TITLE: VACCINE COMPRISING ADJUVANT EMBEDDED IN LIPID BI-LAYER SPHERICAL PARTICLE ENCLOSING AQUEOUS COMPARTMENT CARRYING CONJUGATE
OBJECTIVE:
To provide a vaccine formulation comprising lipid bi-layer spherical particle enclosing aqueous compartment carrying protein-polysaccharide conjugate wherein the particle is embedded with at least one adjuvant
BRIEF DESCRIPTION:
The first embodiment of the invention is a vaccine preparation which comprises lipid bi-layer spherical particle enclosing aqueous compartment carrying antigen -protein conjugate and essentially an adjuvant embedded to the lipid layer.
The second embodiment of the invention is embedding varying amount of adjuvant(s) based on the antigen type.
The third embodiment of the invention is use of antigen-protein conjugate in combination with at least one adjuvant to enhance the immunological effect of antigens conjugates.
The antigens of the instant invention are, but not limited to, bacterial antigen. The antigens may be selected from cellular antigens of various types comprising whole cell, carbohydrates, proteins, lipoproteins, lipopolysaccharides, polysaccharides, nucleic acids, carbohydrates. The antigens may be obtained from microorganism of genus selected from Streptococcus, Haemophilus, Neisseria and Escherichia.
The carrier proteins of the instant inventions are, but not limited to, diphtheria toxoid, Tetanus toxoid, CRM197 or their derivatives e.g. mutants.
The adjuvants of the instant inventions are, but not limited to, monophosphoryl lipid A or derivatives thereof; aluminium compounds preferably aluminium hydroxide, aluminium phosphate, potassium aluminium sulphate; calcium compounds preferably calcium phosphate; outer membrane vesicles or proteins obtained from serotype strains or commensal strains; M. phlei cell wall skeleton; synthetic acylated monosaccharides; DC_chol (lipoidal immunostimulators; CpG motifs; genetically modified toxins; cytokins etc.
ADVANTAGES OF THE INVENTION:
Better vaccine for prevention of various diseases.
