Abstract: The present invention relates to an extraction buffer, method and kit for isolation of RNA. The extraction buffer, method and kit of the present invention are especially useful for isolating RNA from plant specimens comprising high content of phenolics and polysaccharides. The present invention provides a cost effective and short method for high throughput RNA isolations.
CLIAMS:1.An extraction buffer for isolation of RNA, wherein the extraction buffer comprises:
a)1% to 3% cetyltrimethyl ammonium bromide;
b)0.05M to 0.2M tris HCl;
c)0.4M to 2.4M sodium chloride;
d)15mM to 25mM ethylenediaminetetraacetic acid;
e)1% to 3% polyvinylpyrrolidone;
f)200mM to 400mM sodium acetate; and
g)1% to 20% beta mercaptoethanol.
2.The extraction buffer as claimed in claim 1, wherein the extraction buffer comprises:
a)2% cetyltrimethyl ammonium bromide;
b)0.1M tris HCl;
c)1.4M sodium chloride;
d)20mM ethylenediaminetetraacetic acid;
e)2% polyvinylpyrrolidone;
f)300mM sodium acetate; and
g)10% beta mercaptoethanol.
3.The extraction buffer as claimed in claim 1 or 2, wherein the extraction buffer is free of phenol.
4.A method for isolation of RNA from a plant material, wherein the method comprises:
a)pulverizing the plant material to obtain a sample;
b)mixing the sample with an extraction buffer to obtain an extract, wherein the extraction buffer comprises:
i.2% cetyltrimethyl ammonium bromide;
ii.0.1M tris HCl;
iii.1.4M sodium chloride;
iv.20mM ethylenediaminetetraacetic acid;
v.2% polyvinylpyrrolidone;
vi.300mM sodium acetate; and
vii.10% beta mercaptoethanol;
c)adding a mixture of chloroform and isoamyl alcohol to the extract for phase separation to obtain an aqueous phase and an organic phase;
d)separating the aqueous phase from the organic phase;
e)precipitating RNA from the aqueous phase; and
f)collecting the precipitate to obtain RNA.
5.The method as claimed in claim 4, wherein the plant material is selected from the group consisting of flower, leaf, root, seed, embryo, cotyledon, xylem, and stem.
6.The method as claimed in claim 4, wherein the mixture of chloroform and isoamyl alcohol is present in the ratio of 24:1 (v/v).
7.The method as claimed in claim 4, wherein the precipitation is done by using an alcohol.
8.The method as claimed in claim 7, wherein the alcohol is isopropanol.
9.A kit for isolation of RNA, wherein the kit comprises the extraction buffer as claimed in claim 1.
10.The kit as claimed in claim 9, wherein the kit additionally comprises a mixture of chloroform and isoamyl alcohol in the ratio of 24:1 (v/v).
11.The kit as claimed in claim 9, wherein the kit additionally comprises isopropanol. ,TagSPECI:As Attached
