TITLE:
A herbal nutraceutical formulation for anti-obesity and a process for
preparing the same.
FIELD OF THE INVENTION:
This invention retates to a herbal nutraceutical formulation for anti-obesity
and a process for preparing the same.
BACKGROUND OF THE INVENTION:
The term "Nutraceutical" was coined by combining the terms "Nutrition"
and "Pharmaceutical" in 1989 by Dr Stephen DeFelice, Chairman of the
Foundation for lnnovation in Medicine. "Nutraceutical" is any substance
that may be considered a food or part of a food and provides medical or
health benefits, encompassing prevention and treatment of diseases.
Such products may range from isolated nutrients, dietary supplements
and diets to genetically engineered "designer" foods, herba! products and
processed foods such as cereals, soups and beverages. "Nutraceuticals
and functional foods have received considerable interest because of their
presumed safety and potentiat nutritional and therapeutic effects". The
nutraceutical and functional food industry is in a unique position to
capitalize on consumers' interest. Be it a multinational pharmaceutical
corporation, a nutritional company, a large food multinational or a small
vitamin selling firm, all of them recognize the changing trends and are

aware of the more health-seeking consumer trend. There is, thus, a
proliferation of these value-added products aimed at not only keeping
oneself healthy but also prevention/treatment of various ailments ranging
from heart diseases to cancer. The main aim is based on the disease
preventing phytonutrients present in herbal plants.

ex@ss body fat has accumulated to the extent that it may have an adverse
effect on health, leading to reduced life expectancy and / or increased
health problems. People are considered obese when their body mass
index (BMt), a measurement obtained by dividing a person's weight in
kilograms by the square of the person's height in metres' exceeds 30
kg/m2.
OBJECTS OF THE INVENTION:
An object of this invention is to propose a herbal nutraceutical formulation
for anti-obesity.
Another object of this invention is to propose a process for preparing
herbal nutraceutical formulation.
Still another object of this invention is to propose a combination of herbal
plants with promising potential for the development of the base material to
provide essential natural nutrients.
Further object of this invention is to propose a food supplement with
specific functional attributes using medicinal plants / herbs.
BRIEF DESGRIPTION OF THE INVENTION:
According to this invention there is provided a Nutraceutical formulation
for antiobesity comprising: 2O'3Oo/o of Achyranthus aspera; 2040o/o ol
Cyperus rotundus and 40-6 Oo/o ol Amorphophallus konjac. A method for
preparing a nutraceutical formulation for antiobesity comprising: obtaining
and drying the whole plant material of Achyranthus aspera, subjecting the
dried material to the step of defattering an organic solvent, pulverizing the
defatted materiat to coarse powder, extracting the said powder using
aqueous methanotic solvent system drying the said fraction, obtaining and
washing roots and rhizomes of cyperus rotundus, drying and pulverizing

the said materials of Cyperus rotundus, subjecting the dried and
pulverized powder to the step of extraction with an organic solvent
system, drying the extract to powder state obtaining roots of
Amorphophallus konjac, subjecting the roots to the step of extraction with
water, filtering and evaporating root materials to dryness, pulverizing the
dried root material to form a dried powder mixing the powder fractions in
the following ratio:
Achyranthus asPera: 600 mg
Cyperus rotundus: 1.5-2.5 gms
Amorphophallus konjac: 1-1.5 gms
The present formulation relates to the herbal health protective and
promotive nutraceutical formutation for the obese people for their better
health.
The Herbal formulation contains extract fractions mix of following:
Achyranthus aspera, cyperus rotundus and Amorphophallus konjac
The processes described below can be scaled up to produce larger
quantities of extracts. The details provided for preparation of the following
abstracts reflect the presenily preferred method for extract preparations
and not be consider as limiting. The quantities and times described below
can be varied substantially to provide suitable extracts.

It will be appreciated by those skilled in the art that the above approximate
weight percents are dependent generally on the expected potencies of the
individual components, whereby the relative weight percents will vary
sometimes substantially from the above individual amounts. lt wil! be
within the skilled person's knowledge with this disclosure that the objects
of the present invention require the inclusion of each of the components in
retative approximate weight percents above. The whole plant material
was dried under sun, defatted using petroleum ether and was
mechanicatty reduced to moderate coarse powder and extracted using
aqueous methanolic solvent system (80:20%), at 50 C- 70C for 3040 hrs.
The extract fractions, fiuleted and concentrated to dryness using
evaporator aS contain Achrynthus aspera, contain Betaine and
Achyranthine as the principal alkaloids and also some triterpinoid
saponins. The saponins, may not be less than 0.10 - 0.2o/o, which are the
biomarker compound.
The roots and rhizomes of Cyprus rotuindus are collected, washed and
sun dried. The dry portion is pulverized into powder and extracted with
aqueous methanolic solvent system (80-20o/o) and the extraction is done
at temperature 30-60 C for 60-80 hrs. The extract is collected and dried
to powder state. The extract contains alkaloids, flavanoids, terpenes and
oil. The total extract contents of the sesquio terpenes and alkaloids
should be not less than 12-15o/o.
Amorphophallus konjac, is a dietry fibre obtained from the roots contains
glucomanann is extracted with water at temperature 40€0 C, filtered, and

evaporated to dryness at temperature 60-80 C. The dried powder contains
carbohydrates, especially glucomannan, insoluble starch, cellulose and
proteins. The total extractives are not less than 60-80 o/o ol glucomannan
content. The fibre content of the FIour is the pharmacologically active
content.
Achyranthus aspera: 20-30%, 600 mg
Cyperus rotundus: 2040o/o, 1 .5-2.5 g
Amorphophallus konjac: 40$0o/o, 1-1.5 g
To protect the formulation from any harmful growths, suitable
preservatives have to be added. These include alkyl esters of phydrorybenzoic
acid, hydanton, derivatives, propionate salts, potassium
sorbate and sodium benzoate.
The extract mix fractions for the formulation as a treatment mode for
malnutrition contains the fractions in the ratio given above.
The oral dosage from containing the mix of the above fractions of herbs
and derivatives acts as antiobesity formulation. The formulation increases
fat metabolism in liver, and lowers betalipoproteins and triglyceride levels
in the blood.

WE CLAIM:
1. A Nutraceutical formulation for antiobesity comprising:
20-30o/o of Achyranthus asPera
2040o/o of Cyperus rotundus and
40-600/o of Amorphophallus konjac
2. A method for preparing a nutraceutical formulation for antiobesity
comprising:
obtaining and drying the whole plant material of Achyranthus aspera
subjecting the dried material to the step of defattering an organic solvent
putverizing the defatted material to coarse powder
extracting the said powder using aqueous methanolic solvent system
drying the said fraction
obtaining and washing roots and rhizomes of Cyperus rotundus,
drying and putverizing the said materials of Cyperus rotundus
subjecting the dried and pulverized powder to the step of extraction with an
organic solvent sYstem,
drying the extract to powder state obtaining roots of Amorphophallus konjac
subjecting the roots to the step of extraction with water
filtering and evaporating root materials to dryness
pulverizing the dried root material to form a dried powder mixing the
powder fractions in the following ratio:
Achyranthus asPera: 600 mg
Cyperus rotundus: 1 .5-2.5 gms
Amorphophallus konjac: 1-1.5 gms

3. The method as claimed in claim 2 wherein the said step of defatting of
Achyranthine aspera is done using petroleum ether.
4. The method as claimed in claim 2 wherein the said step of extraction is
done using methanolic solvent system in a ratio of 80:20o/o at 50 - 70C for
30 - 40 hrs.
5. The method as claimed in claim 2 wherein the said pulverized powder is
extract in a organic sotvent system using aqueous methanol in a ratio of 8 -
0 - 20%at 30 - 60 c for 60 - 80 hrs.
G. The method as claimed in claim 2 wherein the said root of Amorphophallus
konjac is extracted with water at 40 - 60 C at a temperature range of 60 -
80c.
