Abstract: ABSTRACT “POLYNUCLEOTIDE ASSOCIATED WITH SEQUITERPENOID SYNTHASE AND USES THEREOF” The cDNA encoding a polypeptide associated with Sesquiterpenoid synthase (AiSTS) Biosynthesis is provided herein. The present disclosure also provides recombinant DNA molecule comprising the polynucleotide molecules as disclosed in the disclosure, recombinant plasmid comprising the DNA recombinant molecule, and recombinant host cells are also provided. Method of generating transgenic plants, and transgenic plants comprising a cDNA encoding a polypeptide associated with Sesquiterpenoid synthase (AiSTS) Biosynthesis and transgenic plants comprising the DNA recombinant molecule of the present disclosure are also provided.

1. An isolated cDNA fragment encoding a polypeptide having amino acid sequence as set forth in SEQ ID NO: 9.
2. The isolated cDNA as claimed in claim 1, wherein the polynucleotide sequence of the cDNA is as set forth in SEQ ID NO: 8, wherein said cDNA encodes a polypeptide having sesquiterpenoid synthase activity.
3. A recombinant DNA construct comprising a cDNA fragment encoding a polypeptide having amino acid sequence as set forth in SEQ ID NO: 9, operably linked to a promoter, wherein said polypeptide has sesquiterpenoid synthase activity.
4. The recombinant DNA construct as claimed in claim 3, wherein the polynucleotide sequence of the cDNA fragment is as set forth in SEQ ID NO: 8.
5. A recombinant vector comprising a recombinant DNA construct as claimed in claim 3 or 4.
6. A recombinant host cell comprising a recombinant DNA construct as claimed in claim 3 or 4.
7. A recombinant host cell comprising a recombinant vector as claimed in claim 5.
8. The recombinant host cell as claimed in claim 6 or 7, wherein the host cell is a bacterial or a fungal cell.
9. A method of enhancing the content of a-farnesene in a plant, said method comprising:
a. transforming a plant cell with a recombinant DNA construct as claimed in claim 3 or 4 or a recombinant host cell as claimed in claim 7.
b. selecting and developing a transgenic plant with enhanced content of a-farnesene.
10. The method as claimed in claim 9, wherein the plant cell is transformed using a method selected from the group consisting of Agrobacterium mediated transformation, protoplast mediated transformation, particle gun bombardment, in-planta transformation, and liposome mediated transformation.
