Abstract: The present disclosure relates to a handwash concentrate composition having broad-spectrum antimicrobial activity. Specifically, the present disclosure provides a handwash concentrate composition comprising povidone iodine; a herbal extract comprising neem extract, tulsi extract, or their combination; an anionic surfactant; a thickening agent; a solvent and a moisturizing agent. Said composition is devoid of harsh chemicals and is soft on human skin. The present disclosure further provides a process for preparing the composition.
DESC:FIELD OF THE INVENTION
[0001] The present disclosure relates to a handwash composition. Specifically, the present disclosure provides a handwash concentrate composition comprising an anti-microbial agent, a herbal extract, a moisturizing agent, an anionic surfactant, a solvent and a thickening agent. The present disclosure further provides a process for preparing the handwash concentrate composition.
BACKGROUND OF THE INVENTION

[0003] Micro-organisms can be found on all surfaces like toilets, kitchen slabs, tables, desktops, vegetables etc. Our body is constantly exposed to these micro-organisms through our hands which keep touching over a million places in a day. Because of the over-exposure of our hands to micro-organisms it is important to maintain their hygiene on a regular basis, especially for health care givers. Using plain water may not ensure complete disinfection as it does not kill the microbes instead just washes away the surface bacteria. Also, water may not be effective in washing away a number of resistant floras from the surface of our hands. Therefore, use of handwashes has become important.
[0004] Researchers overtime have come up with several solutions for handwashes. These solutions started from utilizing chlorine to now using more complex formulations using compounds like chlorhexidine, triclosan and benzalkonium chloride. These compounds are known to be harsh chemicals that may be rough on the skin. These are also dangerous for the environment such that the chemicals flow into water bodies and cause further damage to aquatic life. Other anti-microbials used for hand washing include alcohol-based handwashes. These are capable of killing the pathogenic micro-organisms however it does not have persistent activity. These chemicals are also not environmentally suitable. Further, persistent use of alcohol based washes may irritate the skin and lead to dermatitis. Another problem that these anti-bacterial handwashes face is their limited activity over the range of micro-organisms. Some of these maybe effective for gram-positive and others for gram-negative bacteria, however only a few offer broad-spectrum activity.
[0005] Therefore, there is a need in the art to look for compositions that are gentle on the skin and are broad-spectrum anti-microbials. The composition devised by the inventors of the present disclosure help overcome these drawbacks in the art.
OBJECTS OF THE INVENTION
[0006] An object of the present disclosure is to provide a handwash concentrate composition.
[0007] An object of the present disclosure is to provide a handwash composition that is gentle with cleansing and broad-spectrum anti-microbial properties.
[0008] Another object of the present disclosure is to provide a handwash concentrate composition free of harsh chemicals such as parabens, Methylisothiazolinone, Methylchloroisothiazolinone, cocamidopropyl betaine, triclosan, quaternary ammonium compounds, Benzisothiazolinone, diethanolamine, dioxane, formaldehyde, phthalates, polyethylene glycol, retinyl palmitate and retinol.
[0009] An object of the present disclosure is to provide a handwash concentrate composition that has quick action, is eco-friendly and biodegradable.

EXAMPLES
[0064] The disclosure will now be illustrated with working examples, which is intended to illustrate the working of disclosure and not intended to take restrictively to imply any limitations on the scope of the present disclosure. Unless defined otherwise, all technical and scientific terms used herein have the same meaning as commonly understood to one of ordinary skill in the art to which this disclosure belongs. Although methods and materials similar or equivalent to those described herein can be used in the practice of the disclosed methods and compositions, the exemplary methods, devices and materials are described herein. It is to be understood that this disclosure is not limited to particular methods, and experimental conditions described, as such methods and conditions may vary.
Materials: All raw materials were in house (IH) grades and commercially purchased from third party manufacturers.
Example 1
Handwash concentrate composition
[0065] The concentrate composition was prepared as per proportions provided in Table No. 1 below. For pre-mixture A, HPMC and xanthan Gum were added in a clean and oven dried beaker, to which glycerol was added and mixed with a stirring rod or in a homogenizer till uniformity was achieved. TLS was added to this beaker and mixed with stirring rod/homogenizer till uniformity was achieved. This was followed by addition of aloe vera gel with mixing. For pre-mixture B, SLS, PVP-I, EDTA, coloring agent, neem and tulsi extracts were taken in another clean and oven-dried beaker and ground by crushing till uniformity was achieved. Pre-mixtures A and B were mixed together using a stirring road or homogenizer till uniformity was achieved. Fragrance was added and mixed to give the composition (an orange viscous gel). The composition had pH in the range of 5.0-7.5 and specific density in the range 1.0-1.2. The composition was kept tightly closed (avoiding air contact) at room temperature till further use.

Anti-microbial activity
[0066] The composition of Example 1 was tested for its anti-microbial activity by a time kill assay. The assay notes the changes in the microbial population within a specified time in presence of the composition. The time kill assays were performed on the ready-to-use formulation. The ready-to-use formulations were prepared after dilution of 25ml of the concentrate composition of Example 1 in 500 mL of water. The assays were performed as per the American Standard for Testing and Material protocol ASTM-E2315-16.

[0068] TEST PROCEDURE- The working cultures of the test organisms were prepared. 10 mL of the sample was taken with respect to individual test culture.100 µL of adjusted test cultures were inoculated in individual samples prepared. A contact time of 20-60 seconds was considered. Using the pour plate technique 1 mL of the formulation was pipetted to triplicated petri dishes for each dilution prepared by serial dilution technique and then 20mL of melted SCDA was added. The petri dish was incubated at 30-35°C for 72 hours. The colonies on the petri dishes were counted with the help of colony counter after incubation and the observations were recorded in cfu (colony forming unit).

LOG REDUCTION CALCULATION
[0070] The effectiveness of the disinfection procedure against each challenge organism was calculated for each product as a log10 reduction as follows:

ACCEPTANCE CRITERIA
[0072] In case of surface challenge test positive control should yield 50 to 200% recovery of the microorganism in comparison to inoculums added. Bactericidal, fungicidal or yeasticidal activity on surfaces for general purposes is characterized by the concentration of the tested product. Agents should demonstrate a 3 log reduction of vegetative test organisms and mold spores.

[0074] Thus, the composition of the present disclosure provides quick action as the anti-microbial efficacy was about 99.99% within 20 seconds of use. Additionally, the composition shows broad spectrum activity.
EXAMPLE 3
Disk-diffusion test
[0075] The disinfectant activity of povidone-iodine was significantly increased by neem and tulsi extract in the ready-to-use formulation in the specific weight percentage ranges. To study this, a disk diffusion test was performed on E. Coli using the composition of Example 1 and some comparative compositions as shown in Table No. 3 below.

[0076] From the results, it was noted that the combination of povidone iodine, neem extract and tulsi extract mutually regulate the antimicrobial activity.

3. The composition as claimed in claim 1, wherein the herbal extract is present in a range of 0.01% w/w to 1.0%w/w of the composition.

The composition as claimed in claim 1, wherein the surfactant is selected from sodium lauryl sulfate, triethanolamine lauryl sulphate, ammonium lauryl sulfate, sodium laureth sulfate, sodium lauryl sarcosinate, sodium myreth sulfate, sodium pareth sulfate, sodium stearte, a olefin sulfonate, and ammonium laureth sulfate or combinations thereof.
5. The composition as claimed in claim 1, wherein the surfactant is present in a range of 10% w/w to 30%w/w of the composition.
6. The composition as claimed in claim 1, wherein the thickening agent is selected from hydroxypropyl methylcellulose, xanthan gum, guar gum, sodium starch glycolate, polyethylene glycol, polyvinyl pyrrolidone, methyl cellulose or combinations thereof.
7. The composition as claimed in claim 1, wherein the thickening agent is present in a range of 0.5% w/w to 15.0%w/w of the composition.
8. The composition as claimed in claim 1, wherein the moisturizing agent is selected from aloe vera gel, propylene glycol, propylene glycol, jojoba oil or combinations thereof.
9. The composition as claimed in claim 1, wherein the moisturizing agent is present in a range of 0.01% w/w to 0.1%w/w of the composition.
10. The composition as claimed in claim 1, wherein the solvent is glycerol, polyethylene glycol, propylene glycol, or combinations thereof.
11. The composition as claimed in claim 1, wherein the solvent is present in a range of 40% w/w to 55%w/w of the composition.
12. The composition as claimed in claim 1, wherein the composition further comprises one or more additive(s).
13. A handwash concentrate formulation comprising an anti-microbial agent comprising povidone iodine; a herbal extract comprising neem extract, tulsi extract, or their combination; an anionic surfactant; a thickening agent; a solvent; a moisturizing agent; and one or more additive(s).
14. A process for preparing a handwash concentrate composition, wherein the process comprises the steps of: (a) mixing liquid components of the composition in a solvent till homogenous to give a pre-mixture A; (b) grinding and mixing solid components of the composition till homogenous to give pre-mixture B; and (c) blending the pre-mixture A and pre-mixture B by stirring till uniform homogenous composition is obtained; wherein the composition comprises an anti-microbial agent comprising povidone iodine; a herbal extract comprising neem extract, tulsi extract, or their combination; an anionic surfactant; a solvent; a thickening agent and a moisturizing agent.
