DESC:TECHNICAL FIELD OF THE INVENTION
The present invention relates to novel synergistic herbal compositions for improving testosterone levels in athlete and sports personnel for increase the growth of body muscles and contribute to better activation of the nervous system, resulting in more power and strength, a better mood, enhanced libido, and many other benefits.

BACKGROUND OF THE INVENTION
Testosterone is a hormone produced primarily by Leydig cells in testes and is responsible for the development of male sexual characteristics. It is a type of androgen essential for the process of spermatogenesis. Testosterone is an anabolic hormone and regulates a number of functions in men in addition to sperm production, libido, bone mass, muscle growth, muscle strength, stamina, increased energy, mood etc. With ageing in men, the serum testosterone levels gradually decrease resulting in various physical and mental changes such as loss of libido, loss of erectile function, abdominal obesity, decreased muscle mass and strength, fatigue, decreased bone density, mood swings, decreased motivation, and decreased memory and concentration.
Sports person such as athletes and body builders are generally low on fat intake, which has a negative e?ect on anabolic hormones and can result in a reduction in serum concentrations of hormones such as testosterone, IGF-1 and insulin. Athletes and bodybuilders are more predisposed to the use of dietary supplements and hormones. Testosterone is well known to help in sexual desire function and performance, bone density, muscle mass, body composition, mood enhancement, erythropoiesis, better heart and cognition. The basic chemical composition of testosterone is steroidal, and steroids are known to have significant physiological, as well as psychological, effects in males, especially adults. Elevated testosterone levels have been demonstrated to increase the growth of body muscles and contribute to better activation of the nervous system, resulting in more power and strength, a better mood, enhanced libido, and many other benefits.
The PCT patent publication WO2011026500 discloses dietary supplements comprising extracts of Tribulus terrestris and Lepidium meyenii for the treatment of erectile dysfunction by increasing testosterone levels.
The patent publication US20140086999 discloses a biologically active dietary supplement for normalizing the androgen levels in men, improving general health and reducing obesity, said dietary supplement comprises roots, aerial portions and rhizomes of white cinquefoil, or aerial portions of white cinquefoil, a policosanol, icariin, an extract of grapes, and/or L-arginine.
There is a continuous need in the art to provide supplements comprising highly effective herbal ingredients for improving concentrations of testosterone in males in general and particularly in athletes and bodybuilders with minimal side effects thereby making the option safe for human consumption.
Focusing on the above parameters of muscle growth and strength, the inventors have come up with the present invention of herbal compositions that will increase the testosterone level, Luteinizing Hormone (LH) level, muscle strength and mass.

SUMMARY OF THE INVENTION
In one general aspect, the present invention relates to a composition comprising one or more of Withania somnifera (ashwagandha), asphaltum (shilajit), Chlorophytum borivilianum (safed musli), Mucuna Pruriens (kaunch beej), Tribulus Terresteris (gokhru), Trigonella foenum-graecum (fenugreek), Pueraria tuberosa (vidarikand), Dactylorhiza hatagirea (salam panja), Myristica fragrans (Jatiphala), Piper longum (Pippali), Yashad bhasm and Lauh bhasm.
In one embodiment of the above aspect, the composition may additionally contain Ginseng, L-Dopa, Boron, Copper, Vitamin D, Asparagus racemosus (shatawari), Serenoa repens (saw palmetto), Urtica dioica (stinging nettle) , Argyreia nervosa (Vriddhadaruka), and combinations thereof.
In one embodiment of the above aspect, ashwagandha is present in an amount of 1 to 6 grams powder or 0.01 to 1 gm extract, gokhru is present in an amount of 1 to 6 grams powder or 0.01 to 1 gm extract, kaunch beej is present in an amount of 1 to 6 grams powder or 0.01 to 1 gm extract, fenugreek is present in an amount of 1 to 5 grams powder or 0.01 to 1 gm extract, safed musli is present in an amount of 1 to 10 grams powder or 0.01 to 1 gm extract, Vidarikand is present in an amount of 1 to 5 grams powder or 0.01 to 1 gm extract, Shilajit is present in an amount of 0.1 to 1.0 grams powder or 0.01 to 1 gm extract or resin, and salam panja is present in an amount of 1 to 10 grams powder or 0.01 to 1 gm extract.
In another embodiment, the composition comprises Ashwagandha, Safed Musli, Kaunch Beej, Gokhru, Fenugreek Extract, Salam panja, Vidarikand, Jatiphala, Pippali, Yashad Bhasm and Lauh Bhasm.
In another embodiment of the above aspect, Ashwagandha is present in an amount of 100 mg to 700 mg, Shilajit is present in an amount of 100 mg to 500 mg, Safed Musli is present in an amount of 100mg to 500 mg, Kaunch Beej is present in an amount of 100 mg to 500 mg, Gokhru is present in an amount of 100 mg to 500 mg, Fenugreek Extract is present in an amount of 100 mg to 500 mg, Salam panja is present in an amount of 10 mg to 100 mg, Vidarikand is present in an amount of 10 mg to-300 mg, Jatiphala is present in an amount of 10 mg to 100 mg, Pippali is present in an amount of 1 mg to 50 mg, Yashad Bhasm is present in an amount of 1 mg to 50 mg and Lauh Bhasm is present in an amount of 1 mg to 50 mg.
In yet another embodiment of the above aspect, Ashwagandha is present in an amount of 600 mg, Shilajit is present in an amount of 450 mg, Safed Musli is present in an amount of 300 mg, Kaunch Beej is present in an amount of 450 mg, Gokhru is present in an amount of 300 mg, Fenugreek Extract is present in an amount of 300 mg, Salam panja is present in an amount of 75 mg, Vidarikand is present in an amount of 150 mg, Jatiphala is present in an amount of 50 mg, Pippali is present in an amount of 15 mg, Yashad Bhasm is present in an amount of 30 mg and Lauh Bhasm is present in an amount of 30 mg.
In an embodiment of the above aspect, the active ingredients in the form of powder or extract are blended in required quantities and formulated into tablets or capsules and sachets providing effective unit doses. The blended powder or extract may also be used as such.

DETAILED DESCRIPTION OF THE INVENTION
Testosterone is a key androgenic male hormone, and it is primarily produced by testis in Leydig cells. It is responsible for the development of male reproductive tissues and promoting secondary sexual characteristics. In adult males, testosterone plays an important role in the maintenance of muscle mass, muscle strength, energy levels and bone mass, sexual function and psychological well-being. Increased levels of testosterone enhance muscle mass and muscle strength; increase energy levels, improves libido, erectile function and improve the feelings of well-being. The testosterone levels gradually decline with ageing and the undesirable effects associated with this condition, called “andropause”, have been receiving increased attention in recent years. The undesirable effects include decreased desire for sex (libido), diminished erectile quality, particularly for night time erections, changes in mood, reduced intellectual and cognitive function, fatigue, depression, and anger, decrease in muscle mass and strength, decrease in energy levels, decreased body hair, skin alterations, erectile dysfunction, abdominal obesity, decreased memory and concentration, decreased bone mass/mineral density, increase in abdominal fat mass.
The testosterone synthesis in body can be either de novo from the cholesterol, or derived from steroids which enter through blood stream into nervous system. The latter one depends on the enzymatic ability of the neural region or cell. The complexity of testosterone mechanism of action is underlined by its metabolism and steroid nature. The synthesis of sex hormones is ultimately controlled by gonadotropin-releasing hormone (GnRH), which is produced by the hypothalamus and which stimulates the pituitary gland to release LH and FSH (Follicle-stimulating hormone). LH stimulates the testosterone release by Leydig Cells of testes.
This unique herbal composition of the present invention comprises of both adaptogens and steroidal saponins as its active ingredients.
Steroidal saponins (gitonin, protodioscin, and tribulosaponins A and B) present in herbal blend influences androgen receptors in the brain, causing an underestimation of sex hormone levels, which causes the posterior pituitary gland to secrete more LH and, therefore, increased testosterone synthesis in the testes. Additionally, protodioscin is believed to increase the conversion of testosterone to dihydrotestosterone, which promotes red blood cell production and muscle development.
Adaptogen are defined as nontoxic substances, often of plant origin, that increase the body’s ability to resist the damaging effects of stress and promote or restore normal physiological functioning. Cortisol, the main glucocorticoid form in humans, is a catabolic hormone secreted from the adrenal cortex in response to physical and psychological stress. Low serum testosterone levels in humans has also been reported during psychological and physical stress.
Ashwagandha (Withania somnifera) is rich in wide variety of chemical compounds like alkaloids, ergostane steroids, amino acids and neurotransmitters which are responsible for its medicinal properties. It is an adaptogen which exhibit neuroprotective, anti-fatigue, antidepressant, anxiolytic, nootropic, and central nervous system–stimulating activity. It can also influence the hypothalamic–pituitary–gonadal hormonal axis and increase testosterone concentrations. Ashwagandha can be used in the form of powder, or the extract standardised to containing Withanaloids in amount of 2% to 35%. In the present compositions, ashwagandha may be present in an amount of 1 to 6 grams as powder or 0.01 to 1 gm extract.
Gokhru (Tribulus Terrestris) is a small leafy plant, also known as puncture vine. Traditionally has been used for a variety of potential effects, including to enhance libido, keep the urinary tract healthy and anti-inflammatory and increase testosterone levels. It has many chemical compounds, among which the best known are steroidal glycosides (saponins) and alkaloids. The steroidal saponins, such as protodioscin and protogracillin, are thought to confer unique biological activities. Saponins are responsible for the positive physiological effects of gokhru on sexual performance and modulation of hypertension. Moreover, gokhru could reduce inflammation by attenuating muscle damage and oxidative damage. Gokhru is useful for athletes because of its potential ergogenic effects on sports performance, facilitating recovery after exercise, improvement of strength and stimulation of skeletal muscle hypertrophy associated with gokhru as a testosterone booster, and promoter of muscle anabolism. Gokhru may be used in the form of powder or extract for the same standardised to flavonoids, saponins and alkaloids. In the present compositions, gokhru may be present in an amount of 1 to 6 grams as powder or 0.01 to 1 gm as extract.
Kaunch Beej (Mucuna Pruriens) also known as 'velvet bean is considered as a natural source of L - DOPA which has been linked with reduced psychological stress. Further, Mucuna pruriens is reported to contain many bioactive constituents, including alkaloids, coumarins, flavonoids and alkylamines etc. which play an important role in increasing the antioxidant capacity. It helps in increased synthesis of sexual hormone in men. Besides psychological stress, its administration improves semen quality as it restores antioxidant levels and reduces lipid peroxide content. It is a natural growth hormone booster. Additionally, the combined effects of Chlorophytum borivilianum and Mucuna pruriens on serum growth hormone was observed where a significant increase in circulating growth hormone in exercise-trained men providing support for the use of Mucuna pruriens for increasing exercise-related muscle and strength gains. Mucuna pruriens hormonal effects potentially enhance both physiological and psychological parameters of sexual health. We can use powder or the extract for the same standardised to flavonoids, saponins and alkalioids. In the present compositions, Mucuna pruriens may be used in an amount of 1 to 6 grams in the form of powder or 0.01 to 1 gm as extract.
Fenugreek (Trigonella foenum-graecum) is one of the oldest medicinal plants, with its beneficial effects being cited in Ayurveda and traditional Chinese medicine. Fenugreek seeds include fiber, 4-hydroxyisoleucine, steroidal saponins, protodioscin, glycosides, alkaloids, polyphenols, flavonoids, antioxidants, lipids, carbohydrates, amino acids, and hydrocarbons. Extensive pre-clinical and clinical investigations have demonstrated its broad spectrum pharmacological, medicinal, and therapeutic properties including antioxidant, antidiabetic, anti-hyperlipidemic, anti-inflammatory, antifungal, antibacterial, and galactogogue. Furthermore, many studies have demonstrated its beneficial effects is sports nutrition, muscle building, and exercise. Fenugreek contains a variety of sapogenins such as diosgenin and yamogenin. In particular, diosgenin may be an important precursor in testosterone synthesis. Fenugreek can increase testosterone through an aromatase and 5a reductase inhibition. The oral administration of Fenugreek extract in clinical practice is expected to improve sports nutrition performance and increased erectile function. It is an effective ergogenic aid for rapid improvement in exercise performance capabilities and body composition above and beyond that of resistance exercise. Fenugreek may be used in the form of powder or extract for the same standardised to flavanoids, saponins and alkaloids. Fenugreek may be present in an amount of 1 to 5 grams as powder or 0.01 to 1 gm as extract.
Safed Museli (Chlorophytum borivilianum), belongs to the family Liliaceae and is used as a general health promoting substance. It is one of the main constituents of chyawanprash. Saponin glycosides which are the active principles for aphrodisiac activity are present in abundance in safed musli. Antioxidant property of the herb might be responsible for its adaptogenic activity which might correlate with high polyphenol and flavonoid contents of its root. Safed musli may be used in the form of powder or extract for the same standardized to flavonoids, saponins and alkaloids. Safed musli may be present in an amount of 1 to 10 grams as powder or 0.01 to 1 gm as extract.
Vidarikand (Pueraria tuberosa) is a plant widely used in traditional Indian medicine as tonic, aphrodisiac, antirheumatic, diuretic, and galactagogue. It possesses growth hormone inducing and nootropic properties. By induction of growth hormone vidarikand may promote weight gain and physical strength. It has anxiolytic and anti-stress properties confirming the clinical efficacy of the plant mentioned in Ayurveda. Adaptogenic activity of Vidarikand proves its utility in tolerating the stress produced as a result of physical and mental exertion. Physiological and psychological stress increases the intracellular level of ROS (Reactive oxygen species) which are short-lived, unstable, and highly reactive species containing at least one oxygen atom resulting in reduction of sex hormone levels and disrupting the hormonal balance that regulates reproductive functions. In this context Vidarikand has immunomodulatory and anti-oxidant properties which help improving the serum sex hormone levels and increases an individual’s fertility chances. Vidarikand may be used in the form of powder or extract for the same standardized to flavonoids, saponins and alkaloids. Vidarikand may be present in an amount of 1 to 5 grams as powder or 0.01 to 1 gm as extract.
Shilajit (Asphaltum) is a sticky substance found primarily in the rocks of the Himalayas. It develops over centuries from the slow decomposition of plants. It helps in testosterone synthesis and secretion by maintaining levels of two gonadotropic hormones LH and FSH as well as elevation of testosterone precursor DHEAs (dehydroepiandrosterone). Besides boosting testosterone, it helps in improving sperm parameters, increasing energy production and physical activity and providing antioxidant protection against free radicals. Shilajit may be used in the form of powder, extract or resin for the same standardized to humic and fulvic acids. Shilajit may be present in an amount of 0.1 to 1.0 grams as powder or 0.01 to 1 gm as extract.
Salam panja (Dactylorhiza hatagirea) is a herb, used in the Unani and Ayurvedic systems of medicines as a powerful stimulant and virility enhancer. It is a tonic, which boosts the libido by enhancing the testosterone levels in the blood with its strong aphrodisiac properties. It consists of a broad range of phytochemicals like indole alkaloids, phenolics (stilbene, e.g., resveratrol), and saponins along with ascorbic acid, phyllo- and naphthloquinones, glucomannan, and carotenoids which form the active constituents. It has the ability to enhance the stamina and strength in men, which has a favourable effect on their physical powers. Salam Panja promotes the anabolic effect of tissue formation resulting in increased muscle mass and strength. Salam panja may be used in the form of powder or extract for the same standardized to flavonoids, saponins and alkaloids. Salam panja may be present in an amount of 1 to 10 grams as powder or 0.01 to 1 gm as extract.
Vriddha daruka (Argyreia speciosa) is widely distributed throughout India. It has been reported to have activities as CNS depressant, nootropic, aphrodisiac, anticonvulsant, immunomodulatory, antioxidant, analgesic, antiinflammatory, hypoglycemic, hepatoprotective, antibacterial and antifungal. It is reported to contain flavonoids, sterol glycoside, flavonoids and essential oil.
Jatiphala (Myristica fragrans, Nutmeg) is a dried kernel of broadly ovoid seed and is widely used as spices and in alternative medicine as it has been reported to have aphrodisiac, memory enhancer, antidiarrheal property, anti-inflammatory and anti-cancer property.
Pippali (Piper longum) is a climber that grows wild in India, Malaysia, Nepal, Sri Lanka and Vietnam. It has been used as carminative, sedative in insomnia and epilepsy, as general tonic and haematinic, as cholagogue in obstruction of bile duct and gall bladder; as an enumenagogue and abortifacient; as anthelmintic, stomachic, aphrodisiac, alternative, laxative and anti-dysenteric and leprosy.
Yashad bhasma is a unique particulate preparation of zinc, which has been used by traditional practitioners for the treatment of various diseases such as diabetes, eye diseases, night blindness, cataract, asthma, skin disease, and acne etc. Yashad bhasma is zinc oxide prepared by classical ayurvedic method under vigil supervision of ras vaidya. It is prepared by fine processing of pure zinc metal with aloe vera juice and other herbal decoctions.
Lauha Bhasma is another herbo-metallic preparation used in ayurveda, for treating various ailments such as anemia, diarrhea, hyperlipidemia and diabetes. 100put Lauha Bhasma (LB) is a prominent ayurveda medicine and is used as an ingredient to prepare other indigenous medicines in ayurveda. Launha bhasma is iron oxide in nano-particulate form and helps it being more efficient.
The composition may additionally contain Ginseng, L-Dopa, Boron, Copper, Vitamin D, Asparagus racemosus (shatawari), Serenoa repens (saw palmetto), Urtica dioica (stinging nettle) and combinations thereof.
The active ingredients can used in crushed powdered form or the active ingredients can be subjected to extraction with chemical solvents to prepare extracts that contain the active chemical compounds responsible for biological activity. The solvents used for preparing the extracts and fractions used in the compositions can be selected from but not limited to C1-C5 alcohols, like ethanol, methanol, n-propanol, isopropyl alcohol; ketones like acetone, methylisobutyl ketone, chlorinated solvents like methylene dichloride and chloroform, water and mixtures thereof; C1-C7 hydrocarbons such as hexane; esters like ethyl acetate and the like and mixtures thereof.
The compositions of the present invention may be formulated into a dosage form selected from dry powder form, liquid form, beverage, food product, dietary supplement or any suitable form such as tablet, capsule or a sachet providing an effective dosage.
The presented examples illustrate the invention, but they should not be considered to limit the scope of the invention in any way.

The efficacy of the composition (tablets) of Example 9 was assessed on 18 male volunteers aged 25 to 35 years. The composition was dosed three times a day for minimum Sixty days. The results are presented in Table 1 below:

The results show that the composition was effective in improving the testosterone levels and decreased SHBG levels in volunteers.

A composition comprising one or more of Withania somnifera (ashwagandha), asphaltum (shilajit), Chlorophytum borivilianum (safed musli), Mucuna Pruriens (kaunch beej), Tribulus Terresteris (gokhru), Trigonella foenum-graecum (fenugreek), Pueraria tuberosa (vidarikand), Dactylorhiza hatagirea (salam panja), Myristica fragrans (Jatiphala), Piper longum (Pippali), Yashad bhasm and Lauh bhasm.

2. The composition as claimed in claim 1, wherein the composition additionally contain Ginseng, L-Dopa, Boron, Copper, Vitamin D, Asparagus racemosus (shatawari), Serenoa repens (saw palmetto), Urtica dioica (stinging nettle), Vriddhadaruka and combinations thereof.

The composition as claimed in claim 1, wherein ashwagandha is present in an amount of 1 to 6 grams powder or 0.01 to 1 gm extract, gokhru is present in an amount of 1 to 6 grams powder or 0.01 to 1 gm extract, kaunch beej is present in an amount of 1 to 6 grams powder or 0.01 to 1 gm extract, fenugreek is present in an amount of 1 to 5 grams powder or 0.01 to 1 gm extract, safed musli is present in an amount of 1 to 10 grams powder or 0.01 to 1 gm extract, Vidarikand is present in an amount of 1 to 5 grams powder or 0.01 to 1 gm extract, Shilajit is present in an amount of 0.1 to 1.0 grams powder or 0.01 to 1 gm extract or resin, and salam panja is present in an amount of 1 to 10 grams powder or 0.01 to 1 gm extract.

4. The composition as claimed in claim 1, wherein the composition comprises Ashwagandha, Safed Musli, Kaunch Beej, Gokhru, Fenugreek Extract, Salam panja, Vidarikand, Jatiphala, Pippali, Yashad Bhasm and Lauh Bhasm.

The composition as claimed in claim 1, wherein Ashwagandha is present in an amount of 100 mg to 700 mg, Shilajit is present in an amount of 100 mg to 500 mg, Safed Musli is present in an amount of 100mg to 500 mg, Kaunch Beej is present in an amount of 100 mg to 500 mg, Gokhru is present in an amount of 100 mg to 500 mg, Fenugreek Extract is present in an amount of 100 mg to 500 mg, Salam panja is present in an amount of 10 mg to 100 mg, Vidarikand is present in an amount of 10 mg to-300 mg, Jatiphala is present in an amount of 10 mg to 100 mg, Pippali is present in an amount of 1 mg to 50 mg, Yashad Bhasm is present in an amount of 1 mg to 50 mg and Lauh Bhasm is present in an amount of 1 mg to 50 mg.

6. The composition as claimed in claim 1, wherein Ashwagandha is present in an amount of 600 mg, Shilajit is present in an amount of 450 mg, Safed Musli is present in an amount of 300 mg, Kaunch Beej is present in an amount of 450 mg, Gokhru is present in an amount of 300 mg, Fenugreek Extract is present in an amount of 300 mg, Salam panja is present in an amount of 75 mg, Vidarikand is present in an amount of 150 mg, Jatiphala is present in an amount of 50 mg, Pippali is present in an amount of 15 mg, Yashad Bhasm is present in an amount of 30 mg and Lauh Bhasm is present in an amount of 30 mg.

7. The composition as claimed in claim 1, wherein the active ingredients in the form of powder or extract are blended in required quantities and used as such or formulated into tablets or capsules and sachets providing effective unit doses.
