A Process Of Flocculation To Purify Crude Fermentation Broth
Abstract: The present invention relates to process of flocculation to purify recombinant proteins from a crude fermentation broth of Insulin and Insulin analogues or derivatives at a manufacturing scale in the presence of urea and Triton-X-100 at specific pH. The flocculation process is followed by at least one centrifugation and pH adjustment step for further clarification of the broth. This is further followed by filtration for final removal of any floccules present and chromatography steps to capture the pure protein. A high product recovery of more than 95% was achieved using this process.
