Riboflavin Derivative Containing Medium

Abstract: The present invention addresses the problem of providing a medium for growing and culturing pluripotent stem cells such as iPS cells and ES cells, in particular, a cell growth medium which is free from deterioration caused by the preservation of a complete medium, and a method for producing said medium. Provided are a medium for growing stem cells, said medium containing a riboflavin derivative, etc.

Claims

1. A medium for stem cell proliferation comprising a riboflavin derivative. 5 2 . The medium according to claim 1 wherein the riboflavin derivative is at least one kind selected from the group consisting of flavin adenine dinucleotide, flavin mononucleotide, riboflavin tetrabutyrate, salts thereof and 10 hydrates thereof.

3. The medium according to claim 1 or 2 wherein the riboflavin derivative is flavin adenine dinucleotide or a salt thereof, or a hydrate thereof. 15

4. The medium according to any one of claims 1 to 3 wherein the stem cell is an ES cell or an iPS cell.

5. The medium according to any one of claims 1 to 4 wherein the 20 medium does not comprise riboflavin.

6. A medium for stem cell proliferation comprising a mixture of a basal medium comprising an amino acid, a vitamin, a mineral and a buffering agent and one or more supplements, wherein the 25 basal medium comprises a riboflavin derivative.

7. A stabilizer of a medium for stem cell proliferation comprising a riboflavin derivative. 30 8. A method for stabilizing a medium for stem cell proliferation comprising adding a riboflavin derivative.

9. A method for producing a medium for stem cell proliferation comprising adding a riboflavin derivative.

10. A method for producing a medium for stem cell proliferation comprising mixing a riboflavin derivative, a basal medium comprising an amino acid, a vitamin, a mineral and a buffering agent, and one or more supplements. 5

11. A method for culturing a stem cell comprising culturing in the medium according to any one of claims 1 to 6.

12. A method for producing a stem cell comprising culturing in 10 the medium according to any one of claims 1 to 6.

WE CLAIM:
1. A medium for stem cell proliferation comprising a riboflavin derivative.

2 . The medium according to claim 1 wherein the riboflavin derivative is at least one kind selected from the group consisting of flavin adenine dinucleotide, flavin mononucleotide, riboflavin tetrabutyrate, salts thereof and 10 hydrates thereof.
3. The medium according to claim 1 or 2 wherein the riboflavin
derivative is flavin adenine dinucleotide or a salt thereof, or
a hydrate thereof.

4. The medium according to any one of claims 1 to 3 wherein the
stem cell is an ES cell or an iPS cell.
5. The medium according to any one of claims 1 to 4 wherein the
20 medium does not comprise riboflavin.
6. A medium for stem cell proliferation comprising a mixture of
a basal medium comprising an amino acid, a vitamin, a mineral
and a buffering agent and one or more supplements, wherein the
25 basal medium comprises a riboflavin derivative.
7. A stabilizer of a medium for stem cell proliferation
comprising a riboflavin derivative.
30 8. A method for stabilizing a medium for stem cell
proliferation comprising adding a riboflavin derivative.
9. A method for producing a medium for stem cell proliferation comprising adding a riboflavin derivative.

10. A method for producing a medium for stem cell proliferation
comprising mixing a riboflavin derivative, a basal medium
comprising an amino acid, a vitamin, a mineral and a buffering
agent, and one or more supplements.

11. A method for culturing a stem cell comprising culturing in
the medium according to any one of claims 1 to 6.
12. A method for producing a stem cell comprising culturing in
10 the medium according to any one of claims 1 to 6.
