Abstract: ABSTRACT AN ANTI-DIABETIC COMPOSITION The present invention discloses a composition comprising (3 carotene extract and bilberries fruit extract, wherein the (3 carotene extract to the bilberries fruit extract weight ratio in said composition is in the range of 1:0.25 - 1:1. Also disclosed are methods for preparing the composition and uses of the composition.

[001] The present disclosure broadly relates to an anti-diabetic composition and particularly discloses a composition comprising (3 carotene extract and bilberries fruit extract.
BACKGROUND OF INVENTION
[002] Diabetes is a chronic metabolic disease characterized by hyperglycemia majorly because of impaired insulin secretion and/or lack of insulin activity. India ranks second for people with diabetes in top ten countries (IDF diabetes Atlas 5th edition, 2012 update). Obesity is a condition in which body stores excess of fat. Predominantly, it is a lifestyle condition and is caused by sedentary habits which includes lack of exercise and less of physical activity. It is caused by a disproportion between intake and expenditure of energy (Mukherjee and Sengupta IJBT Vol. 12(1); 2013 32-39). Obesity rises the chance of several diseases including heart disease, type 2 diabetes, obstructive sleep apnea, certain types of cancer, and osteoarthritis (Haslam et al. The Lancet, 2010). India ranks third in top ten countries with highest number of obese people. (Global, regional, and national prevalence of overweight and obesity in children and adults during 1980-2013: a systematic analysis for the Global Burden of Disease Study 2013, Marie et al, 2014 The Lancet, DOI: 10.1016/S0140-6736(14)60460-8).
[003] Impaired insulin secretion by beta-cells of the pancreas, increased hepatic glucose production, and decreased peripheral glucose utilization are some of the primary reasons for the progression of diabetes. Apart from these molecular factors, there are wider forces like economics, social policies, and politics of a country or a region that contributes to the prevalence of diabetes. One of the social and economic implication is the ill-affordability of healthy food and access to the medicines in case of diseased condition. Another aspect is the lack of knowledge and awareness to check the progression of disease beyond a certain limit. In spite of investigations going on at a massive scale throughout the globe, a proper medication to check the diseases progression is still lacking. The available synthetic medications cannot be considered for a prolonged usage as they are
associated with considerable side effects. Therefore, new methods of treatment of
diabetes using novel compounds is highly warranted at this point of time.
[004] US4146613 discloses an anti-diabetic drug comprising sulfonyl urea and a
kinin in a retarded form.
[005] US20090286884 discloses compositions and processes for reducing the
level of blood glucose or ameliorating diabetic symptoms in a subject by treatment
with the composition which includes one or more safe and suitable hops acids or
hops acid derivatives.
SUMMARY OF THE INVENTION
[006] In an aspect of present disclosure, there is provided a composition comprising: (a) (3 carotene extract; and (b) bilberries fruit extract, wherein the (3 carotene extract to the bilberries fruit extract w/w ratio in said composition is in the range of 1:0.25-1:1.
[007] In an aspect of present disclosure, there is provided a method for preparing a composition comprising: (a) (3 carotene extract; and (b) bilberries fruit extract, said method comprising: (i) obtaining the (3 carotene extract; (ii) obtaining the bilberries fruit extract; and (iii) contacting the (3 carotene extract, and the bilberries fruit extract to obtain said composition, wherein the (3 carotene extract to the bilberries fruit extract w/w ratio in said composition is in the range of 1:0.25 - 1:1. [008] In an aspect of present disclosure, there is provided a method for preparing a composition comprising: (a) (3 carotene extract; (b) bilberries fruit extract; (c) at least one diluent; (d) at least one carrier; and (e) at least one excipient, said method comprising: (i) obtaining the (3 carotene extract; (ii) obtaining the bilberries fruit extract; (iii) obtaining the at least one carrier; (iv) obtaining the at least one diluent; (v) obtaining the at least one excipient; and (vi) contacting the (3 carotene extract, the bilberries fruit extract, the at least one carrier, the at least one diluent, and the at least one excipient to obtain said composition, wherein the (3 carotene extract to the bilberries fruit extract w/w ratio in said composition is in the range of 1:0.25 - 1:1 [009] These and other features, aspects, and advantages of the present subject matter will be better understood with reference to the following description and

[0023] Vaccinium myrtillus commonly known as Bilberries, is a dark blue fruit, similar in appearance to blueberries. Bilberries are rich in anthocyanosides. Bilberry extract or anthocyanosides are known to have various biological effects such as, they strengthen blood vessels, improve circulation, treats diarrhea, prevent
cell damage, can help in treating retinopathy, and may also help lower the blood glucose levels.

The inhibitors of the alpha-glucosidase may prevent the absorption of carbohydrates and may be used in the treatment of type 2 diabetes (Floris et al. Vase Health Risk Manag, 2008; 4(6): 1189-1195) and obesity (Okada et al. Hepatol Res. 2009 May; 39(5):490-500). The present document discloses herbal composition comprising (3 carotene extract and bilberries fruit extract which synergistically inhibits the activity of a-glucosidase enzyme thus, restricting the availability of glucose for absorption by the body. The present invention provides a solution for the management of diabetes using herbal extracts which are devoid of any known ill-effects.
[0027] The present invention discloses a composition comprising a combination of (3 carotene extract and bilberries fruit extract ranging from 1:0.25 to 1:1 w/w ratio ((3 carotene extract to bilberries fruit extract). The composition as disclosed in the document herein, shows a synergistic inhibition of the alpha glucosidase activity for the management of type 2 diabetes and obesity. The composition can be used in any formulation for managing diabetes. Also described is the method of preparing the composition.

[0099] The examples describe different compositions comprising different concentrations of (3 carotene extract and bilberries fruit extract. The anti-diabetic property of the compositions has been shown in terms of their ability to inhibit a glucosidase enzyme activity. The compositions which are able to exhibit
synergistic inhibition of a glucosidase enzyme activity are directly correlated with
their ability to display anti-diabetic behavior.
Material and Methods
Preparation of (3 carotene extract
[00100] Stock solution of (3 carotene extract was prepared by using commercially
available (3 carotene extract (20%) from Bio-gene extracts, Bangalore. A stock
solution of (3 carotene extract powder was prepared by dissolving 12.5 mg/ml of
solvent, preferably dimethyl sulfoxide (DMSO - 100%). A 125 - 625 ug/ mL of (3
carotene extract was prepared by dissolving the stock solution in phosphate buffer.
Preparation of bilberries fruit extract
[00101] Stock solution of Bilberries fruit extract was prepared by using
commercially available Bilberries fruit extract (25% Anthocyanidins) from Vidya
Herbs, Bangalore. A stock solution Bilberries fruit extract powder was prepared by
dissolving 12.5 mg/ml of solvent, preferably dimethyl sulfoxide (DMSO - 100%).
A 125 - 625 ug/ mL of bilberries fruit extract was prepared by dissolving the stock
solution in phosphate buffer.
a glucosidase assay
[00102] 0.1M phosphate buffer of pH7 was prepared by adding 30.75mL of 1M
K2HP04 and 19.25mL of 1M KH2P04 and diluted to 500rnL with distilled water
and stored at room temperature.
[00103] Substrate 4- nitrophenyl a-D-glucopyranoside (Sigma-N1377) lOmM
stock solution was prepared by dissolving 12.05mg of substrate in 4ml of assay
buffer and stored at -20 °C.
[00104] Alpha-glucosidase enzyme solution was prepared from intestinal acetone
powder from rat (Sigma-11630). 200 mg of intestinal acetone powder was
dissolved in 4 ml of ice cold phosphate buffer and sonicated for 15 mins at 4 °C.
After vigorous vortex, the suspension was centrifuged at 10,000g at 4 °C for 30
mins and the resulting supernatant was used for the assay.
[00105] Alpha-glucosidase assay is a colorimetric assay based on a kinetic
reaction. The principle behind the assay is that the alpha-glucosidase enzyme acts
on p-nitrophenol conjugated and a (1,4) glucosidic linkage containing substrate
(colourless) and hydrolyses it to release free p-nitrophenol (yellow colored compound). The amount of p-nitrophenol thus released, is estimated colorimetrically at 410nm.
[00106] Procedure: The assay was carried out in a final reaction volume of 200 uL in each well. A reaction mixture containing, 164 ul of phosphate buffer in the blank and 100% activity wells and 154 ul of phosphate buffer in the ingredient blank and ingredient activity wells. Then, 20 ul of substrate was added to all the wells. 10 ul of ingredients ((3 carotene extract or bilberries fruit extract or combinations thereof) were added to the ingredient blank and ingredient activity wells. Then, 16 ul of heat inactive enzyme was added to the blank and ingredient blank wells. Finally, 16 ul of active enzyme was added to 100% activity and ingredient activity wells. The reaction mixtures were mixed thoroughly and the readings were taken at 37 °C and 410 nm using kinetic loop for 30 minutes at the interval of every 1 minute.
Example 1
Effect of p carotene extract and bilberries fruit extract on a glucosidase
enzyme inhibition
[00107] The individual effects of (3 carotene extract and bilberries fruit extract was tested on the inhibition of a glucosidase enzyme inhibition by performing the a glucosidase assay as described in the Material and Methods section. Figure 1 depicts the individual effects of both the extracts on a glucosidase inhibition. Effects of (3 carotene extract - The (3 carotene extract in the varying concentrations of 125-625ug/ml was used in the assay to check the effect of a glucosidase inhibition. Figure 1 depicts the effect of different concentrations on the enzyme inhibition. Different concentrations used in the example were 125ug/ml, 250ug/ml, 312.5ug/ml, 375ug/ml, 500ug/ml, and 625ug/ml. From Figure 1, it can be appreciated that the percentage inhibition of a glucosidase enzyme varies from 10.34% for 125ug/ml (3 carotene extract to 26.70% for 625ug/ml (3 carotene extract.
Effects of bilberries fruit extract - The different concentrations used for bilberries fruit extract were 125ug/ml, 250ug/ml, 312.5ug/ml, 375ug/ml, 500ug/ml, and
625ug/ml. From Figure 1, it can be observed that the percentage inhibition of a
glucosidase enzyme varies from 5.72% (125ug/ml) to 25.53% (625ug/ml).
[00108] After studying the individual effect of (3 carotene extract and bilberries
fruit extract on inhibition of a glucosidase enzyme, effect of different compositions
comprising different concentrations of (3 carotene extract and bilberries fruit extract
were studied.
Example 2
Effect of composition comprising p carotene extract and bilberries fruit
extract on a glucosidase enzyme inhibition
[00109] The compositions comprising (3 carotene extract and bilberries fruit extract
in a w/w ratio ranging from 1:0.25 to 1:4 ((3 carotene extract: bilberries fruit
extract) were considered for checking the effect on inhibition of a glucosidase
enzyme inhibition.
[00110] Table 1 summarizes the ingredients of the compositions.
[00111] The effect of the compositions as described in Table 1 on a glucosidase inhibition is depicted in Figure 2.
[00112] The percentage of inhibition of a glucosidase enzyme by 625ug/ml of (3 carotene extract is around 20%, and the percentage of inhibition of a glucosidase enzyme by 625ug/ml of bilberries fruit extract is around 22%. The composition comprising (3 carotene extract and bilberries fruit extract in a w/w ratio of 1:0.25 (575ug/ml (3 carotene extract + 125ug/ml bilberries fruit extract) displays around 40% of a glucosidase inhibition. The percentage inhibition displayed by this composition is higher as compared to the additive effect of higher concentration (625ug/ml) of (3 carotene extract and 125ug/ml of bilberries fruit extract. Thus, the composition comprising (3 carotene extract and bilberries fruit extract in a w/w ratio of 1:0.25 displays synergistic inhibition of a glucosidase enzyme. [00113] The composition comprising (3 carotene extract and bilberries fruit extract in a w/w ratio of 1:0.66 (450ug/ml (3 carotene extract + 250ug/ml bilberries fruit extract) displays around 38% of a glucosidase inhibition. Whereas the additive percentage of inhibition due to a higher concentration (500ug/ml) of (3 carotene extract and 250ug/ml of bilberries fruit extract is only around 33%. Thus, the composition comprising (3 carotene extract and bilberries fruit extract in a w/w ratio of 1:0.66 displays a synergistic inhibition of a glucosidase enzyme inhibition. [00114] The composition comprising (3 carotene extract and bilberries fruit extract in a w/w ratio of 1:1 (312.5ug/ml (3 carotene extract + 312.5ug/ml bilberries fruit extract) displays around 40% of a glucosidase inhibition. It can be appreciated from Figure 1 that the additive inhibition effect by both the components is only around 29%. Thus, the composition comprising (3 carotene extract and bilberries fruit extract in a w/w ratio of 1:1 displays a synergistic inhibition as compared to the extracts in isolation.
[00115] The composition comprising (3 carotene extract and bilberries fruit extract in a w/w ratio of 1:1.5 (250ug/ml (3 carotene extract + 450ug/ml bilberries fruit extract) displays around 30% of a glucosidase inhibition which is lesser than the additive inhibition effect by the components in isolation. Therefore, the compositions comprising (3 carotene extract and bilberries fruit extract in a w/w ratio of 1:1.5 and 1:4 do not exhibit synergistic effect in a glucosidase inhibition. Thus, one can appreciate that considerable experimentation has been done to conclude the working weight ratio range within which the combination of (3 carotene extract and bilberries fruit extract display synergistic anti-diabetic effects.
[00116] Overall, the present disclosure describes the anti-diabetic effect of the compositions comprising (3 carotene extract and bilberries fruit extract in w/w ratio ranging from 1:0.25 to 1:1. The composition in said ratios inhibit the activity of a glucosidase enzyme. The (3 carotene extract as used in the present invention comprises 20% (3 carotene, and the bilberries fruit extract comprises 25% anthocyanidins.

[00117] The present disclosure provides compositions comprising (3 carotene extract and bilberries fruit extract. It has been assertively shown in the present disclosure that within a working range of w/w ratio ((3 carotene extract and bilberries fruit extract) 1:0.25 to 1:1 the compound exhibits anti-diabetic effects by inhibiting the activity of a glucosidase enzyme. The enzyme is mainly responsible for absorbing sugars from the gut to the blood, therefore, inhibiting its activity will assist in reducing blood sugar level which is of prime importance in diabetic patients. The compositions as disclosed, can be used in various formulations to prepare medications for chronic diabetic patients. Since the components ((3 carotene extract and bilberries fruit extract) used are of herbal origin, there should not be ill-effects from prolonged usage.
I/We Claim:
1. A composition comprising:
(a) (3 carotene extract; and
(b) bilberries fruit extract,
wherein the (3 carotene extract to the bilberries fruit extract w/w ratio in said composition is in the range of 1:0.25 - 1:1.
2. The composition as claimed in claim 1, wherein the (3 carotene extract is having a weight percentage in the range of 0.020%-0.072% with respect to the composition, and the bilberries fruit extract is having a weight percentage in the range of 0.008%-0.039% with respect to said composition.
3. The composition as claimed in claim 1, wherein the (3 carotene extract to the bilberries fruit extract w/w ratio in said composition is 1:0.25.
4. The composition as claimed in claim 1, wherein the (3 carotene extract to the bilberries fruit extract w/w ratio in said composition is 1:0.66.
5. The composition as claimed in claim 1, wherein the (3 carotene extract to the bilberries fruit extract w/w ratio in said composition is 1:1.
6. The composition as claimed in any of the claims 1-5, wherein the (3 carotene extract comprises 10%-30% of (3 carotene.
7. The composition as claimed in any of the claims 1-5, wherein the bilberries fruit extract comprises 20%-30% of anthocyanidins.
8. The composition as claimed in any of the claims 1-7, further comprising at least one carrier, at least one diluent, and at least one excipient, and combinations thereof.
9. A method for preparing a composition as claimed in claim 1, said method
comprising:
a) obtaining (3 carotene extract;
b) obtaining bilberries fruit extract; and
c) contacting the (3 carotene extract, and the bilberries fruit extract to obtain said composition.
10. A method for preparing a composition as claimed in any of the claims 1-8,
said method comprising:
a) obtaining (3 carotene extract;
b) obtaining bilberries fruit extract;
c) obtaining at least one carrier;
d) obtaining at least one diluent;
e) obtaining at least one excipient; and
f) contacting the (3 carotene extract, the bilberries fruit extract, the at least one carrier, the at least one diluent, and the at least one excipient to obtain said composition.
11. The composition as claimed in any of the claims 1-8, wherein said
composition exhibits anti-diabetic effect by inhibiting activity of a-
glucosidase enzyme.
