DESC:RENOFLUTHRIN INDOOR SPACE SPRAY COMPOSITION

FIELD OF THE INVENTION
The present invention relates to a novel indoor space spray composition comprising pyrethroid
insecticide as an active ingredient. Particularly, the present invention relates to a novel and
5 effective insecticidal composition comprising Renofluthrin alone or in combination with at
least one another insecticide for delivery via aerosol or trigger spray or single push, wherein
the said composition exhibits significant flying and crawling insect control activity.
BACKGROUND OF THE INVENTION
Pest control in and around the home is a never-ending issue. Mosquitoes and other crawling
10 and flying insects need to be controlled to avoid the spread of vector-borne diseases. Flying
insects such as mosquitoes, flies and the like, and crawling insects such as cockroaches and red
ants are known to carry disease causing bacteria on their bodies. Flying insects cause great
human sufferings and is a global problem. Diseases like malaria, filariasis, chikungunya,
Dengue fever, yellow fever, Japanese encephalitis, Ross River virus, Burma forest virus,
15 Murray Valley encephalitis etc, are known to spread through mosquitoes. Crawling insects,
such as ants and cockroaches, are also responsible for health problems. Cockroaches may play
a role as carriers of intestinal diseases, such as diarrhoea, dysentery, typhoid fever and cholera.
In addition, control of dust mites cannot be neglected as they are the most common indoor
allergens. They are tiny organisms that can barely be seen by naked eye and feed on house dust
20 and moisture in the air. They are potentially capable of triggering asthma in humans.
Spray cans, oil sprays, mosquito coils, and slow-release vaporising systems including mats and
liquid vaporisers are the most commonly utilized techniques to control household insect
menace. Neem oil, Pyrethrin, Malathion, and carbaryl are all common household pesticides.
However, prolonged exposure to the odour or toxic chemical emissions from commonly
25 available commercial pesticide products may constitute a health risk. Negative health effects
from overexposure and an increase in vector resistance to these commonly used insecticides is
a matter of concern.
Pyrethrum is an insecticide of natural origin, and is endowed with very positive characteristics,
such as high insecticide activity and a very low toxicity for mammals. However, natural

3

pyrethrum is readily oxidized by air in the presence of light (photooxidation) and is easily
degraded, and therefore it is not suited for insecticidal applications.
Numerous compounds have been synthetized, which have a slightly modified structure of
natural pyrethrum, in order to preserve the insecticide action and cause low toxicity to
5 mammals.
Whilst some of these compounds (synthetic pyrethroids) possess the characteristics mentioned
above, conventionally used aerosol compositions may have high flammability, inhalational
toxicity and may cause damage to the environment due to the need of formulating at a
perpetually higher active concentration. The aerosols are designed to work even in closed
10 spaces hence the risk of being exposed to such high amount of insecticidal composition can be
perceived as a risk to health of the user. Thus, it is necessary to formulate a suitable composition
having improved efficacy along with protecting the molecules as much as possible from photo-
oxidation, ensuring a high persistence of the insecticide action at lower concentration of active.
Recently, the inventors of IN4121/MUM/2013 have developed an active ingredient
15 'Renofluthrin' which is found to be effective in-home insecticidal formulations. Renofluthrin is
found to be effective and practical for use in existing known formulation types and also has the
potential to adapt for developing new formulations and delivery systems. Active ingredient,
'Renofluthrin' is an isomeric composition of 2,3,5,6-tetrafluoro-4- (methoxymethyl)benzyl3-
(2,2-dichlorovinyl)-2,2-dimethylcyclopropane carboxylate, wherein total trans isomers (1R, 3S
20 and1S, 3R in native form and almost in equal amounts) ranging from 90% to 99.90% and total
cis isomers (1R, 3R and 1S, 3S in native form and almost in equal amounts) ranging from 0.1%
to 10%.
Hitherto, there is a need in the art to formulate an indoor space spray composition which is
safer to use and economically advantageous. These objectives can be achieved by providing an
25 insecticidal composition that can achieve improved knock down efficacy at lower amount of
active as compared to the conventionally used insecticides.
Moreover, there is a further need in the art for an insecticidal pyrethroid composition that has
both insecticidal as well acaricidal properties.
Therefore, the present invention aims to overcome the problems in the prior art and provide a
30 stable and effective indoor space spray composition comprising Renofluthrin alone or in
combination with other insecticides/acaricides known in the art.

Toxicity Profile of Renofluthrin
In an insecticide / pesticide compound, toxicity and efficacy play a vital role. New insecticide
compounds are based on their effect on target pests and their Toxicology profile.
15 Acute Inhalation Toxicity:
In mosquito repellent and flying insect repellent products, Inhalation Toxicity is very
important. Most flying insect repellent products are based on evaporation and vapours of the
active ingredient. Household insecticide product forms like coils, mats, aerosol sprays, trigger
sprays, liquid evaporators, other forms of burning and vaporising formulations are based on
20 evaporation of the active ingredient. Concentration in air of the insecticide and its evaporation
are the most important aspects in flying insect repellent product.

14

Inhalation toxicity is measured in 'Lethal concentration' LC50 values. LC50 (lethal
concentration) is expressed as weight of test substance per standard volume of air (i.e. mg / L).
Higher the mg/L value of LC50, lower the inhalation toxicity and safer the product.
Known insecticide compounds with similar mode of action have the following reported LC50
5 values:
• Insecticide 'Prallethrin' LC50 is 0.658 mg/L (reported in literature in unit of measurement 658
mg/m3 which in mg/L is 0.658)
• Insecticide 'Transfluthrin' LC50 is 0.513 mg/L (reported in literature in unit of measurement
513 mg/m3 which in mg/L is 0.513)
10 • Insecticide 'Metofluthrin' LC50 is reported as > 1.08 and < 1.96 mg/L
These results are published and reported for the respective insecticides.
Acute Inhalation Toxicity Study of Renofluthrin was performed in Rats as per OECD guideline
No. 403. In Globally Harmonized System of Classifications and Labelling of Chemicals
(GHS), for Dust & mists (mg/1), the insecticide 'Renofluthrin' of the present invention falls in
15 category 4 which is > 1.0 and < 5 mg/L.
The insecticide 'Renofluthrin' of the present invention has LC50 greater than 3.81 mg/L. which
shows that Renofluthrin has the lowest Inhalation toxicity amongst all the compounds
commonly used as active ingredient in household insecticide products.
Bio-efficacy and activity of pesticidal aerosol composition comprising Renofluthrin and
20 its Combination.
The comparative, bio-ef?cacy of Reno?uthrin aerosol composition, on target pests has been
established vis-a-vis commonly marketed aerosol composition containing d-Trans Allethrin
and Transfluthrin”. Mosquito species ‘Aedes aegypti’ is a major vector of dengue fever,
chikungunya and yellow fever viruses, and other viral diseases. The effectiveness and ef?cacy
25 of the present aerosol composition was tested using a Peet Grady chamber.
The effectiveness / ef?cacy of mosquito repellent formulation is normally measured in “Knock-
down time” (KT) in minutes of percentage of the test population of mosquitoes i.e. KT50
values, referring to knock down time of 50% of the test insects quantity. Lower the time in
minutes required for KT50, higher the effectiveness of the insecticide. Results can vary based
30 on study conditions, mosquito species, and protocols.

15

A comparative study for bio-efficacy evaluation of the aerosol composition of the present
invention versus other aerosol compositions known in the art was conducted against ‘Aedes
aegypti’ species of mosquitoes and German cockroach; and the results obtained are presented
in the tables below:
1) Bioefficacy comparison of d-Trans Allethrin 0.25% Aerosol and Renofluthrin 0.03%
Aerosol against Aedes aeygypti in Peet Grandy chamber
5 Method: Experiments were conducted with 2 replicates for each aerosol against mosquitoes,
Aedes aegypti. The aerosol was primed in fume hood/out of the test area for 2-3 times before
testing. After priming, pre weight of the aerosol can was recorded. 0.65 ± 0.10g discharge of
the formulated product was sprayed in a single application towards the centre of the chamber
through a port provided at one of the side walls of the chamber. The knock down observations
10 were recorded at 1 minute interval up to 10 mins followed by 2 mins interval till 20 mins and
thereafter 5 mins interval till 60 minutes.
Table 1: Bioefficacy comparison of d-Trans Allethrin 0.25% Aerosol and Renofluthrin
0.03% Aerosol against Aedes aeygypti in Peet Grandy chamber
Sr. No. Product Information Parameter Knock Down Time

1. d-Trans Allethrin 0.25% Aerosol KT50 7.20
KT90 10.71
2. Renofluthrin 0.03% Aerosol KT50 6.41
KT90 10.66

15 Result: From the table above, it is evident that Renofluthrin aerosol formulation showed better
effect as compared to d-trans allethrin aerosol formulation. The knock down time for both 50%
and 90% of mosquitoes was lesser in Renofluthrin as compared to d-Trans allethrin at an active
concentration of 0.03% as against 0.25%.
Thus, Renofluthrin 0.03% Aerosol demonstrates excellent efficacy at a lower concentration of
20 active.
2) Initial and Residual Bioefficacy testing of Aerosols against Free Flying, Aedes aegypti
mosquitoes in Peet Grandy chamber.

16

Method: mosquitoes were released in the Peet Grady chamber and post acclimatisation period
the aerosol product were sprayed @ 0.65 grams ± 0.1 grams and knockdown readings were
recorded up to 60 minutes and the data was plotted to calculate the KT50 and KT 90 values
using SPSS software. The product was sprayed once and the readings were recorded after 0
5 (Fresh), after 4 hours, after 8 hours and after 10 hours from start of spray and the residual
efficacy was evaluated.

Result:
RNF 0.05% Aerosol formulation showed 30% mortality at 24 hrs.
However, RNF 0.03% + CYPERMETHRIN 0.1% Aerosol formulation showed enhanced
efficacy with 80% mortality as compared to Deltamethrin 0.02% + Allethrin 0.13% Aerosol
(55% mortality at 24 hrs.) already known in the art.
10 Thus, Renofluthrin 0.03% + CYPERMETHRIN 0.1% Aerosol formulation demonstrates
excellent control of crawling insects even at low doses of actives.
