Abstract: SYNERGISTIC INSECTICIDAL FORMULATION OF ETHIPROLE AND ACETAMIPRID ABSTRACT The present invention discloses a synergistic insecticidal formulation of Ethiprole and Acetamiprid, wherein the said composition is in Wettable Dispersible Granule (WG) formulation comprising: Ethiprole Technical in the range of 35-50% w/w, Acetamiprid Technical in the range of 15-25% w/w, Dispersing agent in the range of 2-5% w/w, Surfactant in the range of 0.5-5% w/w, Suspending agent in the range of 2-5% w/w, Fillers in the range of 4-8% w/w and adjuvants. FIELD OF THE INVENTION:

The present invention relates to a synergistic insecticidal formulation of Ethiprole and Acetamiprid. The present invention particularly relates to a synergistic insecticidal formulation of Ethiprole and Acetamiprid, wherein the said composition is Wettable Dispersible Granule (WG) formulation and the process of preparation thereof.

BACKGROUND OF THE INVENTION:

Pesticides are the chemicals which are applied on plants to kill or eliminate fungus, bacteria, insects, snails, slugs, or weeds. Pesticides are broadly used in agriculture and have facilitated a significant increase in yield of crops and productivity.

In order to control the harmful effects of insects on crops, an extensive range of active insecticide compounds have been developed. Many active insecticide compounds in different formulations are commercially available. The efficacy of these conventional insecticide compounds is restricted in controlling certain groups of insect. Thus, the problem of controlling a broad spectrum of insects by a single product still exists. Presently, several insecticidal formulations are used in comparatively high concentrations and doses. This leads to a higher cost of the product, higher residues on plants and soil causing toxicity and have deleterious effects on the desirable insect species and other living organisms.

Many researchers have developed a variety of chemicals and their formulations which are effective in controlling the insects from destroying the crops. Nevertheless, there is a continuous requirement for novel compositions and methods to control the broad range of insects without harming the natural enemies, ecosystem and water bodies. Specifically, in view to achieve economical results and eco-friendly compositions, many efforts have been made to maximize the performance of known active ingredients. Also, there is a need for insecticidal compositions which demonstrate synergistic effect at lower cost.

Ethiprole (5-amino-1- (2, 6-dichloro - a, a, a-trifluoro-p-tolyl) - 4 - ethyl sulfinylpyrazole - 3 - carbonitrile) is a synthetic insecticide that belongs to the class of ‘Phenylpyrazole’. It was first discovered by Bayer Crop Science Japan (formerly Rhone-Poulenc Agrochemicals) in 1994.

Ethiprole is a broad spectrum insecticide for control of chewing and sucking insects like psylla, plant bugs, weevils, leaf miners, aphids, stink bugs, plant hoppers and grasshoppers. It is majorly used as a seed treatment or foliar spray in rice, fruits, cotton and vegetables.

Ethiprole was first disclosed in patent US5814652A and chemically represented as below structure:

The molecular formula of Ethiprole is C13H9Cl2F3N4OS and molecular weight is 397.197 g/ mol and having water solubility of 0.0092 g/L at 20oC and is non-volatile. Ethiprole is the insecticide from IRAC (Insecticide Resistance Action Committee) Group 2B class. Ethiprole is a chiral molecule. The technical material is an isomeric mixture of the S- and R-enantiomers.

Ethiprole is an antagonist of ?-amino butyric acid (GABA – abbreviation of gamma-amino butyric acid) on the GABA receptor. It causes the receptor to be blocked and stops the exchange of chloride ions. This leads to over excitation of the central nervous system, which can lead to paralysis, respiratory arrest and death of the insect.

Acetamiprid ((E)-N1- [(6-chloro-3-pyridyl) methyl] -N2 – cyano - N1 –methyl acetamidine) is a carboxamidine that is an acetamidine in which a (6-chloropyridin-3-yl) methyl and a methyl group substitutes the amino hydrogen while cyano group replaces the hydrogen attached to the imino nitrogen.

Acetamiprid is an insecticide that belongs to the class of ‘neonicotinoids’. It is derived from 2-chloropyridine. The specific structure of acetamiprid is that of a chloronicotinyl compound and it is a potent agonist at the nicotinic acetylcholine receptors in insects. The major application of acetamiprid is to control insects such as Hemiptera, Thysanopterous, Lepidoptera and especially aphids.

Acetamiprid is a broad-spectrum and contact insecticide that is extensively used as replacement of other conventional insecticides for controlling sucking-type insects on various crops, especially leafy vegetables, fruits and tea bushes.

Acetamiprid is an agonist of the nicotinic acetylcholine receptor in various insects and affects the synapses in the insect central nervous system. It is highly soluble in water and is volatile. Acetamiprid has moderate toxicological, environmental and eco-toxicological properties.

Acetamiprid is useful to provide protection in crops like soybean, leafy vegetables, fruiting vegetables, citrus fruits, pome fruits, grapes, cotton and ornamental plants and flowers. Acetamiprid is usually formulated as soluble granules for spray application.

Acetamiprid was first disclosed in US 5304566 and chemically represented as below structure:

The molecular formula of Acetamiprid is C10H11ClN4 and molecular weight is 222.67 g/ mol and having water solubility of 2950 mg/L at 20oC. Acetamiprid is stable in buffered solutions at pH 4, 5 and 7.

Acetamiprid is the insecticide from IRAC (Insecticide Resistance Action Committee) Group 4A class. Acetamiprid is a systemic insecticide with trans laminar activity and with contact and stomach mode of action. The primary mechanism of acetamiprid toxicity against insects is due to its action at nicotinic cholinergic receptors. The unique nature of the neonicotinoids as insecticides is that the negatively charged cyano (or nitro) group will specifically interact with a cationic binding region that is unique to insects. This action will convey selectivity of action against insects and leave mammalian nicotinic receptors relatively unaffected.

In India or anywhere in the world, in agricultural production, using synthetic or chemical agent is the most effective means in order to control pests. However, the main disadvantage of such insecticide is that on long run, the insects develop resistance towards that insecticide and as a result to produce effective result, higher concentration of the insecticide is to be used. Here again using higher dose of insecticide may result in some detrimental effect to soil as well as environment. Therefore in general practice, usually combination of insecticide with another insecticide or another herbicide or fungicide is used in different concentration as per requirement.

However, when combination of one or more insecticide are used, the main challenge comes is to develop a formulation or composition which is stable, easy to use, safe and cost effective with superior efficacy. The present invention satisfies the existing needs, as well as others, and generally overcomes the deficiencies found in the prior art.

CN103651500A describes a composite insecticidal composition containing Ethiprole and Dinotefuron, characterized in that the composite insecticidal composition is an insecticidal active ingredient of Ethiprole and Dinotefuron, wherein Ethiprole and Dinotefuron the weight ratio is 0.1~80:0.5~80. This prior art does describe the combination of Ethiprole with neonicotinoids but nowhere discloses combination of Ethiprole and Acetamiprid in Wettable Granule formulation.

Indian Patent 201721029744 disclose a synergistic insecticidal composition comprising a) Fipronil; b) Neonicotinoids selected from Acetamiprid, Dinotefuran, Nitenpyram, Thiacloprid, Clothianidin, Thiamethoxam, Imidacloprid; and c) at least one more insecticide selected from Cartap Hydrochloride, Thiocyclam, Bifenthrin, Chlorpyrifos, Chlorantraniliprole and Flubendiamide and one or more inactive excipients. This prior art does describe the combination of Phenylpyrazole with neonicotinoids but nowhere discloses combination of Ethiprole and Acetamiprid in Wettable Granule formulation.

Indian patent 1510/DEL/2015 relates to An insecticidal synergistic composition comprising Diafenthiuron, Acetamiprid, wetting, dispersing, anti-caking agent, stabilizing antifoaming agent and inert carrier silica and lavigated clay to obtain said composition in a wettable powder form. This prior art does not comprise the combination of Ethiprole and Acetamiprid in Wettable Granule formulation.

CN101548684A relates to an insecticidal composition, comprising: the first active ingredient Ethiprole; the second active ingredient Imidacloprid; and pesticide-acceptable diluents or carriers, where the weight ratio of first active ingredient and the second active is 20: 1 to 1:20. This prior art does describe the combination of Ethiprole with neonicotinoids but nowhere discloses the combination of Ethiprole and Acetamiprid in Wettable Granule formulation.

However, there is no formulation or composition available of Ethiprole and Acetamiprid in Wettable Granule formulation which is stable and yet effective.

In the present invention, inventors have arrived to a synergistic insecticidal formulation of Ethiprole and Acetamiprid in Wettable Granule formulation and the process of preparation which is stable, effective and overcome all the problems mentioned in prior art as described herein.

OBJECT OF THE INVENTION

The principal object of the present invention is to provide a synergistic insecticidal formulation of Ethiprole and Acetamiprid.

Another object of the present invention is to provide a synergistic insecticidal formulation of Ethiprole and Acetamiprid, wherein the said composition is in Wettable Dispersible Granule (WG) formulation.

Yet another object of the present invention is to provide a synergistic insecticidal formulation of Ethiprole and Acetamiprid, wherein the said composition is in Wettable Dispersible Granule (WG) formulation, which is effective against a broad spectrum of pests and insects.

Another object of the present invention is to provide a process to manufacture a synergistic insecticidal formulation of Ethiprole and Acetamiprid, wherein the said composition is in Wettable Dispersible Granule (WG) formulation.

Yet another object of the present invention is to provide a synergistic insecticidal Wettable Granule formulation of Ethiprole and Acetamiprid which is stable.

Another object of the present invention is to provide a synergistic insecticidal Wettable Granule formulation of Ethiprole and Acetamiprid which is easy to manufacture.

Another object of the present invention is to provide a synergistic insecticidal Wettable Granule formulation of Ethiprole and Acetamiprid which is effective, farmer friendly and in use at field.

Yet another object of the present invention is to provide a synergistic insecticidal formulation of Ethiprole and Acetamiprid, wherein the said composition is in Wettable Granule formulation which provides enhanced insecticidal effect on insects which in turn results in better yield of crops.

SUMMARY OF THE INVENTION
The present invention discloses a synergistic insecticidal formulation of Ethiprole and Acetamiprid, wherein the said composition is in Wettable Dispersible Granule (WG) formulation comprising: Ethiprole Technical in the range of 35-50% w/w, Acetamiprid Technical in the range of 15-25% w/w, Dispersing agent in the range of 2-5% w/w, Surfactant in the range of 0.5-5% w/w, Suspending agent in the range of 2-5% w/w, Fillers in the range of 4-8% w/w and adjuvants. The present invention more particularly discloses a synergistic insecticidal formulation of Ethiprole and Acetamiprid and process of preparation thereof which is easy to manufacture, effective on broad spectrum of pests, lower the rate of application of the active ingredients, enhanced insecticidal effect on pests, farmer friendly, easily dispersible in water and cost effective.

DETAILED DESCRIPTION OF THE INVENTION

The detailed description set forth below is intended as a description of exemplary embodiments and is not intended to represent the only forms in which the exemplary embodiments may be constructed and/or utilized. The description sets forth the functions and the sequence of steps for constructing and/or operating the exemplary embodiments. However, it is to be understood that the same or equivalent functions and sequences which may be accomplished by different exemplary methods are also intended to be encompassed within the spirit and scope of the invention.

As defined herein, all scientific and technical terms used herein have the same meaning as commonly understood by one of ordinary skill in the art to which the invention belongs. Although any process and materials similar or equivalent to those described herein can be used in the practice or testing of the present invention, the preferred methods and materials are now described.

Unless specifically defined otherwise, all technical and scientific terms used herein shall be taken to have the same meaning as commonly understood by one of ordinary skill in the art. Unless the context requires otherwise, throughout the specification which follow, the word “comprise” and variations thereof, such as, “comprises” and “comprising” are to be construed in an open, inclusive sense that is as “including”, but not limited to.

As used in the description herein and throughout the claims that follow, the meaning of “a”, “an” and “the” includes plural reference unless the context clearly dictates otherwise. In some embodiments, the numerical parameters set forth in the written description are approximations that can vary depending upon the desired properties sought to be obtained by a particular embodiment.

As used herein, the terms "insecticide effect" and "insecticide activity" mean any direct or indirect action on the target insect that results in reduced feeding damage on the seeds, roots, shoots and foliage of plants as a result of foliar spray of said formulation.

As used herein, the term "pest" means any insect, rodent and microbes targeted for treatment with a formulation of the invention. Target pest means any insect, rodent and microbes that are harmful for the plants and decrease the productivity of plants.

The present invention is all about providing a synergistic insecticidal formulation of Ethiprole and Acetamiprid, wherein the said composition is in Wettable Dispersible Granule (WG) formulation and process of preparation thereof.

As per one embodiment, the present invention provides a synergistic insecticidal Wettable Dispersible Granule (WG) formulation comprising Ethiprole and Acetamiprid with the following mass percentage of the composition:

Active ingredient is the actual chemical in the product mixture that controls the pests, insects and microbes. Active ingredient plays a key role in the functioning of the product mixture.

As per one embodiment, the active ingredient for the present invention is Ethiprole and Acetamiprid, wherein the concentration range of Ethiprole is in the range of 35-50% w/w and Acetamiprid is in the range of 15-25% w/w in WG formulation.

A dispersing agent is a substance which adsorbs onto the surface of particles and helps to preserve the state of dispersion of the particles and prevents them from re-aggregating. Dispersing agents are added to agrochemical formulations to facilitate dispersion and suspension during manufacture, and to ensure the particles re-disperse into vehicle or carrier.

As per one embodiment, the concentration range of dispersing agent in the insecticidal composition of the present invention is in the range of 2-5% w/w. The dispersing agent in the present invention is selected from a group of Sodium salt of sulfonate naphthalene formaldehyde condensate, Sodium alkyl naphthalene sulfonate condensate, sodium salt of Alkylated naphthalene sulfonate or a combination thereof.

Wetting agents/ surfactants are chemical agents which lower the surface tension of liquid by a considerable amount in order to allow it to spread upon the penetrating surface. It is well within the competence of those skilled in the art to determine whether a particular surface active agent (surfactant) is a wetting agent or not.

As per one embodiment, the concentration range of surfactant in the insecticidal combination of the present invention is in the range of 0.5-5% w/w. The surfactant for present invention is selected from group consisting of polycarboxylate sodium salt, sodium lauryl sulphate, polydimethyl siloxane or combination thereof.

The Suspending agent helps active agent stay suspended in the formulation and prevent sedimentation at the bottom of the container. The suspending agent helps formulation to remain stable without problem of sedimentation.

As per one embodiment, suspending agent for the present invention is selected from group consisting of Silicon Dioxide, colloidal silicon dioxide or combination thereof.

An adjuvant is an additive which is added to make the formulations more effective, that improves or enhances application, performance, safety, storage, or handling of an active ingredient.

As per one embodiment, the insecticide formulation also contains one or more additional adjuvants selected from filler, polysaccharides, anti-foaming agent, anti-microbial agent, thickening agent, biocides and vehicle in the concentration range of 0.5-5% w/w.

As per one embodiment, filler in the present invention is selected from lactose monohydrate, hydrated aluminum silicate or a combination thereof in the concentration range of 4-8% w/w.

In yet another embodiment of the present invention, the benefits of the synergistic insecticidal formulation of Ethiprole and Acetamiprid and its process of preparation are described as follows:
1. Easy to manufacture.
2. Effective on wide range of insects and broadening the spectrum of insecticidal activity to other pests.
3. Reduction in the rate of application of the active ingredients.
4. Adequate control of the pests with the aid of the compositions according to the invention, even at a rate of application at which the individual compounds are totally ineffective.
5. Product is adapted to use for practice by farmers.
6. Cost effective
7. Novel combination of Ethiprole and Acetamiprid in WG formulation.
8. Better flowability due to WG formulation.
9. WG formulation of the combination of Ethiprole and Acetamiprid easily gets dispersed in water.

In yet another embodiment of the present invention there is provided a process for preparation of Ethiprole and Acetamiprid Wettable Dispersible Granule (WG) formulation.

The following examples are meant to illustrate the present invention. The examples are presented to exemplify the invention and are not to be considered as limiting the scope of invention.

Example 3: Field Trial Studies
Field trial experiments were conducted to evaluate the efficacy of Ethiprole and Acetamiprid WG formulation in comparison to its solo applications. The observations were recorded by counting the no. of pests before the treatment, 7 days after of treatment and after 15 days after treatment on the crops. The percent population reduction/control of pests was calculated by below formula:

The results of the above experiment show that the Ethiprole 41.05% + Acetamiprid 20.65% WG formulation show synergistic effect in terms of % Population reduction/ Control of BPH in rice. Maximum synergism is obtained at the dosage application rate of 150 g/ha with 76.09% population reduction/ control of BPH on rice.

Example 3(b): Efficacy of Ethiprole 41.05% + Acetamiprid 20.65% WG on Chilly whitefly (Bemesia Tabaci) population control.

The results of the above experiment show that the Ethiprole 41.05% + Acetamiprid 20.65% WG formulation show synergistic effect in terms of % Population reduction/ Control of whitefly in chilly. Maximum synergism is obtained at the dosage application rate of 150 g/ha with 66.67% population reduction / control of whitefly of Chilly crop.

Technical advancements:
The present disclosure described herein above has several technical advantages including, but not limited to, the realization of:
• A synergistic insecticidal Wettable Granule formulation comprising Ethiprole and Acetamiprid;
• A synergistic insecticidal Wettable Granule formulation comprising Ethiprole and Acetamiprid which is stable and is in a ready to use form;

From the results in Examples 1-3, the pesticidal combination of Ethiprole and Acetamiprid in WG formulation claimed in the present invention has a synergistic effect on control of pests.

1. A synergistic insecticidal formulation comprising:
a) Ethiprole Technical (Purity-98.00%) in the concentration range of 35-50% w/w,
b) Acetamiprid Technical (Purity-98.00%) in the concentration range of 15-25% w/w,
c) One or more Dispersing agent in the concentration range of 2-5% w/w,
d) One or more Surfactant in the concentration range of 0.5-5% w/w,
e) One or more Suspending agent in the concentration range of 2-5% w/w,
f) One or more Fillers in the concentration range of 4-8% w/w,
g) One or more Adjuvants in the concentration range of 0.5-5% w/w.

2. The synergistic insecticidal formulation as claimed in claim 1, wherein the formulation is Wettable Dispersible Granule (WDG) formulation.

3. The synergistic insecticidal formulation as claimed in claim 1, wherein Ethiprole Technical (Purity-98.00%) is in the concentration range of 35-50% w/w.

4. The synergistic insecticidal formulation as claimed in claim 1, wherein Acetamiprid Technical (Purity-98.00%) in the concentration range of 15-25% w/w.

The synergistic insecticidal formulation as claimed in claim 1, wherein the dispersing agent is selected from a group of Sodium salt of sulfonate naphthalene formaldehyde condensate, Sodium alkyl naphthalene sulfonate condensate, sodium salt of Alkylated naphthalene sulfonate or a combination thereof in the concentration range of 2 - 5% w/w.

6. The synergistic insecticidal formulation as claimed in claim 1, wherein the surfactant is selected from group consisting of polycarboxylate sodium salt, sodium lauryl sulphate, polydimethyl siloxane or combination thereof in the concentration range of 0.5-5% w/w.

7. The synergistic pesticide composition as claimed in claim 1, wherein the suspending agent is selected from group consisting of Silicon Dioxide, colloidal silicon dioxide, Polyalkoxylated butyl ether or combination thereof in the concentration of 2-5% w/w.

8. The synergistic pesticide composition as claimed in claim 1, wherein the filler is selected from lactose monohydrate, hydrated aluminum silicate or combination thereof in the concentration range of 4-8% w/w.

9. A pesticidal formulation of Chlorantraniliprole and Lambda-cyhalothrin comprising:
a) 41.05% w/w of Ethiprole Technical,
b) 20.65% w/w of Acetamiprid Technical,
c) 2-5% w/w of combination of Sodium salt of sulfonate naphthalene formaldehyde condensate, Sodium alkyl naphthalene sulfonate condensate and sodium salt of alkylated naphthalene sulfonate,
d) 0.5-5% w/w of combination of Polycarboxylate sodium salt and sodium lauryl sulphate,
e) 0.5% w/w of Polydimethyl siloxane,
f) 2% w/w of Silicone dioxide,
g) 5% w/w of Lactose monohydrate,
h) 7.8% w/w of Hydrated Aluminum silicate.
