Abstract: The present invention relates to the process for the preparation of solid forms of Betrixaban maleate (1). The present invention further relates to the process for the preparation of amorphous forms, crystalline forms and solid dispersions of Betrixaban maleate (1) with suitable pharmaceutical acceptable excipients.
1. A process for the preparation of amorphous form of Betrixaban maleate (1),comprising:
a) providing a solution of Betrixaban maleate (1) in a suitable first solvent;
b) stirring and filtering the reaction mass;
c) adding the filtrate to a suitable pre-cooled second solvent;
d) stirring the reaction mass at suitable temperature; and
e) isolating and drying amorphous form of Betrixaban maleate (1) using a suitable technique.
2. The process as claimed in claim 1, wherein the suitable first solvent used is selected from a group but not limited to organic acids comprising acetic acid, formic acid or the like.
3. An alternative process for the preparation of amorphous form of Betrixaban maleate (1), comprising:
a) providing a solution of Betrixaban maleate (1) in a suitable solvent or mixtures;
b) heating the reaction mass;
c) stirring and filtering the reaction mass;
d) removing the solvent from the filtrate at a suitable temperature; and
e) isolating and drying amorphous form of Betrixaban maleate (1) using a suitable technique.
4. A process for the preparation of crystalline form of Betrixaban maleate (1)
comprising of the following steps:
a) providing Betrixaban maleate (1) in a suitable first solvent;
b) stirring and filtering the reaction mass;
c) adding second solvent to the filtrate; and
d) isolating and drying crystalline form of Betrixaban maleate (1) using a suitable technique.
5. A process for the preparation of solid dispersions of Betrixaban maleate (1)
with apharmaceutically acceptable excipient comprising of the following steps:
a) providing Betrixaban maleate (1) in a suitable solvent;
b) adding a suitable pharmaceutically acceptable excipient;
c) stirring and filtering the reaction mixture;
d) removing the solvent by heating or any other suitable technique; and
e) isolating and drying amorphous solid dispersion of Betrixaban maleate (1) using a suitable technique.
6. The process, as claimed in claims 1,3,4 and 5 wherein the Betrixaban maleate
(1) used herein is having purity greater than 99.0% and can be prepared by a
process comprising:
a) adding betrixaban and maleic acid to a solvent mixture;
b) heating to suitable temperature; and
c) isolating Betrixaban maleate (1).
7. The process as claimed in claim 6, wherein the suitable pharmaceutically
acceptable excipients used in the present invention may be selected from a
group comprising of lactose, sorbitol, mannitol, saccharose, cellulose, methyl
cellulose, ethyl cellulose, microcrystalline cellulose, polyethylene glycol
(PEG), polyvinylpyrrolidone (PVP), polyvinylpyrrolidone K-30, (PVP K-30),
polyvinyl acetate hydroxyethyl cellulose (HEC), hydroxy propyl methyl
cellulose (HPMC), hydroxypropyl cellulose (HPC), hydroxy propyl methyl
cellulose acetate succinate (HPMC-AS), y-cyclodextrin, hydroxypropyl beta cyclodextrin (HPBCD), sulfobutylether-[3-cyclodextrin (SBCED) , Soluplus , Eudragit or the like.

The process as claimed in claims 1, 2, 3, 4, 5 and 6 wherein the solvent used for preparation of Betrixaban maleate (1) may be selected from a group comprising of water, methanol, ethanol , ethylene glycol, isopropyl alcohol, benzyl alcohol, acetone, acetonitrile, dioxane, ethyl acetate, n-propyl acetate, n-butyl acetate, t-butyl acetate, isobutyl acetate, toluene, heptane, hexane, cyclohexane, 1,4-dioxane, diethyl ether, methyl tert-butyl ether, tetrahydrofuran, and mixtures thereof.

The process as claimed in claims 1, 2, 3, 4 and claim 5 wherein the solid forms of Betrixaban maleate (1) obtained may be isolated by methods which include but not limited to cooling, precipitation, re-crystallization, concentrating the mass, adding seed crystals , evaporation, flash evaporation, simple evaporation, fast solvent evaporation, distillation, rotational drying, spray drying, thin-film drying, agitated thin-film drying, freeze-drying or the like, preferably precipitation and fast solvent evaporation was used in the present invention.
10. The process as claimed in claim 1,3,4 and 5 wherein the suitable drying technique is selected but not limited to tray dryer, air tray drier, vacuum oven, air oven, cone vacuum dryer, rotary vacuum dryer, fiuidized bed dryer, spin flash dryer, flash dryer, at temperatures less than about 100° C, less than about 80° C, less than about 60° C, less than about 50° C, or any other suitable temperatures.
