’s sensory characteristics and nutritional value.
In the preferred embodiment of the present invention, wherein the synergism of the nutritional drink composition is highlighted with its high ORAC value of 4620 µmol TE/200 ml that promotes good health and overall wellbeing.
HTST- (High Temperature Short Time): Pasteurization is performed to kill spoilage causing microorganisms prior to filling containers, in order to make the products safer. Here, the liquid moves in a controlled, continuous flow while subjected to temperature of 105 ± 1 0C, for about 51 seconds, followed by rapid cooling to 25-30 0 C. HTST pasteurization is an effective method of making beverages safe for consumption, without undergoing changes to either its sensory characteristics or its nutritional value. The most important determinants to ensuring to keep the good quality of ensuring time- temperature conditions, reducing or eliminating post pasteurization contamination.

,CLAIMS:We Claim,
1. A Nutritional drink composition with anti-oxidants and immunity boosters wherein the different herbal components synergistically act to provide a high Oxygen Radical Absorbance Capacity (ORAC) value of 4620 µmol TE/200ml, comprising of :
fruit extracts of quarantined herb Centella asiatica in the range of 0.02g to 0.04g wt% and 0.04 to 0.06 parts per 100 parts;
fruit juice concentrate in the range of 0.03-0.1 wt% and 1.4 to 1.5 parts per 100 parts;
0.00014 to 0.1 parts per 100ml of fortifying vitamins;
0.001 to 0.01 parts per 100 ml of fortifying minerals;
10 to 15 parts of sweetening agent;
0.07 to 0.01 parts of colouring and flavouring agents.
2. The Nutritional drink composition with anti-oxidants and immunity boosters as claimed in claim 1, wherein said fortifying vitamins is selected from group consisting of Beta Carotene, Vitamin B1, Vitamin B2, Vitamin B3, Vitamin B5, Vitamin B6, Vitamin B9, Vitamin C Vitamin E and/or combination thereof.
3. The Nutritional drink composition with anti-oxidants and immunity boosters as claimed in claim 1, Wherein said fortifying minerals is selected from group consisting of Zinc, Magnesium, Calcium, Selenium and/or combination thereof.
4. The Nutritional drink composition with anti-oxidants and immunity boosters, as claimed in claim 1, Wherein said sweetening agent is selected from group consisting of Sugar, Dextrose, Natural Fruit Sugar, Fructose, Honey, Stevia and/or combination thereof.
5. The Nutritional drink composition with anti-oxidants and immunity boosters, as claimed in claim 1, wherein said group of fortifying vitamins are added in the range of :
a) Beta Carotene forming 0.0014 to 0.002 parts parts per 100 ml;
b) Vitamin B1 contributing 0.00017 to 0.0002 parts per 100 ml;
c) Vitamin B2 is found in 0.00018 to 0.00025 parts per 100 ml;
d) Vitamin forming B3 0.00055 to 0.0008 parts per 100 ml;
e) Vitamin B5 making up 0.00055 to 0.0007 parts per 100 ml;
f) Vitamin B6 constituting 0.00012 to 0.0002 parts per 100 ml;
g) Vitamin B9 found in 0.000058 to 0.0000090 to parts per 100 ml;
h) Vitamin C contributing 0.05 to 0.08 parts per 100ml; and
i) Vitamin E found in 0.0006 to 0.001 parts per 100 ml
6. The Nutritional drink composition with anti-oxidants and immunity boosters, as claimed in claim 1, wherein said group of fortifying minerals are added in the range of :
a. 0.00006 to 0.00009 Parts of Manganese sulphate in 100ml;
b. 0.002 to 0.003 parts of Zinc Sulphate in 100ml;
c. 0.0000018 to 0.000002 parts of Sodium Selenate in 100ml;
d. 0.25 to 0.35 Parts of Sodium Citrate in 100ml.
7. The Nutritional drink composition with anti-oxidants and immunity boosters, as claimed in claim 1, wherein said fortification of the nutritional drink composition comprises of :
a. 10 to 15 parts of sweetening agent;
b. 0.15 to 0.25 parts of Malic acid; and
c. 0.1-0.15 parts of coloring and 0.3-0.6 ml of flavoring agents.
8. A method of preparation of Nutritional drink composition with anti-oxidants and immunity boosters, comprising steps of:
a. dry blending of fortifying agents;
b. Preparing Sugar Syrup with Malic acid, Ascorbic acid, Sodium Citrate, Fruit juices, Mineral, fruit Aroma, and Additive Mixture in a Sugar Syrup Tank, with feed hopper;
c. Above ingredients are recirculated in high shear mixer for 15 minutes;
d. Wet blending is carried out by adding Centella asiatica extract in slurry form;
e. Centella asiatica slurry is recirculated in High shear mixer for 15 minutes
f. filtering the solution through kitten filters (20-200 micron);
g. blending of all the ingredients to obtain a uniform solution;
h. Addition of Vitamin Mixture to the blending tank slowly and mixed using an agitator;
i. Addition of Flavour Mixture slowly to the blending tank and mixed using an agitator;
j. Performing volume makeup with the addition of High Purity water;
k. Performing batch standardization using high purified water;
l. Filtration and transferring the product to Ready Beverage Tank (RBT);
m. Sterilization of the bulk solution stored in the Ready Beverage Tank; and
n. Packing of the sterilized product packed into Form using Aseptic packing technology.

The method of preparation of Nutritional drink composition with anti-oxidants and immunity boosters, as claimed in claim 8, wherein dry blending of Vitamin mixture includes Thiamine Hydrochloride, Pyridoxine Hydrochloride, Calcium Pantothenate and Nicotinamide at 50% – 60% Relative Humidity (RH), maintaining a temperature of 30--32°Cin normal light radiation.
10. The method of preparation of Nutritional drink composition with anti-oxidants and immunity boosters, as claimed in claim 8, Wherein dry blending of Vitamin mixture further includes Natural Beta Carotene (10 % Dispersible), Riboflavin 5 Phosphate Sodium and Folic Acid in 50 – 60 % RH maintaining a temperature of 30 to 32° C without visible radiation conditions.

The method of preparation of Nutritional drink composition with anti-oxidants and immunity boosters, as claimed in claim 8, Wherein dry blending of minerals mixture includes Zinc Sulphate, and Sodium Selenate are dry blended in an RH of 50 – 60 % maintaining a temperature of 30-to-32°Cin normal light conditions.
12. The method of preparation of Nutritional drink composition with anti-oxidants and immunity boosters, as claimed in claim 8, Wherein dry blending of Additives includes Caramel colour are dry blended in an RH of 40%-60% maintaining a temperature of 30° C to 32°C in normal light conditions.
13. The method of preparation of Nutritional drink composition with anti-oxidants and immunity boosters, as claimed in claim 8, Wherein wet blending of Flavours, Tocopherol Acetate and other are done in an RH of 40%-60% maintaining a temperature of 30° C to 32°C in normal light conditions.

The method of preparation of Nutritional drink composition with anti-oxidants and immunity boosters, as claimed in claim 8, wherein said batch standardisation is done by checking the Bulk Solution for Sensory test like, Colour, Appearance & taste and Physical properties like Total Soluble Solids (TSS), Acidity and pH.
15. The method of preparation of Nutritional drink composition with anti-oxidants and immunity boosters, as claimed in claim 8, wherein said sterilization performed is High Temperature Short Time ( HTST) sterilization, followed by rapid cooling which protects the nutritional drink’s sensory characteristics and nutritional value.
