Abstract: The present invention relates to a stable nutraceutical composition for enhancing the workout performance in a human subject comprising an effective amount of plurality of energy- yielding ingredients selected from one or more of amino acids, vitamins, caffeine, ginseng panax, phytonutrients or mixtures thereof and optionally other additives. The performance may be, for example, athletic, academic, or other performances requiring physical stamina and/or mental alertness. The nutraceutical composition is useful for conditions characterized by altered mitochondrial functioning and biogenesis, such as increasing physical performance, reducing fatigue, increasing energy and focus, building heart strength, various liver diseases, improve muscle/fat ratio, muscle endurance thus leading to improved VO2 max production.

BACKGROUND OF THE INVENTION
Muscle activity is primarily based on a very fundamental biochemical mechanism, the breakdown of energy-rich phosphate bonds (ATP, adenosine triphosphate). ATP is the direct source of energy for muscle work and is in fact the only form of chemical energy, which can be converted by the muscle into mechanical work. During high physical activity of the body, the ATP level in the muscles diminishes rapidly. Several substrates are available as sources for replenishing the ATP. One strategy for minimizing fatigue and muscle weakness is to enhance a diet with nutritional supplements that help meet/exceed additional nutritional demands.
There are many types of nutritional compositions currently on the market. The dietary supplements are appealing as a quick means for attaining adequate nutrition, boosting the immune and metabolic systems, enhancing exercise and sport and work performance, and assisting with weight loss. These supplements include performance enhancing supplements (steroids like blood doping, human growth hormone; non-steroids like creatine), energy supplements (caffeine), and supplements that aid in recovery (protein, branched chain amino acids or BCAAs). Commonly known energy supplements such as caffeine, Guarana, Vitamin B12, and Asian ginseng are known to increase the efficiency of an individual to have extended performance.
US Publication No. 2013/0122148 describes puree nutritional compositions comprising at least one fruit puree, at least one vegetable puree and carbohydrates fraction comprising glucose and fructose in a weight ratio from about 0.5:1 to about 5:1 for maximizing performance.
US Publication No. 2016/0213673 provides compositions containing dihydrocapsiate, caffeine and arginine to increase human endurance during times of physical activity. The compositions enhance endurance by increasing the sparing of glycogen stores, forcing aerobic activity, increasing vasodilation, and reducing the build-up of metabolites that can lead to the feeling of exhaustion during and after physical activity.
The benefits of available workout supplements include: improvement in time to failure and work capacity by giving intense amounts of energy (ATP); enhancement of physical performance, enhancement of blood flow and helping muscles to have a speedy recovery and the time to failure during exercise; boosting adrenaline which helps breakdown fat cells faster; promoting lipid oxidation using energy produced by burning fat rather than carbohydrates; protecting muscle loss under fasted training and improving focus, mood, and attention.
Focusing on the above parameters of energy, the present inventors have prepared an alternative workout supplement that can be taken pre or post workout depending on need and comprises of thermic energy yielding ingredients that enhances the body’s own capacity for energy generation and/or enhances cellular energy generation.
SUMMARY OF THE INVENTION
The present invention provides a unique combination of ingredients that are designed to provide enhanced endurance by multiple ways. This advanced workout supplement, trade marked as “MB ENERGY PROTM” exhibits increased mitochondrial ATP generation, enhanced energy flow and helps to improve the quality of exercise.
In one of the aspects of this invention, there is provided a nutraceutical composition for enhancing the workout performance in a human subject comprising an effective amount of plurality of energy-yielding ingredients selected from one or more of amino acids, vitamins, caffeine, ginseng panax, phytonutrients or mixtures thereof and optionally other additives.
In an embodiment of the above aspect, the phytonutrient comprises one or more of phenolic antioxidants, phenolic acids, stilbenoids, flavonoids, flavones, isoflavones, alkaloids or mixtures thereof.
In a further embodiment, the phenolic antioxidants are selected from one or more of catechins, rutin, epigallocatechin gallate, resveratrol, quercetin and tyrosol esters such as hydroxytyrosol, oleuropein, oleocanthal.
In another embodiment, the phytonutrient is a stilbenoid selected from pterostilbene, piceatannol, pinosylvin and resveratrol.
In the second aspect of the invention, there is provided a nutraceutical composition for enhancing the workout performance in a human subject comprising an effective amount of one or more amino acids, vitamins, caffeine, ginseng panax, pterostilbene, hydroxytyrosol, catechins, rutin and optionally other additives selected from flavours, colors and sweeteners, wherein the composition is in powder form.
In an embodiment of above aspect, the amino acids comprises of arginine and beta-alanine.
In another embodiment, the amino acids comprise arginine and citrulline malate.
In another embodiment, the amino acids comprise beta-alanine and citrulline malate.
In another embodiment, the composition comprises at least one vitamin selected from vitamin B, vitamin K or mixtures thereof.
In a particular embodiment, the composition comprises vitamin B12.
In another particular embodiment, the composition comprises vitamin K2.
In another embodiment, the composition comprises a mixture of vitamin B12 and vitamin K2.
In the third aspect of the invention, there is provided a stable nutraceutical powder composition for enhancing the pre/post workout performance in a human subject comprising effective amount of mixtures of arginine, citrulline and beta-alanine as amino acids; vitamin B12, caffeine, ginseng panax, pterostilbene, juniperus communis extract and optionally other additives selected from flavors, colors and sweeteners.
In fourth aspect of the invention, there is provided a nutraceutical composition for enhancing the workout performance in a human subject comprising:
about 1 to 10 grams of amino acid,
about 0.01 to 2grams of vitamin,
about 0.05 to 0.5grams of caffeine,
about 0.01 to 5grams of phytonutrients,
about 0.01 to 1grams of ginseng panax, and
optional additives selected from flavors, colors and sweeteners.
In an embodiment of the above aspect, the composition comprises mixtures of beta- alanine and citrulline as amino acids; mixtures of vitamin K2 and vitamin B12, hydroxytyrosol, caffeine and optionally other additives selected from flavors, colors and sweeteners.
In a preferred embodiment, the sweetener is sucralose.
In another embodiment, the composition further comprises about 1 to 5 g of beetroot powder.
In another embodiment, the composition comprises a mixture of beta-alanine and citrulline as amino acids, vitamin B12, hydroxytyrosol, caffeine, sucralose, flavors and colors.
In a particular embodiment, the composition comprises citrulline; vitamin K2, hydroxytyrosol, caffeine, pterostilbene, ginseng panax, beetroot powder, sucralose, flavors and colors.
In another particular embodiment, the composition comprises beta-alanine as amino acid, vitamin K2, hydroxytyrosol, caffeine, pterostilbene, beetroot powder, sucralose, flavors and colors.
In another particular embodiment, the composition comprises a mixture of beta-alanine and arginine as amino acids, vitamin K2, hydroxytyrosol, caffeine, pterostilbene, beetroot powder, sucralose, flavors and colors.
In another embodiment, the composition comprises citrulline, vitamin K2, hydroxytyrosol, pterostilbene, caffeine and ginseng panax.
In another embodiment, the composition is in the form of a drinkable product.
In an embodiment of the present invention, there is provided a process of preparing a stable nutraceutical composition comprising the steps of:
(a) sifting the required quantities of ingredients through an appropriate sieve;
(b) adding the sifted ingredients into a suitable blender;
(c) mixing the ingredients at 15 rpm for about 15-20 minutes or until a uniform blend is obtained; and
(d) optionally compressing the final blend into suitable size tablets/Bar.
In fifth aspect of the invention, there is provided a method of improving the workout performance comprising administering a therapeutically effective amount of stable nutraceutical composition of the present invention to a human subject in need thereof.
In an embodiment of above aspect, the composition is administered 15 to 45 minutes prior to physical exercise or 10-30 minutes post exercise.
In a preferred embodiment, the composition is administered 30 minutes prior to physical exercise and 10 minutes post exercise.
In sixth aspect of the invention, there is provided a method of maintaining or increasing mitochondrial function and activity via increased mitochondrial biogenesis comprises administering an effective amount of nutraceutical composition of the present invention to the human subject in need thereof.
In seventh aspect of the invention, there is provided a nutraceutical composition for enhancing the workout performance in a human subject, wherein the nutraceutical composition comprises an effective amount of plurality of energy-yielding ingredients: vitamin, caffeine, ginseng panax and phytonutrients and optionally other additives.
In an embodiment of the above aspect, the vitamin is selected from vitamin K2 and vitamin B12 and combination thereof.
In an embodiment of the above aspect, the phytonutrient comprises one or more of phenolic antioxidants, phenolic acids, stilbenoids, flavones, isoflavones, alkaloids or mixtures thereof.
In a further embodiment, the phenolic antioxidants are selected from one or more of catechins, rutin, epigallocatechin gallate, resveratrol, quercetin and tyrosol esters such as hydroxytyrosol, oleuropein, oleocanthal.
In another embodiment, the phytonutrient is a stilbenoid selected from pterostilbene, piceatannol, pinosylvin and resveratrol and combination thereof.
In another embodiment, the catechin is juniperus cummunis.
In yet another embodiment of the above aspect, the nutraceutical composition further comprises amino acid. The amino acid is selected from arginine, citrulline and beta-alanine and combinations thereof.
In yet another embodiment of the above aspect, the composition comprises:
about 0.01 to 2 grams of vitamin,
about 0.05 to 0.5 grams of caffeine,
about 0.01 to 5 grams of phytonutrients,
about 0.01 to 1 grams of ginseng panax, and
optional additives selected from flavors, colors and sweeteners.
In yet another embodiment, the nutraceutical composition is in the form of a drinkable product.
The nutraceutical compositions are stable over a period of time suitable for use or during storage.
DETAILED DESCRIPTION OF THE INVENTION
While the present invention may be embodied in many different forms, disclosed herein are specific illustrative embodiments thereof that exemplify the principles of the invention. It should be emphasized that the present invention is not limited to the specific embodiments illustrated. As used herein, the following definitions apply unless clearly indicated otherwise.
The term "stable" as used herein refers to formulations capable of maintaining their physical and chemical stability for a minimum of 3 months, more preferably at least 6 months, even more preferably 9 months and most preferably 12 months or more when stored under ambient conditions.
The term "nutraceutical" as used herein denotes the usefulness in both the nutritional and pharmaceutical field of application.
The term “workout” as used herein, represents the preparation of human body prior to intense exercise or workout and also to retrieve body function post exercise.
The term “effective amount” or “therapeutically effective amount” refers to that amount having therapeutic effects on the condition intended to be treated or prevented.
The term “phytochemicals” and “phytonutrients” as used herein refers to any chemical produced by a plant that imparts one or more health benefit on the user.
The term "mitochondrial biogenesis" as used herein, includes all processes involved in maintenance and growth of the mitochondria, including those required for mitochondrial division and segregation during the cell cycle.
Examples of vitamins include but are not limited to one or more of vitamin A, vitamin Bl, vitamin B2, vitamin B3, vitamin B5, vitamin B6, vitamin B7, vitamin B8, vitamin B9, vitamin B12, vitamin C, vitamin D, vitamin E, vitamin K such as phylloquinone (vitamin K1), menaquinone (vitamin K2), vitamin K3, vitamin K4, vitamin K5, vitamin K6 and vitamin K7 or any precursor or analogue to any of these vitamins.
Vitamin K2 in particular has been demonstrated to restore mitochondrial function and has a key role in production of mitochondrial adenosine triphosphate. Thus, the dietary supplementation with vitamin K2 would increase the function of muscle with high mitochondrial content.
Caffeine is a xanthine alkaloid that acts as a stimulant drug. Caffeine affects the CNS by stimulating the secretion of serotonin in the cerebral cortex, enhancing the action of the sympathetic system, and diminishing the activity of inhibitory neurons as a result of the binding and blocking of adenosine receptors. Adenosine and caffeine work opposite of one another with respect to cellular regulations. Adenosine, a neuromodulator, binds to adenosine receptors and slows nerve cell activity, whereas caffeine blocks adenosine receptors and speeds up the activity of cells. This mechanism greatly affects the user’s cognition and mood. Caffeine significantly enhances concentration, visual vigilance, choice reaction time, and self-reported fatigue (i.e., after a period of exercise, work, and a long period of sleep deprivation) in a dose-dependent manner.
Ginseng is material extracted from the root of a plant that is a member of the ginseng family (Araliaceae), genus Panax. According to Traditional Chinese Medicine, Panax Ginseng promotes Yang energy, improves circulation, increases blood supply, revitalizes and aids recovery from weakness after illness, and stimulates the body. The types and relative abundance of the chemical constituents of ginseng depend on the species, the part of the plant, the place of origin, the method of cultivation, and the technique of extraction used to obtain the ginseng. Different types of ginseng may have slightly different constituent profiles, but the type of ginseng used in this composition will be the material extracted from Panax ginseng or Panax quinquefolius. Panax ginseng contains many active substances. The substances thought to be most important are called ginsenosides or panaxosides.
Examples of phytonutrients or phytochemicals include but are not limited to one or more of phenolic antioxidants, phenolic acids, stilbenoids, flavonoids, flavones such as catechins, isoflavones, alkaloids or mixtures thereof.
Hydroxytyrosol is chemically 3,4-dihydroxyphenylethanol and encompasses any material or extract of a plant or any material or extract of parts of a plant or any extract/concentrate/juice of fruits of a plant (such as olives) containing it. Furthermore, it encompasses physiologically/pharmaceutically acceptable salts thereof such as sodium or potassium salts. Hydroxytyrosol induces mitochondrial biogenesis and leads to an increased mitochondrial function in tissues. An increased mitochondrial biogenesis results in a number of desirable physical states such as increase in cardiac health, energy, strength and endurance.
Pterostilbene (trans-3,5-dimethoxy-4'-hydroxystilbene) is a naturally occurring stilbenoid compound that is a structural analog of resveratrol. It is a well-known antioxidant that is more bioavailable than resveratrol; reduces muscle damage, immune dysfunction and fatigue, and thus improve performance.
Catechin is a flavan-3-ol, a type of natural phenol and antioxidant commonly found in berries, tea, cocoa etc. Catechin shows potential for fat oxidation thus prolonged exercise performance can be improved by an increased utilization of fat as energy during exercise, since fat utilization reduces anaerobic carbohydrate consumption in providing energy substrate to the muscle is reduce and minimizes production of lactate in the muscle. A good source of catechin is juniperus cumminus extract.
Examples of amino acids include but are not limited to one or more of arginine, asparagine, aspartate, citrulline, cysteine, glutamate, carnosine, carnitine, beta-alanine, glutamine, glycine, histidine, hydroxyproline, hydroxyserine, hydroxytyrosine, hydroxylysine, isoleucine, leucine, lysine, methionine, phenylalanine, proline, serine, taurine, threonine, tryptophan, tyrosine, valine, or mixtures thereof.
Arginine is characterized as a nonessential amino acid and plays a significant role in healing, cell division, immune function, the elimination of ammonia from the body and the release of hormones. Arginine is used in the dietary supplements to supplement to boost Human Growth Hormone (HGH) production, increase vasodilation, enhance blood circulation, increase oxygen flow to the muscles, and boost Nitric Oxide (NO) production.
Citrulline is an alpha-amino acid naturally occurring in the human body and is made from the amino acid ornithine, along with carbamoyl phosphate in one of the central reactions in the Urea Cycle. It is also produced from arginine together with nitrogen oxide (NO) known as a substance with a vasodilating activity and is condensed with aspartic acid to regenerate arginine. Citrulline is commercially available as supplements for improvement of blood flow, prevention of arteriosclerosis and energy enhancement in the U.S. and citrulline-malate is commercially available as a medicament for recovery from fatigue in Europe. Ingestion of citrulline malate resulted in a significant reduction in the sensation of fatigue, a 34% increase in the rate of oxidative ATP production during exercise, and a 20% increase in the rate of phosphocreatine recovery after exercise, indicating a larger contribution of oxidative ATP synthesis to energy production. Considering individual variables characterization aerobic function, extrema were measured after either eight or 15 days of treatment, indicating chronological heterogeneity of treatment induced changes. One way analysis of variance confirmed improved aerobic function, which may be the result of an enhanced malate supply activating ATP production from the tricarboxylic acid cycle through anaplerotic reactions.
The nutraceutical compositions may comprise additional additives such as sweetening agents, flavoring agents, coloring agents, preservatives, pH modifiers, and buffering agents.
Examples of sweetening agents that may be optionally included in the composition of the invention include but are not limited to sucrose, lactose, aspartame, acesulfame-K, sucralose, monatin, stevia, saccharin and the like.
Examples of flavoring agents include but are not limited to fruit flavors, botanical flavors, and the like. Non-limiting examples of fruit flavors include orange, lemon, lime, grapefruit, apple, grape, cherry, pineapple, fruit punch, strawberry, kiwi, tangerine, raspberry, mango, cocoa, papaya, apricot, passion fruit, and cupuacu.
Examples of coloring agents include but are not limited to fruit, vegetable, and/or plant extracts from sources such as grape, black currant, aronia, carrot, beetroot, red cabbage, and hibiscus.
Examples of preservatives include but are not limited to acetic acid, citric acid, benzoic acidsorbic acid, sulphur dioxide, potassium benzoate, potassium sorbate, sodium benzoate, sodium sorbate, calcium benzoate, calcium sorbate, sodium citrate, potassium citrate, sodium hexametaphosphate, nitrites, sulfites etc.. Preservatives may also include antioxidants such as ascorbic acid, EDTA, BHA, BHT, TBHQ, dehydroacetic acid, dimethydicarbonate, ethoxyquin, and parabens.
Examples of pH modifiers include but are not limited to lactic acid, citric acid, malic acid, fumaric acid, adipic acid, phosphoric acid, gluconic acid, tartaric acid, ascorbic acid, acetic acid, succinic acid, and malic acid; sodium hydroxide, potassium hydroxide, calcium hydroxide and magnesium hydroxide.
Examples of buffering agents include but are not limited to carbonic acid, citric acid, acetic acid, phosphoric acid, succinic acid and various salts of such acids.
The nutraceutical compositions according to the present invention is suitable for administrating to the human body in any form that is conventional for oral administration, e.g. in solid form such as (additives/supplements for) food or feed, food or feed premix, tablets, pills, granules, capsules, and effervescent formulations such as powders and tablets, or in liquid form such as solutions, emulsions or suspensions as e.g. beverages, pastes and oily suspensions. In a preferred embodiment, the composition is in the form of a drinkable product.
The nutraceutical composition according to the present invention is administered 15 to 45 minutes prior to physical exercise or 10-30 minutes post exercise. In a preferred embodiment, the composition is administered 30 minutes prior to physical exercise and 10 minutes post exercise.
Various liquid and powder compositions according to the present disclosure may be prepared by conventional methods.
While the methods have been described in terms of what are presently considered to be the most practical and preferred embodiments, it is to be understood that the disclosure need not be limited to the disclosed embodiments. It is intended to cover various modifications included within the spirit and scope of the claims, the scope of which should be accorded the broadest interpretation so as to encompass all such modifications and similar structures.

CLAIMS:1. A nutraceutical composition for enhancing the workout performance in a human subject, wherein the nutraceutical composition comprises an effective amount of plurality of energy-yielding ingredients: vitamin, caffeine, ginseng panax and phytonutrients and optionally other additives.
2. The nutraceutical composition of claim 1, wherein the vitamin is selected from vitamin K2 and vitamin B12 and combination thereof.
3. The nutraceutical composition of claim 1, wherein the phytonutrient comprises one or more of phenolic antioxidants, phenolic acids, stilbenoids, flavones, isoflavones, alkaloids or mixtures thereof.
4. The nutraceutical composition of claim 3, wherein the phenolic antioxidants are selected from one or more of catechins, rutin, epigallocatechin gallate, resveratrol, quercetin and tyrosol esters such as hydroxytyrosol, oleuropein, oleocanthal.
5. The nutraceutical composition of claim 3, wherein the stilbenoid is selected from pterostilbene, piceatannol, pinosylvin and resveratrol and combination thereof.
6. The nutraceutical composition of claim 4, wherein the catechin is juniperus cummunis.
7. The nutraceutical composition of claim 1, wherein the nutraceutical composition further comprises amino acid.
8. The nutraceutical composition of claim 7, wherein the amino acid is selected from arginine, citrulline and beta-alanine and combinations thereof.
9. The nutraceutical composition of claim 1, wherein the composition comprises:
about 0.01 to 2 grams of vitamin,
about 0.05 to 0.5 grams of caffeine,
about 0.01 to 5 grams of phytonutrients,
about 0.01 to 1 grams of ginseng panax, and
optional additives selected from flavors, colors and sweeteners.
10. The nutraceutical composition of claim 1, wherein the composition is in the form of a drinkable product.
