Abstract: The present disclosure provides a herbal composition comprising: a) Zingiber officinale extract; and b) at least one organic compound, wherein the Zingiber officinale extract to the at least one organic compound weight ratio is in a range of 0.2:1 to 5:1. The present disclosure further provides a convenient process for preparing the composition.
TECHNICAL FIELD
[001] The subject matter described herein in general relates to the pharmaceutical and food supplement compositions, and in particular, relates to the field of herbal compositions.
BACKGROUND OF INVENTION
[002] Iron is an important mineral for human health. Iron helps in the physical and mental growth of humans and the requirement increases with age. Recent survey revealed that iron deficiency is most prevalent in India (Rekha Kumari et.al, J Clin Diagn Res. 2017). Some of the dietary factors are known to impair the nonheme iron absorption (Richard Hurrell et. al., Am J Clin Nutr 2010). Therefore, the iron bioavailability is found to be low in our diet. To overcome this problem, first approach is that people can take iron supplements. However, daily intake of iron supplementation has some known side effects such as gastrointestinal problems (Zoe Tolkien et.al, PLoS One. 2015). US7585527 discloses the usage of iron polysaccharides as an effective iron supplement. EP2661273 discloses an iron chelate as a food supplement.
[003] The second approach involves taking some adjuvants which have the ability to improve the non-heme iron absorption from regular dietary intake. The usage of herbal extracts in this approach may be particularly advantageous as it is known to have low cellular toxicity. WO2006068729 discloses a combination of two organic acids for enhancing iron-uptake. SUMMARY OF THE INVENTION
[004] In an aspect of the present disclosure, there is provides a herbal composition comprising: a) Zingiber officinale extract; and b) at least one organic compound, wherein the Zingiber officinale extract to the at least one organic compound weight ratio is in a range of 0.2:1 to 5:1.
[005] In another aspect of the present disclosure, there is provided a process for preparing the herbal composition comprising: a) Zingiber officinale extract; and b) at least one organic compound, wherein the Zingiber officinale extract to the at least one organic compound weight ratio is in a range of 0.2:1 to 5:1, said process
comprising steps of: a) obtaining Zingiber officinale extract; b) at least one organic
compound; and c) contacting the Zingiber officinale extract with at least one
organic compound to obtain the composition.

absorption on Caco2 cells in the presence of FeS04 (lOuM), in accordance with an
implementation of the present subject matter.

[0021] As mentioned previously, the existing food supplements suffer from drawbacks of toxicity and high cost. Therefore, there is a scope for exploration of synergistic actives combinations which can provide the better efficacy of health promoting benefits such as enhanced iron uptake. Herbal or natural combinations could prove effective in providing enhanced iron-uptake while being non-toxic. The present disclosure aims to solve the said problem by employing a combination consisting of ginger extract and an organic compound shows the synergistic effect on the iron absorption.
[0022] In an embodiment of the present disclosure, there is provided a herbal composition comprising: a) Zingiber officinale extract; and b) at least one organic compound, wherein the Zingiber officinale extract to the at least one organic compound weight ratio is in a range of 0.2:1 to 5:1. In another embodiment of the present disclosure, the Zingiber officinale extract to the at least one organic compound weight ratio is in arange of 0.3:1 to 4.8:1.
[0023] In an embodiment of the present disclosure, there is provided a herbal composition comprising: a) Zingiber officinale extract; and b) at least one organic compound, wherein the Zingiber officinale extract to the at least one organic compound weight ratio is in a range of 0.5:1 to 4.5:1. Succinic acid, sodium acetate and sodium ascorbic acid -Vitamin C were obtained commercially from Sigma. Example 1: Identification of active for iron absorption
[0086] Iron uptake assay was studied on Caco2 cells (P47-P52). Caco2 cells were seeded on a 24 well plate at density 5X104 cells/ well. Post confluency of 14 days, cells were used for iron uptake assay. Caco2 cells were washed with iron free HBSS (Hanks Balanced Salt Solutions from thermo fisher scientific) then cells were treated with the actives and its combinations in the presence of ferrous sulphate (lOuM) for 60 min at 5% C02, 37°C. After incubation, media was removed and
cells were washed twice with ice cold HBSS. Samples were lysed with 50mm NaOH and followed by sonication at 25%amp for 5s. Then some quantity (50ul)
used for protein estimation by BCA method and remaining used for total
intracellular iron content by using ferrozine method.
[0087] Estimation of total iron by ferrozine method: Aliquots of tested lysate
samples (lOOul) were mixed with equal quantity of lOmM of HC1 and freshly
prepared iron releasing reagent (1.4MHC1 and 4.5%KMn04 in 1:1 ratio) and
incubated for 2h at 60°C. Post incubation, samples were cooled to room
temperature and mixed with 30ul of iron detection reagent (freshly mixed 6.5mM
ferrozine, 2.5m ammonium acetate and 1M of Ascorbic acid) and incubated for
30min, then absorbance was measured at 550nm on a microplate reader. Percentage
of iron absorption was calculated for each tested samples.
[0088] Among the all combinations, Zingiber officinale or ginger extract
(0.005%) and succinic acid (0.0025%) combination (Figures 1 and 2) showed the
high iron absorption ability (-31%) as compared to control (FeS04). The
combination pertaining to Zingiber officinale or ginger extract (0.005%) and
sodium acetate (0.0025%) was also found to show synergistic enhancement
(19.53% in Figure 1).
Example 2: Evaluation of selected combination at different ratios for iron
absorption ability
[0089] The process as described in Example 1 was followed in order to test
various ratios of a combination comprising: a) Zingiber officinale or ginger extract;
and b) at least one organic compound for iron uptake.
[0090] Among the all ratios tested for iron absorption (Figure 3), Zingiber
officinale or ginger extract and succinic acid combination at 1.5:1 ratio showed
synergistic iron absorption rate (-52%). The other combinations pertaining to 2:1,
4:1, 2:3 or 0.67:1 and 1:4 or 0.25:1 were also found to enhance iron uptake. Thus,
the combination (ginger extract and succinic acid) proved to have synergistic iron
absorption ability as compared to individual.
Advantages of the present disclosure:
[0091] The present disclosure reveals a herbal composition comprising Zingiber
officinale or ginger extract; and b) at least one organic compound. The composition
was found to enhance cellular iron uptake. The composition can be easily adapted to obtain useful commercial formulations such as health supplements.
I/We Claim;
1. A herbal composition comprising:
a) Zingiber officinale extract; and
b) at least one organic compound,
wherein the Zingiber officinale extract to the at least one organic compound weight ratio is in a range of 0.2:1 to 5:1.
2. The herbal composition as claimed in claim 1, wherein the Zingiber officinale extract to the at least one organic compound weight ratio is in a range of 0.5:1 to 1:4.5.
3. The herbal composition as claimed in claim 1, wherein the Zingiber officinale extract to the at least one organic compound weight ratio is in a range of 1.5:1 to 2.5:1.
4. The herbal composition as claimed in claim 1, wherein the Zingiber officinale extract comprises gingerol having a weight percentage in a range of 6 - 8% with respect to the extract.
5. The herbal composition as claimed in claim 1, wherein the at least one organic compound is selected from a group consisting of succinic acid, sodium acetate, citric acid, tartaric acid, malic acid, propionic acid, fumaric acid, and combinations thereof.
6. The herbal composition as claimed in any one of the claims 1-5, wherein the composition comprises:
a) Zingiber officinale extract having weight percentage in a range of 0.001 - 0.007% with respect to the composition; and
b) at least one organic compound having a weight percentage in a range of 0.001 - 0.007% with respect to the composition.
7. The herbal composition as claimed in claim 6, wherein the composition
comprises:
a) Zingiber officinale extract having a weight percentage in a range of 0.0025 - 0.0065% with respect to the composition; and
b) at least one organic compound having a weight percentage in the range of 0.001 - 0.005% with respect to the composition.
8. The herbal composition as claimed in any one of the claims 1-7, wherein the composition further comprises at least one additive selected from a group consisting of carriers, chelating agent, and combinations thereof.
9. A process for preparing the herbal composition as claimed in claim 1, said process comprising the steps of: a) obtaining Zingiber officinale extract; b) obtaining at least one organic compound; and c) contacting the Zingiber officinale extract with the at least one organic compound to obtain the composition.
10. The herbal composition as claimed in any one of the claims 1-8, wherein the composition provides enhanced iron uptake.
