Method For Preparation Of Pneumococcal Conjugate Vaccine With Low Cdap

Abstract: The present invention relates to a method for covalent coupling of polysaccharides to proteins for the preparation of Pneumococcal Conjugate Vaccines (monovalent) using low quantities/ratios of 1-Cyano 4-Dimethyl Amino Pyridinium tetrafluoroborate (CDAP) thereby preventing the destruction of immunogenic epitopes and prevention of self-aggregation of pneumococcal polysaccharides during CDAP activation. The method results in stable monovalent conjugates and also prevents the formation of self- aggregated conjugate molecules during conjugation reaction.
