FIELD OF THE INVENTION:
The present invention is in the field of nutritional composition, particularly enriched collagen
building composition and method thereof. In particularly, the invention provides polyphenols
enriched collagen composition extracted from Amla Fruit, Green tea powder & Green coffee
seeds.
BACKGROUND OF THE INVENTION:
The following background discussion includes information that may be useful in understanding
the present invention. It is not an admission that any of the information provided herein is prior
art or relevant to the presently claimed invention, or that any publication specifically or
implicitly referenced is prior art.
Collagen is a protein that forms many structures of the body, including tendons, bones, teeth,
and sheets that support the skin and internal organs. Collagen is composed of three strands
wrapped around a triple helix. The structure results from repetition of 3 amino acids. In the
helix, there is glycine every 3 amino acids, and most of the remaining amino acids are proline
or hydroxyproline.
Collagen has been used commercially and clinically for some time. Collagen can now be used
to replace or augment hard or soft connective tissue such as skin, tendons, cartilage, bone and
stroma. Each collagen composition has certain physical properties that can be advantageous or
disadvantageous for its use in a particular technique. Due to the various drawback associated
with the synthetic collagen, natural alternative of collagen is in great demand. Naturally derived
collagen is usually plant based and animal based.
The majority of the collagen supplements you see on the shelves are animal-based collagen
supplements. That means these supplements come from the ground up hooves, hides, bones,
and joints of animals. Most animal collagen is a by-product of factory farming, where
pesticides and herbicides like glyphosate are stored in the bones. Companies that make these
animal-based collagen supplement powders transform the collagen of animals into mainly
tasteless or flavoured powders you can mix into your coffee or put into a smoothie.
There are a couple of issues with animal-based collagen, though. The first issue is contaminants
are often found in animal-based collagen supplements. These contaminants include antibiotics,

prescription drug metabolites, steroids, insecticides, and parabens. None of these are good for
human body.
Alternate option is vegan collagen peptide builders. The vegan collagen peptide builder work
in a very different way from animal-based collagen supplements. Vegan collagen peptide
builders are not made from collagen. Like the name suggests, these plant-based collagen

as artificial endocranium, artificial ligaments, artificial tendons, surgical sutures, surgical
prostheses, surgical reinforcement, wound protecting materials, artificial skin and artificial
cornea, characterized by filling or having inside a substance having biocompatibility that can
be degraded and absorbed in the body into a matrix of a non-woven fabric-like multi-element
structure of collagen fibers having ultra-fine fibers of collagen as its basic unit.
Further, JP2021104360A discloses a composition that contains human placenta telopeptide
collagen, a method for preparing said composition, a method for using the same, and a kit
containing said composition, and said composition, kit and method are useful, for example, for
augmenting or replacing mammalian tissue.
Moving further, US7615373B2 discloses a formation and use of electroprocessed collagen,
including use as an extracellular matrix and, together with cells, its use in forming engineered
tissue. The engineered tissue can include the synthetic manufacture of specific organs or tissues
which may be implanted into a recipient. The electro processed collagen may also be combined
with other molecules in order to deliver substances to the site of application or implantation of
the electro processed collagen. The collagen or collagen/cell suspension is electrodeposited
onto a substrate to form tissues and organs.
There is a need of natural alternative to non-vegetarian sourced collagen supplement.

OBJECTIVE OF THE INVENTION:
The primary object of the present invention is to overcome the drawback associated with prior
art.
Another object of the present invention is to provide a nutritional composition, particularly an
enriched collagen building composition extracted from plant based.
Another object of the present invention is to provide an polyphenols enriched collagen
composition extracted from Amla Fruit, Green tea powder & Green coffee seeds.
Another object of the present invention is to provide a method of preparing polyphenols
enriched collagen composition extracted from Amla Fruit, Green tea powder & Green coffee
seeds.
Summary of the Invention:
The Invention provides a nutritional enriched collagen building composition comprising
polyphenols enriched collagen composition extracted from Amla Fruit, Green tea powder &
Green coffee seeds.
The nutritional composition comprises pro- collagen blend present in an amount of 2200mg
comprising the components:
i. Lemon Fruit powder present in an amount of 701 mg;
ii. Guava Fruit powder present in an amount of 499mg;
iii. Sea Buckthorn present in an amount of 200mg;
iv. Rose petal powder present in an amount of 200mg;
v. Orange juice fruit powder present in an amount of 200mg;
vi. Spinach leaf powder present in an amount of 200mg;
vii. Barley grass leaf powder present in an amount of 100mg
viii. Holy Basil leaf powder present in an amount of 100mg
The nutritional composition also comprises anti-ageing blend present in an amount of 2100mg
comprising the components:
a) Pomegranate fruit powder present in an amount of 499mg;
b) Lemon fruit powder present in an amount of 402mg;
c) Pink salt present in an amount of 399mg;
d) Strawberry fruit powder present in an amount of 302mg;

e) Acai berry fruit extract present in an amount of 200mg;
f) Ginger root powder present in an amount of 200mg;
g) Cinnamon bark extract present in an amount of 100mg.
The nutritional composition also comprises collagen- vitamin blend of 1500mg, comprising:
a) Lemon fruit powder present in an amount of 1000mg;
b) Acerola cherry fruit extract present in an amount of 200mg;
c) Sesbania leaf extract present in an amount of 100mg;
d) Bamboo shoot extract present in an amount of 100mg.
The Invention also provides a method of preparing the nutritional composition as described
above, comprising the steps of:
a) grinding the amla fruit, green tea powder & green coffee seeds coarsely;
b) carrying out extraction at 90:10 water: Ethanol in the extraction ratio of 10:1 at 50
degree celsius for 6 hrs cycle

For the purpose of promoting an understanding of the principles of the invention, reference will
now be made to the embodiment illustrated in the drawings and specific language will be used
to describe the same. It will nevertheless be understood that no limitation of the scope of the
invention is thereby intended, such alterations and further modifications in the illustrated
system, and such further applications of the principles of the invention as illustrated therein
being contemplated as would normally occur to one skilled in the art to which the invention
relates.

following detailed description are exemplary and explanatory of the invention and are not
intended to be restrictive thereof.
The Invention provides a nutritional composition. Particularly, the invention provides an
enriched collagen building composition extracted from plant based.
In an embodiment, the enriched collagen composition comprises synergistic extracts from
Amla Fruit, Green tea powder & Green coffee seeds.

In another embodiment, the invention provides a method of preparing polyphenols enriched
collagen composition extracted from Amla Fruit, Green tea powder & Green coffee seeds.
In an embodiment, the amla fruit, green tea powder & green coffee seeds are grinded coarsely
and the extraction is carried out at 90:10 water: Ethanol in the extraction ratio of 10:1 at 50
degree celsius for 6 hrs cycle. The process gives the maximum quantity of polyphenols which
majorly contribute to collagen.
In an embodiment, the invention provides green solvent extraction process which maintains
the collagen activity through enrichment of polyphenols.
In an embodiment, Collagen building blend specifically targeted with enrichment of
polyphenols which are key responsible for collagen building activity in the body.
In an embodiment, the composition of the present invention with 38% polyphenols gave the
best collagen building activity.
In an embodiment, the nutritional composition is an oral supplement.
In an embodiment, the nutritional composition is a skin care supplement.
In an embodiment, the nutritional enriched collagen building composition comprises
polyphenols enriched collagen composition extracted from Amla Fruit, Green tea powder &
Green coffee seeds.

• Samples showed an increase in collagen production for marker Human Col IV
• Collagen building blend sample showed higher production at concentration as low as
0.025 mg/ml
• The samples showed cell proliferating capacity which can promote production of
proteins such as collagen.

1. A nutritional enriched collagen building composition comprising polyphenols
enriched collagen composition extracted from Amla Fruit, Green tea powder & Green
coffee seeds.
2. The nutritional composition as claimed in claim 1, wherein the composition comprises
38% polyphenols.
3. The nutritional composition as claimed in claim 1, wherein the composition comprises
pro- collagen blend present in an amount of 2200mg comprising the components:
i. Lemon Fruit powder present in an amount of 701 mg;
ii. Guava Fruit powder present in an amount of 499mg;
