Process For Conversion Of Lactose To Galacto Oligosaccharides

Abstract: The present disclosure relates to a process which uses a combination of a lactase and a metal salt and suitable enhancers to obtain high percentage conversion of lactose to galacto-oligosaccharides. The process results in enhanced conversion and thereby reduction of lactose along with good galacto-oligosaccharide yield in a medium. Also disclosed is a product obtained by the disclosed process, which has high galacto-oligosaccharide and reduced lactose content.

Specification

Claims:1. A process for conversion of lactose to galacto-oligosaccharides in a medium, said process comprising:
(a) obtaining a medium comprising lactose;
(b) adding almond extract to the medium to obtain a first mixture;
(c) obtaining a premix of a lactase and a salt;
(d) heating the premix to obtain a preheated premix;
(e) adding the preheated premix to the first mixture obtained in step (b) to obtain a second mixture; and
(f) incubating the second mixture at a temperature in the range of 3-8 ? for a time period in the range of 4-8 hours to obtain a product with lower concentration of lactose as compared to the medium.
2. The process as claimed in claim 1, wherein lactose concentration in the medium is in the range of 0.5-20%.
3. The process as claimed in claim 1, wherein the medium is selected from the group consisting of milk, skimmed milk, partly skimmed milk, whey protein, casein, lactose, and combinations thereof.
4. The process as claimed in claim 1, wherein the medium is milk.
5. The process as claimed in claim 1, wherein the almond extract concentration in the first mixture is in the range of 0.05-0.5%.
6. The process as claimed in claim 1, wherein the premix comprises 1-10 units/g of the lactase and 0.005-0.05% of the salt with respect to the medium.
7. The process as claimed in claim 1 and 6, wherein the salt is selected from the group consisting of KCl, MgSO4, MnSO4, NaCl, CaCl2, and combinations thereof.
8. The process as claimed in claim 1, wherein heating of the premix is at a temperature in the range of 35 ?-45 ? for a time period in the range of 2-10 minutes.
9. The process as claimed in claim 1, wherein lactose conversion in the medium is in the range of 50-90%.
10. The process as claimed in claim 1, wherein incubating of the second mixture leads to conversion of lactose to galacto-oligosaccharides, monosaccharides, and combination thereof.
11. A product, obtained from the process as claimed in any of the claims 1-10, wherein the product has galacto-oligosaccharide content in the range of 14-21%.
12. A product comprising lactose content in the range of 0.17-8%, and galacto-oligosaccharide content in the range of 14-21%.
