Abstract: The present invention relates to an anti-aging composition. More particularly, the present invention relates to an anti-aging composition which has the ability to stimulate collagen and elastin production, has brightening properties, maintains skin tone and suppleness, gives smoothness to rough skin surfaces and helps improve skin appearance and further reduces the formation of the signs of premature aging.
FIELD OF THE INVENTION
The present invention relates to an anti-aging composition. More particularly, the present invention relates to an anti-aging composition which has the ability to stimulate collagen and elastin production, has brightening properties, maintains skin tone and suppleness, gives smoothness to rough skin surfaces and helps improve skin appearance and further reduces the formation of the signs of premature aging.
BACKGROUND OF THE INVENTION
Vitamin A (Retinol) is oil soluble vitamin and it is considered essential for many metabolic processes. On skin it has anti-oxidant properties as well as anti-aging capabilities due to its ability to stimulate collagen and elastin production.
Vitamin C (Ascorbic acid) is one of the queen ingredients in anti-aging treatments due to its multifunctional properties. Vitamin C is a strong anti-oxidant and so it helps reduce the formation of the signs of premature aging caused by environmental oxidative stress factors and free-radicals. Moreover, Vitamin C is a cofactor involved in the collagen synthesis and so helps to maintain skin tone and suppleness. Moreover, it has brightening properties due to its ability to inhibit melanin production.
Ascorbic acid (Vitamin C) is unstable in water or water-based systems so in the products are contained stabilized forms of Vitamin C with the same properties.
Hyaluronic acid is a non-sulfated linear glycosaminoglycan consisting of recurrent units of D-glucuronic acid and N-acetyl-D-glucosamine (Tammi R., Agren UM., Tuhkanen AL., Tammi M. Hyaluronan metabolism in skin. Progress in Histochemistry & Cytochemistry. 29(2): 1-81, 1994).
Sodium Hyaluronate is the salt form of Hyaluronic Acid, which is a polysaccharide naturally found in connective tissue of all mammalians and has the function of maintaining the optimal water content thanks to its water binding capabilities. Applied on skin has moisturizing and film-forming properties. The hyaluronic acid film helps preserve the most important characteristics of youthful, healthy skin, such as suppleness, elasticity and tone. It is a natural and strong moisturizer with excellent water binding capabilities. Its ability to retain water gives smoothness to rough skin surfaces and helps improve skin appearance.
Vitamin B5, also known as pantothenic acid, is a water-soluble vitamin that possesses good skin permeability. Panthenol has been reported to improve hydration, reduce itching and inflammation of the skin and accelerate and improve healing of epidermal wounds and is also often used in sunburn treatment products. According to research, when Panthenol is applied topically, it penetrates into lower skin layers, is absorbed into skin cells and processed into Pantothenic Acid also known as Vitamin B5. Because it is absorbed deeply into the skin, it adds essential moisture and is also considered a potential acne treatment because of its absorption properties that can counteract bacteria and its anti-inflammatory properties.
US6379716 discloses the use of echinacea extract for reducing skin irritation and boosting anti-aging efficacy of the compositions containing hydroxy acids.
US8084062 discloses anti-aging compositions containing Hibiscus esculentus extracts and at least one selected from the group consisting of oleanolic acid, ursolic acid, glycyrhetinic acid and retinol for wrinkle improvement effect.
EP1388339 discloses an alkaline anti-wrinkling cosmetic composition comprising carnosine, derivative thereof or mixture of carnosine and derivative thereof.
Increasingly, women and men all around the world seek ways to enhance their appearance and, consequently, to fade out the signs and marks of aging that appear in the form of wrinkles and fine lines. Hence, there is an increasing demand for anti-aging products to combat the signs of skin aging.
Therefore, the present invention on the current state of the above prior arts, aims to provide a novel composition for anti-ageing and a basic skin care which contains the same directed to prevent or improve skin changes such as wrinkles, fine wrinkles, blemishes, sagging in the skin that are inducible with aging.
OBJECT OF THE INVENTION
Accordingly, the main object of the invention is to provide a synergistically effective composition for topical application comprising of vitamin A, vitamin C, panthenol and hyaluronic acid for anti-aging action, long lasting hydration and has brightening properties.
Another object of the invention is to provide a composition for topical application comprising vitamin A, vitamin C, panthenol and hyaluronic acid wherein the composition has ability to stimulate collagen and elastin production.
Yet another object of the invention is to provide a process for preparing a composition for topical application comprising vitamin A, vitamin C, panthenol and hyaluronic acid which helps to reduce the formation of the signs of premature aging caused by environmental oxidative stress factors and free-radicals.
Yet another object of the invention is to provide a composition for topical application with minimal side-effects.
Yet another object of the present invention is to provide a composition to maintain skin tone and suppleness.
Still another object of the invention is to provide a process of preparing a composition for topical application comprising vitamin A, vitamin C, panthenol and hyaluronic acid.
SUMMARY OF THE INVENTION
The present invention relates to a novel, synergistically effective composition for topical application comprising vitamin A, vitamin C, panthenol and hyaluronic acid and a process of preparing the same. More particularly, the present invention pertains to a composition for topical application comprising vitamin A, vitamin C, panthenol and hyaluronic acid wherein the composition is effective for long lasting hydration, anti-aging action and has brightening properties.
In an embodiment the present invention provides a composition comprising of vitamin A, vitamin C, panthenol and hyaluronic acid. The cumulative effect results into skin lightening, skin illumination, even-toning of all skin types, dark spot reduction, reduced tanning and enhanced moisturizing of skin.
In another embodiment the present invention provides a composition comprising of vitamin A, vitamin C, panthenol and hyaluronic acid which helps to reduce the formation of the signs of premature aging caused by environmental oxidative stress factors, free-radicals, maintain skin tone and suppleness, results in strong anti-oxidant action to fight free radicals, promotes strong collagen and elastin production thus restoring firmness and a youthful appearance, fights past damage, premature lines, repairs and restores healthy skin. The composition further provides deep and long-lasting hydration and further helps in improving skin texture, suppleness, elasticity and tone.
In yet another embodiment, the present invention provides a process of preparing an anti-aging composition comprising steps of (a) heating a predefined amount of distilled water at a predefined temperature; (b) dissolving a predefined amount of sodium hyaluronate and panthenol in heated distilled water obtained in step (a) with continuous stirring to obtain a solution; (c) cooling down the solution as obtained in step (b) to a predefined temperature with continuous stirring; (d) adding predefined amount of retinol solution to the cooled solution of step (c) and mixing well until the solution turns to hazy from transparent and further cooling down the solution to room temperature; and (e) adding predefined amount of vitamin C to the solution obtained in step (d) followed by stirring until complete dissolution to obtain an anti-aging composition; wherein, the predefined amount of sodium hyaluronate is in range of 0.01 to 0.05%; the predefined amount of panthenol is in range of 0.05 to 0.1%; the pre-defined amount of vitamin C is in range of 0.001 to 2%; the pre-defined amount of retinol solution is in range of 0.075 to 3%; the pre-defined amount of distilled water is up to 100%, and the heating in step (a) is performed at a pre-defined temperature in range of 70-80°C, preferably 75°C; and the solution in step (c) is cooled down to a pre-defined temperature in range of 25-35°C, preferably 30°C.
DETAILED DESCRIPTION OF THE INVENTION
The present invention will now be described in detail hereinafter with reference to the accompanying drawings in which a preferred embodiment of the invention is shown. This invention may, however, be embodied in many different forms and should not be construed as being limited to the embodiment set forth herein. Rather, the embodiment is provided so that this disclosure will be thorough, and will fully convey the scope of the invention to those skilled in the art.
The present invention relates to a novel, synergistically effective composition for topical application comprising vitamin A, vitamin C, panthenol and hyaluronic acid and a process of preparing the same. More particularly, the invention pertains to a composition for topical application comprising vitamin A, vitamin C and hyaluronic acid wherein the composition is effective for anti-aging action, long lasting hydration and has brightening properties. The composition preferably comprising (I) about 0.075 to 3% of vitamin A; (II) about 0.001 to 2% of vitamin C; (III) about 0.05 to 0.1% of panthenol; and (IV) about 0.01 to 0.05% of hyaluronic Acid. The ratios of the ingredients in the composition may vary depending upon type of product which includes but is not limited to serum, day crème, foam cleanser, toner, under eye serum, night crème etc.
In another preferred embodiment, said composition further comprises a cosmetically acceptable delivery system including water-based composition, oil-based composition, emulsions, suspensions, colloids, or anhydrous systems. The composition further comprises the cosmetically acceptable delivery system is selected from a toner, cleanser, eye contour, lotion, cream, shampoo, shower gel, cleanser, bath oil, salve, paste, lip balm, serum, gel, body splash, cologne, and other such as well-known topical cosmetic and pharmaceutical delivery systems.
In a preferred embodiment the present invention provides an anti-aging composition comprising of vitamin A, vitamin C, panthenol and hyaluronic acid. The cumulative effect results into anti-aging action, skin lightening, skin illumination, even-toning of all skin types, dark spot reduction, reduced tanning and enhanced moisturizing of skin. When anti-aging skin gets thinner and reduces its metabolism with aging while advancing years are a macroscopically significant factor causing cutaneous aging; influences of dryness, oxidation, and ultraviolet (UV) rays of sunlight are believed to be causes directly concerning cutaneous aging.
In another preferred embodiment the present invention provides a composition comprising of vitamin A, vitamin C, panthenol and hyaluronic acid which helps to reduce the formation of the signs of premature aging caused by environmental oxidative stress factors, free-radicals, maintain skin tone and suppleness, results in strong anti-oxidant action to fight free radicals, promotes strong collagen and elastin production thus restoring firmness and a youthful appearance, fights past damage, premature lines, repairs and restores healthy skin. The composition further provides deep and long-lasting hydration and further helps in improving skin texture, suppleness, elasticity and tone.

In yet another preferred embodiment, the present invention is to provide a synergistically effective composition for topical application comprising vitamin A, vitamin C, panthenol and hyaluronic acid wherein the composition minimises oxidative effects leading to anti-aging effects.
The skin has three layers: the epidermis, dermis, and subcutaneous tissue. With the aging process of the skin, these three components undergo degenerative changes, and changes to the dermis are the most obvious. Skin aging can be classified into two categories: Intrinsic and extrinsic. Intrinsic aging occurs with advancing age and is characterized by fine wrinkles and a thinning epidermis. In contrast, extrinsic aging is characterized by deep wrinkles, skin laxity, and hyperpigmentation, and is mainly caused by chronic sun exposure. Regardless of aging type, wrinkles and reduced elasticity are typical phenomena of skin aging and the result of progressive atrophy of the dermis. One of the main mechanisms of dermal atrophy is thought to be a reduction in the amount of extracellular matrix (ECM), particularly collagen in the dermis. In aged skin, the production of collagen decreases and its degradation increases, which leads to an overall reduction in collagen amount. Most anti-aging approaches target and aim to reverse this process. Structural changes in the collagen and other ECM proteins are also involved in dermal aging.
ROS (Reactive Oxygen Species) generated in the aging process increase MMP expression and inhibit TGF-ß signaling, which leads to collagen fragmentation and decreased collagen biosynthesis. This hinders the mechanical interaction between fibroblasts and the ECM, and consequently leads to a reduction in the size of dermal fibroblasts. The aged fibroblasts produce more ROS, which further increase the expression of MMPs and inhibit TGF-ß signaling, creating a positive feedback loop that accelerates dermal aging. Interestingly, fibroblasts in aged skin recovered their appearance and function when in contact with intact ECM implying that the ECM microenvironment is a major factor in the aging of fibroblasts. Antiaging strategies have been developed with the aim of achieving a young and healthy skin.
Oxidative stress initiated by ROS generation is an important factor modulating dermal alteration in the aging process. Topical antioxidants enhance resistance to oxidative stress and prevent dermal damage and consequently reduce the rate of skin aging. Ascorbic acid (vitamin C) has been used widely as an antiaging and hyperpigmentation topical agent for several decades. Ascorbic acid eliminates most ROS due to the oxidation of ascorbate to monodehydroascorbate, and then to dehydroascorbate, and can maintain the normal physiological state of human skin.
Regarding oral and topical photoprotection, exogenous nutrients such as a-tocopherol, oral and topical retinoids such as vitamin A and ß-carotene (a precursor of vitamin A), lycopene, and lutein all contribute to the formation of a UV-radiation barrier and some of them act as antioxidants. Indeed, it has been shown that retinoids protect skin from UV radiation because they have the ability to increase the proliferation of epidermal keratinocytes and dermal fibroblasts, and/or inhibit the expression of matrix metalloproteinases (MMPs), matrix-degrading enzymes, leading to an increase in overall protein and extracellular matrix content.
On the other hand, vitamins E and D also protect skin from ROS damage. In fact, it has been described that vitamin E inhibits MMP-1 expression, thus avoiding collagen breakdown, and acts as an anti-inflammatory agent, whereas vitamin D has been shown to stimulate innate immunity.
Vitamin A (Retinol) is an oil soluble vitamin and it is considered essential for many metabolic processes. The stabilized form of Vitamin A is a hydro-soluble solution of retinol in cyclodextrins.
Vitamin C is one of the queen ingredients in anti-aging treatments due to its multifunctional properties. Vitamin C is a strong anti-oxidant and so it helps reduce the formation of the signs of premature aging caused by environmental oxidative stress factors and free-radicals. Moreover, Vitamin C is a cofactor involved in the collagen synthesis and so helps to maintain skin tone and suppleness. Moreover, it has brightening properties thanks to its ability to inhibit melanin production. Ascorbic acid (Vitamin C) is unstable in water or water-based systems so in the products are contained stabilized forms of Vitamin C with the same properties. Pure Vitamin C (ascorbic acid) is a water-soluble ingredient but is also renowned for its poor stability in presence of water where it oxidizes rapidly. Thus, there is a need of using stabilized forms of Vitamin C. The water-based formulation which includes the serum, the toner and the cleanser and the like there need is for a water-soluble form of stabilized Vitamin C, like the Ascorbyl Glucoside. While for emulsions which includes day cream, night cream and eye contour Ascorbyl glucoside potentially be solubilized in the water phase of the emulsions but it maintains the acidic behaviour of the Ascorbic Acid which can affect badly the stability of the emulsions. Therefore, a different form of stabilized Vitamin C for the emulsion is required, in particular Ascorbyl Tetraisopalmitate, which is a liposoluble form of Vitamin C and is compatible with emulsions.

The composition may further comprise skin conditioning agents, humectants, antioxidants and UV light absorbers in varying supportive percentage ranges suitably mixed to the weight of the composition.
Human assessment trial was conducted for 4 weeks with following compositions:
Composition 1: preferably comprising (I) about 0.00375% to about 1.5% of Vitamin A, (II) about 0.0005% to about 1% of Vitamin C, (III) about 0.025% to about 0.05% of Panthenol, (IV) about 0.005% to about 0.025% of Hyaluronic Acid.
The activity of the composition was assessed on human subjects that did not provide satisfactory anti-aging effect except skin hydrating efficacy in this concentration range. The composition was further developed to obtain a result-oriented composition.
Composition 2: preferably comprising (I) about 0.05625% to about 2.25% of Vitamin A, (II) about 0.00075% to about 1.5% of Vitamin C, (III) about 0.0375% to about 0.075% of Panthenol, (IV) about 0.0075% to about 0.0375% of Hyaluronic Acid.
Upon study, it is observed that the composition provides satisfactory results when the ingredients were used in high concentration range but took longer time to show results therefore the further higher concentration was explored.
Composition 3: preferably comprising (I) about 0.075 to about 3% of Vitamin A, (II) about 0.001 to about 2% of Vitamin C, (III) about 0.05 to about 0.1% of Panthenol, (IV) about 0.01 to about 0.05% of Hyaluronic Acid.
A significant improvement is observed as it helps to reduce the formation of the signs of premature aging caused by environmental oxidative stress factors, free-radicals, maintain skin tone and suppleness. The human assessment trial was satisfactory to the extent that there was strong anti-oxidant action to fight free radicals, promotes strong collagen and elastin production thus restoring firmness and a youthful appearance, fights past damage, premature lines, and repairs and restores healthy skin. The composition also found satisfactory in synergistically improving deep and long-lasting hydration and further helps in improving skin texture, suppleness, elasticity and tone.

The combination of the four selected ingredients provides synergistic results. Vitamin A or Retinol has also a collateral activity compared with Vitamin C. These active ingredients have cumulative effect resulting into anti-aging action, skin lightening, skin illumination, even-toning of all skin types, dark spot reduction, reduced tanning and enhanced moisturizing of skin. These active ingredients helps to reduce the formation of the signs of premature aging caused by environmental oxidative stress factors, free-radicals, maintain skin tone and suppleness, results in strong anti-oxidant action to fight free radicals, promotes strong collagen and elastin production thus restoring firmness and a youthful appearance, fights past damage, premature lines, repairs and restores healthy skin.
Composition 4: preferably comprising (I) about 0.09375 to about 3.75% of Vitamin A, (II) about 0.00125 to about 2.5% of Vitamin C, (III) about 0.0625 to about 0.125% of Panthenol, (IV) about 0.0125 to about 0.0625% of Hyaluronic Acid.
The human assessment trial was satisfactory to the extent that reduce the formation of the signs of premature aging caused by environmental oxidative stress factors, free-radicals, maintain skin tone and suppleness, results in strong anti-oxidant action to fight free radicals, promotes strong collagen and elastin production thus restoring firmness and a youthful appearance, fights past damage, premature lines, repairs and restores healthy skin. However, subjects reported heaviness in texture on application and experienced discomfort during the trial period. The formulation was further adjusted with change in concentrations of skin conditioners and humectants. However, the subjects were not satisfied with the texture of the composition.
Composition 5: preferably comprising (I) about 0.1125% to about 4.5% of Vitamin A, (II) about 0.0015% to about 3% of Vitamin C, (III) about 0.075 to about 0.15% of Panthenol, (IV) about 0.015 to about 0.075% of Hyaluronic Acid
When ingredients are used in high concentration range, it is observed that the composition possesses stability and hypersensitivity issues as subjects experienced irritation and discomfort upon application. Hence, the high concentration of the ingredients was not recommended for topical formulations.
Many modifications and other embodiments of the invention set forth herein will readily occur to one skilled in the art to which the invention pertain having the benefit of the teachings presented in the foregoing descriptions and the associated drawings. Therefore, it is to be understood that the invention is not to be limited to the specific embodiments disclosed and that modifications and other embodiments are intended to be included within the scope of the appended claims. Although specific terms are employed herein, they are used in a generic and descriptive sense only and not for purposes of limitation.
We claim:
1. An anti-aging composition comprising:
vitamin A in range of 0.075 to 3%;
vitamin C in range of 0.001 to 2%;
panthenol in range of 0.05 to 0.1%; and
hyaluronic acid in range of 0.01 to 0.05%;
wherein,
the anti-aging composition has long lasting hydration, anti-aging action, brightening properties and promotes strong collagen and elastin production.
2. The composition as claimed in claim 1, wherein the composition reduces the formation of signs of premature aging caused by environmental oxidative stress factors and free-radicals.
3. The composition as claimed in claim 1, wherein the composition provides even skin tone and suppleness.
4. The composition as claimed in claim 1, wherein the composition provides skin brightening properties which includes skin illumination, even-toning of all skin types, dark spot reduction and enhanced moisturizing of skin.
5. The composition as claimed in claim 1, wherein the composition is synergistic and minimizes oxidative effects leading to anti-aging effects.
6. The composition as claimed in claim 1, wherein the composition further comprises a cosmetically acceptable delivery system including water-based composition, oil-based composition, emulsions, suspensions, colloids, or anhydrous systems.
7. The composition as claimed in claim 6, wherein the cosmetically acceptable delivery system is selected from a lotion, cream, shampoo, shower gel, cleanser, bath oil, salve, paste, lip balm, serum, gel, body splash, cologne, and other such as well-known topical cosmetic and pharmaceutical delivery systems.
8. The composition as claimed in claim 6, wherein for the emulsions stabilized form of Vitamin C is selected which includes but is not limited to Ascorbyl Tetraisopalmitate.
9. The composition as claimed in claim 6, wherein for the water-based compositions water-soluble form of stabilized Vitamin C is selected which includes but is not limited to the Ascorbyl Glucoside.
10. The composition as claimed in claim 1, wherein Vitamin A is a hydro-soluble solution of retinol in cyclodextrins.
11. A process of preparing an anti-aging composition comprising steps of:
(a) heating a predefined amount of distilled water at a predefined temperature;
(b) dissolving a predefined amount of sodium hyaluronate and panthenol in heated distilled water obtained in step (a) with continuous stirring to obtain a solution;
(c) cooling down the solution as obtained in step (b) to predefined temperature with continuous stirring;
(d) adding predefined amount of retinol solution to the cooled solution of step (c) and mixing well until the cooled solution turns to hazy from transparent and further cooling down to room temperature; and
(e) adding pre-defined amount of vitamin C to the solution obtained in step (d) followed by stirring until complete dissolution to obtain the anti-aging composition;
wherein,
the pre-defined amount of sodium hyaluronate is in range of 0.01 to 0.05%;
the pre-defined amount of panthenol is in range of 0.05 to 0.1%;
the pre-defined amount of vitamin C is in range of 0.001 to 2%;
the pre-defined amount of retinol solution is in range of 0.075 to 3%;
the pre-defined amount of distilled water is up to 100%;
the anti-aging composition has long lasting hydration, anti-aging action, brightening properties and promotes strong collagen and elastin production.
12. The process as claimed in claim 1, wherein the heating in step (a) is performed at a pre-defined temperature in range of 70-80°C.
13. The process as claimed in claim 1, wherein the solution in step (c) is cooled down to a pre-defined temperature in range of 25-35°C.
